A Host KH RNA-Binding Protein Is a Susceptibility Factor Targeted by an RXLR Effector to Promote Late Blight Disease

Plant pathogens deliver effector proteins that alter host processes to create an environment conducive to colonization. Attention has focused on identifying the targets of effectors and how their manipulation facil- itates disease. RXLR effector Pi04089 from the potato blight pathogen Phytophthora infestans accumu- lates in the host nucleus and enhances colonization when transiently expressed in planta. Its nuclear local- ization is required for enhanced P. infestans colonization. Pi04089 interacts in yeast and in planta with a putative potato K-homology （KH） RNA-binding protein, StKRBPI. Co-localization of Pi04089 and StKRBP1, and bimolecular fluorescence complementation between them, indicate they associate at nuclear speckles. StKRBP1 protein levels increased when it was co-expressed with Pi04089. Indeed, such accumu- lation of StKRBP1 was observed also on the first day of leaf colonization by the pathogen. Remarkably, overexpression of StKRBP1 significantly enhances P. infestans infection. Mutation of the nucleotide- binding motif GxxG to GDDG in all three KH domains of StKRBP1 abolishes its interaction with Pi04089, its localization to nuclear speckles, and its increased accumulation when co-expressed with the effector. Moreover, the mutant StKRBP1 protein no longer enhances leaf colonization by P. infestans, implying that nucleotide binding is likely required for this activity. We thus argue that StKRBP1 can be regarded as a sus- ceptibility factor, as its activity is beneficial to the pathogen.

Structure and heterogeneity of habitat determine diet of predators despite prey abundance:Similar response in Long-eared,Short-eared Owls and Common Kestrels

According to one of the theses of optimal foraging theory,main prey species abundance in the hunting area is the main factor determining the diet and habitat choices of birds of *** other factors can also be *** habitat structure influences the predators’diets as *** this study we examined the influence of habitat structure on diet compositions of three species of birds:Long-eared Owl(Asio otus),Short-eared Owl(***)and Common Kestrel(Falco tinnunculus).The study was carried out from 2007 to 2019 in a 48 km2area of the Crane’s Homeland Reserve,Moscow Region,*** habitat structures of model species’hunting territories(ratio of different types of landscape elements)were classified in module"Semi-Automatic Classification Plugin"based on the QGIS.A boosted regression tree analysis identified that the share of the main prey species in the diet is primarily determined by the landscape structure of hunting *** largest share of Common Vole(Microtus arvalis)in birds’diet was determined by the shrubs area(15%of hunting area),the meadow area(75%),the habitat heterogeneity(70%)and the arable land area(5%).The same predictors determined the largest share of Root Vole(Microtus oeconomus):the shrubs area 25%,the meadow area 70%,and the arable land area 3%.The annual mean abundance of prey species did not determine their importance in the diet of birds of ***,the main prey abundance in the hunting area is not a determining factor for the formation of diet composition of birds of prey.

CRITICAL ISSUES IN THE NUMERICAL TREATMENT OF THE PARAMETER ESTIMATION PROBLEMS IN IMMUNOLOGY

A robust and reliable parameter estimation is a critical issue for modeling in immunology. We developed a computational methodology for analysis of the best-fit parameter estimates and the information-theoretic assessment of the mathematical models formulated with ODEs. The core element of the methodology is a robust evaluation of the first and second derivatives of the model solution with respect to the model parameter values. The critical issue of the reliable estimation of the derivatives was addressed in the context of inverse problems arising in mathematical immunology. To evaluate the first and second derivatives of the ODE solution with respect to parameters, we implemented the variational equations-, automatic differentiation and complex-step derivative approximation methods. A comprehensive analysis of these approaches to the derivative approximations is presented to understand their advantages and limitations.

The plastic deformation of the pipe made of Cu-based alloy hardened by incoherent nanoparticles and subjected to theuniform internal pressure was investigated. The limits of elastic and plastic resistance are determined. The insignificantexcess in the limit of the elastic resistance enables the plastic deformation in the most part of the pipe wall. The densities ofshear-forming dislocations and prismatic dislocation loops are higher in alloys strengthened with coarse particles than inalloys strengthened with fine particles. At small distances between the strengthening particles, this effect is the mostpronounced.

