Melanoma is one of the most aggressive types of malignant tumors,commonly affecting young *** treatment of metastatic tumors remains obscure due to the resistance of tumor cells to drugs mediated by various *** acquisition of a resistant phenotype is associated with both genetic and epigenetic alterations in cancer ***,the current study aimed to investigate whether microRNA(miR)-204-5p could promote alterations in the cell cycle and apoptosis of dacarbazine(DTIC)-treated melanoma *** real time PCR showed that transfection of DTIC-treated SK-MEL-2 melanoma cells with miR-204-5p mimics signiﬁcantly upregulated ***,ﬂow cytometric analysis revealed that the proportion of cells in different phases of the cell cycle remained ***,the proportion of early apoptotic cells was notably enhanced following cell treatment with DTIC,accompanied by a profound increase in Ki-67 negative cells,as veriﬁed by an immunoﬂuorescence ***,miR-204-5p overexpression reduced the percentage of early apoptotic DTIC-treated melanoma *** proportion of Ki-67 negative cells was only increased by 3%.Overall,the results of the current study indicated that miR-204-5p overexpression could mostly attenuate cell apoptosis in DTIC-treated cells rather than promote their transition from the G0 phase of the cell cycle in response to chemotherapeutic agent-induced stress.

Nearly half of the global population are carriers of Helicobacter pylori(H. pylori),a Gram-negative bacterium that persists in the healthy human stomach. H. pylori can be a pathogen and causes development of peptic ulcer disease in a certain state of the macroorganism. It is well established that H. pylori infection is the main cause of chronic gastritis and peptic ulcer disease(PUD). Decontamination of the gastric mucosa with various antibiotics leads to H. pylori elimination and longer remission in this disease. However,the reasons for repeated detection of H. pylori in recurrent PUD after its successful eradication remain unclear. The reason for the redetection of H. pylori in recurrent PUD can be either reinfection or ineffective anti-Helicobacter therapy. The administration of antibacterial drugs can lead not only to the emergence of resistant strains of microorganisms,but also contribute to the conversion of H. pylori into the resting(dormant) state. The dormant forms of H. pylori have been shown to play a potential role in the development of relapses of PUD. The paper discusses morphological H. pylori forms,such as S-shaped,C-shaped,U-shaped,and coccoid ones. The authors proposes the classification of H. pylori according to its morphological forms and viability.

A host of abiotic factors modify fish social behavior. However, few studies have characterized the effects of temperature on behavior. In this study, brown trout Salmo trutta fry were reared at 5 dif- ferent temperatures （4℃, 6℃, 8℃, 10℃, and 12℃）. In order to characterize group structure, 3 be- havioral parameters were investigated： group social structure （based on inter-individual distances）, inter-individual relationships （based on physical contacts）, and individual activity. These behavioral parameters were studied at the emergence stage, which corresponds to a switch from a social gre- garious life in the gravel to a solitary one in the water column. Data analysis showed that the inter- individual distances increased with increasing temperature, particularly the nearest neighbor distance. The mean number of physical contacts between fry increased at both low and high tem- peratures. At high temperatures, most fry moved apart from each other after a physical contact. Swimming activity decreased at both the lower and upper temperatures （18% of activity at 4℃, 38% at 8℃, and 12% at 12℃）. This study showed that temperature modifies brown trout fry activity, inter-individual relationships, and social behavior, which all affect group cohesion before emer- gence and can influence their survival and dispersal.

Comparative analysis of breast and lung cancer survival rates and clinical trial enrollments among rural and urban patients in Georgia

Objectives:Rural patients have poor cancer outcomes and clinical trial(CT)enrollment compared to urban patients due to attitudinal,awareness,and healthcare access *** of cancer survival disparities and CT enrollment is important for designing interventions and innovative approaches to address the stated *** study explores the potential disparities in cancer survival rates and clinical trial enrollments in rural and urban breast and lung cancer *** hypotheses are that for both cancer types,urban cancer patients will have longer 5-year survival rates and higher enrollment rates in clinical trials than those in rural ***:We compared breast and lung cancer patients’survival rates and enrollment ratios in clinical trials between rural(RUCC 4-9)and urban counties in Georgia at a Comprehensive Cancer Center(CCC).To assess these differences,we carried out a series of independent samples t-tests and Chi-Square ***:The outcomes indicate comparable 5-year survival rates across rural and urban counties for breast and lung cancer patients,failing to substantiate our *** clinical trial enrollment rates demonstrated a significant difference between breast and lung cancer patients at CCC,no significant variation was observed based on rural or urban ***:These findings underscore the need for further research into the representation of rural patients with diverse cancer types at CCC and other cancer ***,the findings have considerable implications for the initiation of positive social change to improve CT participation and reduce cancer survival disparities.

