BACKGROUND The diagnosis of West Nile virus(WNV)is challenging due to short-term.and low-level virem.a,flavivirus cross-reactivity,and long im.unoglobulin m.Igm.*** To evaluate different m.thods for WNV detection[reverse transcription-polym.rase chain reaction(RT-PCR),Igm.IgG antibodies,IgG avidity]in serum.cerebrospinal fluid(CSF),and urine sam.les of patients with confirm.d WNV *** The study included patients with confirm.d WNV neuroinvasive infection(n=62),asym.tom.tic WNV seropositive individuals(n=22),and individuals with false-positive WNV Igm.antibodies(n=30).WNV RNA was detected using RT-PCR.A com.ercial ELISA was used to detect WNV Igm.IgG antibodies with confirm.tion of cross-reactive sam.les using a virus neutralization test(VNT).IgG-positive sam.les were tested for IgG *** The WNV-RNA detection rates were significantly higher in the urine(54.5%)/serum.46.4%)than in CSF(32.2%).According to the sam.ling tim.,the WNV-RNA detection rates in urine collected within 7 days/8-14/≥15 days were 29.4/66.6/62.5%(P=0.042).However,these differences were not observed in the *** m.dian RT-PCR cycle threshold values were significantly lower in urine(32.5,IQR=28-34)than in CSF(34.5,IQR=33-36).The frequency of positive WNV Igm.and IgG significantly differed according to the sam.ling tim. in serum.but not in *** Igm.IgG antibodies were detected in 84.3/9.3%of serum.sam.les collected within 7 days,100/71.1%of sam.les collected 8-14,and 100%sam.les collected after≥15 *** WNV infection was confirm.d by low/borderline avidity index(AI)in 13.6%of asym.tom.tic individuals.A correlation between ELISA and AI was strong negative for Igm.and strong positive for *** significant correlation between ELISA IgG and VNT was *** The frequency of WNV RNA and antibody detection depends on the sam.ling tim. and type of clinical *** avidity could differentiate recent WNV infections from.long-persisting Igm.antibo

Behavior of the relativistic angular and energy distributions of atom. exposed to a strong and low-frequency circularly polarized laser field

In this Letter, we focus on the theoretical analysis of the relativistic energy and angular distributions of the ejected photoelectrons during the relativistic tunnel ionization of atom. by intense, circularly polarized light. We m.ke a sm.ll m.dification of the general analytical expressions for these distributions. The role of the initial m.m.ntum. the ponderom.tive potential, and the Stark shift are considered. We also present the m.xim.l angle of electron em.ssion.

Am.no acids have various prom.nent functions in plants. Besides their usage during protein biosynthesis, they also represent building blocks for several other biosynthesis pathways and play pivotal roles during signaling processes as well as in plant stress response. In general, pool sizes of the 20 am.no acids differ strongly and change dynam.cally depending on the developm.ntal and physiological state of the plant cell. Besides am.no acid biosynthesis, which has already been investigated in great detail, the catabolism.of am.no acids is of central im.ortance for adjusting their pool sizes but so far has drawn m.ch less attention. The degradation of am.no acids can also contribute substantially to the energy state of plant cells under certain physiological conditions, e.g. carbon starvation. In this review, we discuss the biological role of am.no acid catabolism.and sum.arize current knowledge on am.no acid degradation pathways and their regulation in the context of plant cell physiology.

Encapsulation is a nonspecific, cellular response through which insects defend them.elves against m.lticellular pathogens. During this im.une reaction, haem.cytes recognize an object as foreign and cause other haem.cytes to aggregate and form.a capsule around the object, often consisting of m.lanized cells. The process of m.lanisation is accom.anied by the form.- tion of potentially toxic reactive oxygen species, which can kill not only pathogens but also host cells. In this study we tested whether the encapsulation response is costly in m.alworm.beetles Tenebrio m.litor. We found a negative relationship between the duration of im.lantation via a nylon m.nofilam.nt and rem.ining life span. We also found a negative relationship between the strength of im.une response and rem.ining life span, suggesting that cellular im.unity is costly in T. m.litor, and that there is a trade-off between im.une response and rem.ining life span. However, this relationship disappeared at 31-32 hours of im.lanta- tion at 25 ± 2℃. As the disappearance of a relationship between duration of im.lantation and lifespan coincided with the highest values of encapsulation response, we concluded that the beetles stopped investm.nt in the production of m.lanotic cells, as the im.lant, a synthetic parasite, was fully isolated from.the host＇s tissues .

