BACKGROUND The prevalence of type 2 diabetes(T2D)has been increasing dramatically in recent decades,and 47.5%of T2D patients will die of cardiovascular disease.Thallium-201 myocardial perfusion scan(MPS)is a precise and noninvasive method to detect coronary artery disease(CAD).Most previous studies used traditional logistic regression(LGR)to evaluate the risks for abnormal CAD.Rapidly developing machine learning(Mach-L)techniques could potentially outperform LGR in capturing non-linear relationships.AIM To aims were:(1)Compare the accuracy of Mach-L methods and LGR;and(2)Found the most important factors for abnormal TMPS.METHODS 556 T2D were enrolled in the study(287 men and 269 women).Demographic and biochemistry data were used as independent variables and the sum of stressed score derived from MPS scan was the dependent variable.Subjects with a MPS score≥9 were defined as abnormal.In addition to traditional LGR,classification and regression tree(CART),random forest,Naïve Bayes,and eXtreme gradient boosting were also applied.Sensitivity,specificity,accuracy and area under the receiver operation curve were used to evaluate the respective accuracy of LGR and Mach-L methods.RESULTS Except for CART,the other Mach-L methods outperformed LGR,with gender,body mass index,age,low-density lipoprotein cholesterol,glycated hemoglobin and smoking emerging as the most important factors to predict abnormal MPS.CONCLUSION Four Mach-L methods are found to outperform LGR in predicting abnormal TMPS in Chinese T2D,with the most important risk factors being gender,body mass index,age,low-density lipoprotein cholesterol,glycated hemoglobin and smoking.

Leaf morphogenesis:The multifaceted roles of mechanics

Plants produce a rich diversity of biological forms,and the diversity of leaves is especially notable.Mechanisms of leaf morphogenesis have been studied in the past two decades,with a growing focus on the interactive roles of mechanics in recent years.Growth of plant organs involves feedback by mechanical stress:growth induces stress,and stress affects growth and morphogenesis.Although much attention has been given to potential stress-sensing mechanisms and cellular responses,the mechanical principles guiding morphogenesis have not been well understood.Here we synthesize the overarching roles of mechanics and mechanical stress in multilevel and multiple stages of leaf morphogenesis,encompassing leaf primordium initiation,phyllotaxis and venation patterning,and the establishment of complex mature leaf shapes.Moreover,the roles of mechanics at multiscale levels,from subcellular cytoskeletal molecules to single cells to tissues at the organ scale,are articulated.By highlighting the role of mechanical buckling in the formation of three-dimensional leaf shapes,this review integrates the perspectives of mechanics and biology to provide broader insights into the mechanobiology of leaf morphogenesis.

Reduced prostate branching morphogenes,s in stromal fibroblast, but not in epithelial, estrogen receptor a knockout mice

Early studies suggested that estrogen receptor alpha （ERa） is involved in estrogen-mediated imprinting effects in prostate development. We recently reported a more complete ERa knockout （KO） mouse model via mating β-actin Cre transgenic mice with floxed ERa mice. These ACTB-ERaKO male mice showed defects in prostatic branching morphogenesis, which demonstrates that ERa is necessary to maintain proliferative events in the prostate. However, within which prostate cell type ERa exerts those important functions remains to be elucidated. To address this, we have bred floxed ERa mice with either fibroblast-specific protein （FSP）-Cre or probasin-Cre transgenic mice to generate a mouse model that has deleted ERa gene in either stromal fibroblast （FSP-ERaKO） or epithelial （pes-ERaKO） prostate cells. We found that circulating testosterone and fertility were not altered in FSP.ERaKO and pes-ERaKO male mice. Prostates of FSP-ERaKO mice have less branching morphogenesis compared to that of wild-type littermates. Further analyses indicated that loss of stromal ERa leads to increased stromal apoptosis, reduced expression of insulin-likegrowth factor-1 （IGF-1） and FGFIO, and increased expression of BMP4. Collectively, we have established the first in vivo prostate stromal and epithelial selective ERaKO mouse models and the results from these mice indicated that stromal fibroblast ERa plays important roles in prostatic branching morphogenesis via a paracrine fashion. Selective deletion of the ERa gene in mouse prostate epithelial cells by probasin-Cre does not affect the regular prostate development and homeostasis.