Polydimethylsiloxane(PDMS)is a dominant material in the fabrication of microfluidic devices to generate water-in-oil droplets,particularly lipid-stabilized droplets,because of its highly hydrophobic ***,its key property of hydrophobicity has hindered its use in the microfluidic generation of oil-in-water droplets,which requires channels to have hydrophilic surface *** this article,we developed,optimized,and characterized a method to produce PDMS with a hydrophilic surface via the deposition of polyvinyl alcohol following plasma treatment and demonstrated its suitability for droplet *** proposed method is simple,quick,effective,and low cost and is versatile with respect to surfactants,with droplets being successfully generated using both anionic surfactants and more biologically relevant *** method also allows the device to be selectively patterned with both hydrophilic and hydrophobic regions,leading to the generation of double emulsions and inverted double emulsions.

The effects of enhanced UV-B radiation on the oogenesis and morpho- anatomical characteristics of the European solitary red mason bee Osmia bicornis L. （Hy- menoptera： Megachilidae） were tested under laboratory conditions. Cocooned females in the pupal stage were exposed directly to different doses （0, 9.24, 12.32, and 24.64 kJ/m2/d） of artificial UV-B. Our experiments revealed that enhanced UV-B radiation can reduce body mass and fat body content, cause deformities and increase mortality. Following UV exposure at all 3 different doses, the body mass of bees was all significantly reduced compared to the control, with the highest UV dose causing the largest reduction. Similarly, following UV-B radiation, in treated groups the fat body index decreased and the fat body index was the lowest in the group receiving the highest dose of UV radiation. Mortality and morphological deformities, between untreated and exposed females varied considerably and increased with the dose of UV-B radiation. Morphological deformities were mainly manifested in the wings and mouthparts, and occurred more frequently with an increased dose of UV. Cell death was quantified by the Terminal deoxynucleotidyl transferase dUTP nick end labeling （TUNEL） assay （DNA fragmentation） during early stages of oogenesis of O. bicornis females. The bees, after UV-B exposure exhibited more germarium ceils with fragmented DNA. The TUNEL test indicated that in germarium, low doses of UV-B poorly induced the cell death during early development. However, exposure to moderate UV-B dose increased programmed cell death. In females treated with the highest dose of UV-B the vast majority of germarium cells were TUNEL-positive.

Regional tree-ring width chronology of the Scots pine (Pinus sylvestris L.) was constructed from 8 sites in the forest-steppe belt situated in the foothills of the Selenga River basin, Russia. Moisture information contained in tree-ring width chronology was obtained through linear regression reconstruction models of annual August–July precipitation and annual water discharge of the Selenga River during the period 1767–2015. Comparison of the smoothed series allowed estimating long-term variation component of these moisture regime parameters with a high precision. At the same time, regional drought indices are less correlated with pine radial growth, because they have contribution of the other environmental variables, which are much less reflected in the tree-ring of the investigated pine forest stands. Reconstructed dynamic of the moisture regime parameters is supported by documental evident of many socially significant events in the regional history, such as crop failures caused by both droughts and floods, and catastrophic fire in the Irkutsk City in 1879. Also, dependence of the amount of precipitation in the study area on the atmospheric circulation in Central Asia is revealed to have a similar pattern with other regions, i.e., a negative relationship of precipitation with the development of large high atmospheric pressure area within its center in the Altai and Tianshan mountains.

Control of combustion area using electrical resistivity method for underground coal gasification

Underground coal gasification （UCG） is one of the clean technologies to collect heat energy and gases （hydrogen, methane, etc.） in an underground coal seam. It is necessary to further developing environ- mentally friendly UCG system construction. One of the most important UCG＇s problems is underground control of combustion area for efficient gas production, estimation of subsidence and gas leakage to the surface. For this objective, laboratory experiments were conducted according to the UCG model to iden- ti[y the process of combustion cavity development by monitoring the electrical resistivity activity on the coal samples to setup fundamental data for the technology engineering to evaluate combustion area. While burning coal specimens, that had been sampled from various coal deposits, electrical resistivity was monitored. Symmetric four electrodes system （ABMN） of direct and low-frequency current electric resistance method was used for laboratory resistivity measurement of rock samples. Made research and the results suggest that front-end of electro conductivity activity during heating and combusting of coal specimen depended on heating temperature. Combusting coal electro conductivity has compli- cated multistage type of change. Electrical resistivity method is expected to be a useful geophysical tool to for evaluation of combustion volume and its migration in the coal seam.