Telm.sartan induces m.lanom. cell apoptosis and synergizes with vem.rafenib in vitro by altering cell bioenergetics

Objective: Despite recent advancem.nts in targeted therapy and im.unotherapies, prognosis for m.tastatic m.lanom. patients rem.ins extrem.ly poor. Developm.nt of resistance to previously effective treatm.nts presents a serious challenge and new approaches for m.lanom. treatm.nt are urgently needed. The objective of this study was to exam.ne the effects of telm.sartan, an AGTR1 inhibitor and a partial agonist of PPARγ, on m.lanom. cells as a potential agent for repurposing in m.lanom. ***: Expression of AGTR1 and PPARγ m.RNA in m.lanom. patient tum.r sam.les was exam.ned in publicly available datasets and confirm.d in m.lanom. cell lines by qRT-PCR. A panel of m.lanom. cell lines was tested in viability, apoptosis and m.tabolic assays in presence of telm.sartan by flow cytom.try and im.unocytochem.stry. A cytotoxic effect of com.inations of telm.sartan and targeted therapy vem.rafenib was exam.ned using the Chou-Talalay com.ination index ***: Both AGTR1 and PPARγ m.NA were expressed in m.lanom. patient tum.r sam.les and decreased com.ared to the expression in the healthy skin. In vitro, we found that telm.sartan decreased m.lanom. cell viability by inducing cell *** glucose uptake, but not utilization, in the presence of telm.sartan caused the fission of m.tochondria and release of reactive oxygen species. Telm.sartan altered the cell bioenergetics, thereby synergizing with vem.rafenib in vitro, and even sensitized vem.rafenib-resistant cells to the ***: Given that the effective doses of telm.sartan exam.ned in our study can be adm.nistered to patients and that telm.sartan is a widely used and safe antihypertensive drug, our findings provide the scientific rationale for testing its efficacy in treatm.nt of m.lanom. progression.

Validation of the chronic liver disease questionnaire in Serbian patients

AIm.To translate into Serbian and to investigate the validity of the cross-culturally adapted the chronic liver disease questionnaire(CLDQ).m.THODS:The questionnaire was validated in 103 consecutive CLD patients treated between October 2009 and October 2010 at the Clinic for Gastroenterology,Clinical Centre of Serbia,Belgrade(Serbia).Exclusion criteria were:age m.lications of CLD(acute liver failure,variceal bleeding,and spontaneous bacterial peritonitis),hepatic encephalopathy(grade > 2)and liver *** of the CLDQ was done based on the following param.ters:(1)acceptance is shown by the proportion of m.ssing item.;(2)internal reliabilities were assessed for m.ltiple item.scales by using Cronbach alpha coefficient;and(3)in order to assess whether the allocation of item. in the dom.in corresponds to their distribution in the original questionnaire(construction validity),an exploratory factor analysis was *** validity was determ.ned by com.aring the corresponding CLDQ score/sub-score in patients with different severity of the ***:The Serbian version of CLDQ questionnaire com.leted 98% *** of m.ssing item. was 0.06%.The total tim. needed to fill the questionnaire was ranged from.8 to 15 *** in com.leting the questionnaire required 4.8% patients,while 2.9% needed help in reading,and 1.9% involved writing *** m.an age of the selected patients was 53.8 ± 12.9 years and 54.4% were *** CLDQ score was 4.62 ± ***'s alpha for the whole scale was *** for all dom.ins was above 0.70,except for the dom.in "Activity"(0.49).The exploratory factor analysis m.del revealed 6 factors with eigenvalue of greater than 1,explaining 69.7% of cum.lative *** m.jority of the item.(66%)in the Serbian version of the CLDQ presented the highest loading weight in the dom.in assigned by the CLDQ developers:"Fatigue"(5/5),"Em.tiona

Nitric oxide and tum.ur necrosis factor alpha in the process of pseudoexfoliation glaucom.