Indocyanine green dye and its application in gastrointestinal surgery:The future is bright green

Indocyanine green(ICG)is a water-soluble fluorescent dye that is minimally toxic and widely used in gastrointestinal surgery.ICG facilitates anatomical identi-fication of structures(e.g.,ureters),assessment of lymph nodes,biliary mapping,organ perfusion and anastomosis assessment,and aids in determining the adequacy of oncological margins.In addition,ICG can be conjugated to artificially created antibodies for tumour markers,such as carcinoembryonic antigen for colorectal,breast,lung,and gastric cancer,prostate-specific antigen for prostate cancer,and cancer antigen 125 for ovarian cancer.Although ICG has shown promising results,the optimization of patient factors,dye factors,equipment,and the method of assessing fluorescence intensity could further enhance its utility.This review summarizes the clinical application of ICG in gastrointestinal surgery and discusses the emergence of novel dyes such as ZW-800 and VM678 that have demonstrated appropriate pharmacokinetic properties and improved target-to-background ratios in animal studies.With the emergence of robotic technology and the increasing reporting of ICG utility,a comprehensive review of clinical application of ICG in gastrointestinal surgery is timely and this review serves that aim.

Early Growth Response gene 1 (Egr-1) regulates HSV-1 ICP4 and ICP22 gene expression

The molecular mechanisms mediating herpes simplex virus type 1 (HSV-1) gene silencing during latent infection arenot clear. Five copies of early growth response gene 1 (Egr-1) binding elements were identified in the intron of HSV-1ICP22 (infected cell protein No. 22) gene, leading to the hypothesis that Egr-1 binds to the viral genome and regulates theviral gene expression. Transient co-transfection assays indicated that Egr-1 negatively regulated the transcription of bothfull-length and intron-removed ICP22 promoters. The same assays also revealed that Egr-1 repressed ICP4 (infected cellprotein No. 4) promoter activity in a dose-dependent manner but showed less inhibition when the intron was removed.Histone deacetylation was not involved in this regulation since histone deacetylase inhibitor trichostatin Adid not exhibitany effect on Egr-1-mediated repression. Chromatin immunoprecipitation assays showed that Egr-1 reduced the bindingof Spl to the promoters and that the co-repressor Nab2 (NGFI-A/EGR1-binding protein) was recruited to the proximityof ICP4 in the presence of Egr-1. These results suggested that the multifunctional transcription factor Egr-1 can repressHSV-1 immediate-early gene expression through the recruitment of co-repressor Nab2 and reduction of Sp1 occupancy,and thus may play a critical role in HSV-1 gene silencing during latency.

INTRODUCTION The Global Adult Tobacco Survey （GATS） is a component of Global Tobacco Surveillance System （GTSS） under auspices of the Bloomberg philanthropy and the Bill and Melinda Gates Foundation. GATS is a household survey with a standard protocol and its goals are to measure tobacco use, to assess changes due to policy and to facilitate cross country comparison. China is the largest consumer and producer of tobacco in the world. China was selected as one of 14 countries of high burden of tobacco use, large population, and mostly low income, to conduct the GATS.

The Large sky Area Multi-Object Fiber Spectroscopic telescope(LAMOST) medium-resolution spectral survey of Galactic Nebulae(MRS-N) has conducted for more than three years since 2018 September and observed more than190 thousand nebular spectra and 20 thousand stellar spectra.However,there is not yet a data processing pipeline for nebular spectra.To significantly improve the accuracy of nebulae classification and their physical parameters,we developed the MRS-N Pipeline.This article presented in detail each data processing step of the MRS-N Pipeline,such as removing cosmic rays,merging single exposure,fitting sky light emission lines,wavelength recalibration,subtracting skylight,measuring nebular parameters,creating catalogs and packing spectra.Finally,a description of the data products,including nebular spectra files and parameter catalogs,is provided.

Findings from 2010 Global Adult Tobacco Survey: Implementation of MPOWER Policy in China

Objective To assess the implementation of five key tobacco control policies in China： protection from second-hand smoke （SHS）; offering help to quit; health warnings regarding tobacco use; the enforcement of bans on tobacco advertising, promotion, and sponsorship; and increasing tobacco taxes and prices. Methods Using 2010 Global Adults Tobacco Survey in China （GATS-China）, 10 indicators are used to assess the implementation of five key tobacco control policies of MPOWER in China. Results Overall, 63.3% and 72.7% of adultsnoticed people smoking indoor workplaces and public places, respectively. Approximately 60% of smokers were not asked about their smoking habits and approximately 67% were not advised to quit on their visit to a health worker. Sixty percent of adults noticed health warning messages on cigarette packaging and in the media in the last 30 days, 63.6% stated that they would not consider quitting. Twenty percent of respondents noticed tobacco advertising, promotion, and/or sponsorship activities in the 30 days prior to the survey. Among them, 76.3% noticed the direct advertising and 50% noticed from TV programs. Although purchasing price of one pack of cigarettes ranged from 1 to 200 RMB, 50% of current smokers （about 150 million） spent 5 RMB or less on one pack of cigarette. The expenditure on 100 packets of cigarettes represents 2% of 2009 GDP per capita. Conclusion The average score for the implementation of the 5 policies of MPOWER in China is 37.3 points, indicating tobacco control policies in China is poor and there is a large gaps from the FCTC requirements.