Testing of new ionospheric models along the meridian 110°E over the Northern Hemisphere

Despite the continuous improvement of the widely used empirical model international reference ionosphere(IRI),the recently appeared new models must be tested *** along the meridians has the advantage of dealing with the latitudinal dependent *** paper uses new models of parameters foF2(critical frequency),TEC(total electron content),andτ(equivalent slab thickness of the ionosphere),which are of great importance for evaluating the effects of space ***-Plas,NNT2F2,and NTSM models were tested using data from 6 ionosondes located along the meridian 110°E in March *** is shown that the IRI-Plas model provides the closest values to experiment with respect toτ,while the NTSM model provides a rather limited reflection of the latitude *** of foF2(NNT2F2)have shown that,the NNT2F2 model provides good conformity with experimental values in this area,but it is very dependent on the TEC processing *** latitudinal dependences of foF2 obtained with TEC and polynomial dependenceτ(Appr)showed the presence of positive deviations from medians not only during disturbances but also quiet periods,longitudinally at the meridian.

Mid-Miocene thermal impact on the lithosphere by sub-lithospheric convective mantle material:Transition from high-to moderate-Mg magmatism beneath Vitim Plateau,Siberia

High-Mg lavas are characteristic of the mid-Miocene volcanism in Inner *** the Vitim Plateau,small volume high-Mg volcanics erupted at 16-14 *** were followed with voluminous moderate-Mg lavas at 14-13 *** the former unit,we have recorded a sequence of（1） initial basaltic melts,contaminated by crustal material,（2） uncontaminated high-Mg basanites and basalts of transitional（K-Na-K） compositions,and（3） picrobasalts and basalts of K series;in the latter unit a sequence of（1） initial basalts and basaltic andesites of transitional（Na-K-Na） compositions and（2） basalts and trachybasalts of K-Na *** pressure estimation,we infer that the high-Mg melts were derived from the sublithospheric mantle as deep as 150 km,unlike the moderate-Mg melts that were produced at the shallow *** 14-13 Ma rock sequence shows that initial melts equilibrated in a garnet-free mantle source with subsequently reduced degree of melting garnet-bearing *** melting of relatively depleted lithospheric material,evidenced by mantle xenoliths,was involved in melting,*** suggest that the studied transition from high-to moderate-Mg magmatism was due to the mid-Miocene thermal impact on the lithosphere by hot sub-lithospheric mantle material from the Transbaikalian low-velocity（melting） domain that had a potential temperature as high as 1510℃.This thermal impact triggered rifting in the lithosphere of the Baikal Rift Zone.

Ectomycorrhiza of Larix sibirica Ledeb. along the gradients of main ecological factors and elevation at the Northern and Subpolar Urals

Siberian larch(Larix sibirica Ledeb.) forests cover the largest areas in the Eurasian boreal zone, but there are insufficient data on its root system including the structure and functional traits of ectomycorrhizas(EM). The aim of this research is to find out if the morphological parameters of Larix sibirica EMs responded to the changes in elevation and main ecological factors(soil humidity, soil richness, soil acidity and habitat illumination). Using light microscopy, we studied EM diameter, root diameter, mantle width, and mantle volume share, share of tannin cells layers, EM density and EM length of Larix sibirica in two main types of plant communities along the elevation gradient at the Northern and Subpolar Urals. Differences in the environment were traced using phytoindication approach and the Ellenberg ecological scales. All the studied traits depend on the elevation and studied ecological factors. The diversity of fungal mantles is low, and the proportion of unstructured and pseudoparenchymatous mantles is high in response to the deterioration of the humidity, soil nitrogen content and acidity at higher-altitude habitats. Results of EM quantitative parameters measurements confirmed this pattern. We found a decline in the EM linear dimensions accompanied by a compensatory growth of the EM density with the raised elevation and the deterioration of environmental conditions.

The effect of space weather on the ionosphere at the 110°meridian during CAWSES-Ⅱperiod

Space weather determines the state of the ionosphere,which is especially important to know during *** study this state,the period of March 7-17,2012,which was recommended by SCOSTEP for detailed studies and called CAWSES-II,was *** this period,the behavior of parameters of the solar wind(SW)and the interplanetary magnetic field(IMF)show a number of *** this paper,we study their relationship with the total electron content(TEC)and the critical frequency(foF2)of the ionosphere at the 110°meridian,near which several ionosondes are located,in the range of 20°N-62.5°*** correlation coefficients of TEC and foF2 with the SW and IMF parameters for the month and the selected period are *** ionosphere showed the considered disturbances in different ways in different latitudinal zones:negative anomalies are observed at high latitudes,and positive ones at low latitudes.A distinctive feature is the presence of signs of a super-fountain *** of TEC and foF2 from their medians can be *** IRI model reacted in the latitude range 40°N-62.5°N in the form of negative anomalies,although positive anomalies were observed in the *** lower latitudes,no reaction was *** all factors(F10.7,Dst,Kp,IMF,Np),three zones(high-latitude,midlatitude,and low-latitude)are distinguished for different *** strongest influence of disturbances on the correlation coefficients is observed in the zone of 37.5°N 62.5°N.