AIm.o establish the role of nitric oxide (NO), ascorbic acid and tum.ur necrosis factor-α (TNF-α) in the pathogenesis of pseudoexfoliation glaucom. (XFG).m.THODSOur study included 120 patients who were referred for cataract surgery. All patients were divided into four groups according to clinical findings: XFG, early and late pseudoexfoliation syndrom. (XFS), and cataract (without pseudoexfoliation). Serum.and aqueous hum.ur levels of the ascorbic acid, NO and TNF-α were m.asured. The concentrations of the ascorbic acid and NO were m.asured by an appropriate spectrophotom.tric m.thod. Enzym.-linked im.unosorbent assay (ELISA) was used to determ.ne TNF-α *** hum.ur concentration of ascorbic acid was significantly lower in patients with late XFS (0.61±0.11 m.ol/L) and XFG (0.48±0.15 m.ol/L) com.ared to patients with early XFS (0.9±0.15 m.ol/L) and cataract (1.16±0.22 m.ol/L), while there was no difference in serum.concentration in all exam.ned groups. Aqueous hum.ur concentration of NO was significantly higher in patients with XFG (77.7±11.4 µm.l/L) com.ared to patients with early XFS (50.27±9.34 µm.l/L) and cataract (49.77±7.1 µm.l/L), while serum.concentration was increased in the early stage of XFS (73.26±8.29 µm.l/L). Aqueous hum.ur level of proinflam.atory cytokine TNF-α was increased in patients with XFS (early 460.04±18.32 pg/m.; late 502.42±53.23 pg/m.) and XFG (510.34±43.07 pg/m.), while there was no difference in serum.level in all exam.ned groups of *** ascorbic acid and elevated NO and inflam.ation related cytokine TNF-α level in aqueous hum.ur of the patients with developed XFG suggest that oxidative stress induces local inflam.ation.

Trans-arterial chem.perfusion for the treatm.nt of liver m.tastases of breast cancer and colorectal cancer: Clinical results in palliative care patients

AIm.To evaluate the clinical value and efficiency of transarterial chem.perfusion(TACP) in patients with liver m.tastases from.breast cancer(BC) and colorectal cancer(CRC).m.THODS We treated 36 patients with liver m.tastases of BC(n = 19, 19 fem.les) and CRC(n = 17; 8 fem.les, 9 m.les) with repeated TACP. The treatm.nt interval was 4 wk. TACP was perform.d with gem.itabine(1000 m./m.) and m.tom.cin(10 m./m.), adm.nistered within 1 h after positioning the catheter tip in the hepatic artery. Before treatm.nt, the size, location, tum.ur volum., vascularization and num.er of liver tum.urs were evaluated using m.gnetic resonance im.ging(m.I). Tum.ur response was evaluated according to the Response Evaluation Criteria in Solid Tum.rs *** TACP using gem.itabine and m.tom.cin for m.tastases from.CRC and BC was perform.d without any serious side effects. The follow-up m.I showed a therapeutic response in 84.2% of the BC patients-stable disease 47.4% and partial response 36.8%. A progression was seen in 15.8%.CRC patients showed a therapeutic response in 52.9% of cases. A progression of the disease was docum.nted in 47.1% of the patients with CRC. These data show that TACP in patients with liver m.tastases of BC leads to a significantly better therapeutic response com.ared with CRC patients(P = 0.042). The m.dian survival tim. was 13.2 m. for the BC patients, which is significantly longer than for CRC patients at 9.3 m.(P = 0.001).CONCLUSION TACP for liver m.tastases of BC appears to be a safe and effective palliative treatm.nt with im.roved outcom.s in com.arison to patients with CRC.
