ALKBH5 suppresses autophagic flux via N6-methyladenosine demethylation of ZKSCAN3 mRNA in acute pancreatitis

BACKGROUND Increasing evidence has demonstrated that N6-methyladenosine(m6A)RNA modification plays an essential role in a wide range of pathological conditions.Impaired autophagy is a critical hallmark of acute pancreatitis(AP).AIM To explore the role of the m6A modification of ZKSCAN3 in the regulation of autophagy in AP.METHODS The AP mouse cell model was established by cerulein-treated mouse pancreatic acinar cells(MPC-83),and the results were confirmed by the levels of amylase and inflammatory factors.Autophagy activity was evaluated by specific identification of the autophagy-related microstructure and the expression of autophagy-related genes.ZKSCAN3 and ALKBH5 were knocked down to study the function in AP.A m6A RNA binding protein immunoprecipitation assay was used to study how the m6A modification of ZKSCAN3 mRNA is regulated by ALKBH.RESULTS The increased expression of amylase and inflammatory factors in the supernatant and the accumulation of autophagic vacuoles verified that the AP mouse cell model was established.The downregulation of LAMP2 and upregulation of LC3-II/I and SQSTM1 demonstrated that autophagy was impaired in AP.The expression of ZKSCAN3 was upregulated in AP.Inhibition of ZKSCAN3 increased the expression of LAMP2 and decreased the expression of the inflammatory factors,LC3-II/I and SQSTM1.Furthermore,ALKBH5 was upregulated in AP.Knockdown of ALKBH5 downregulated ZKSCAN3 expression and restored decreased autophagic flux in AP.Notably,the bioinformatic analysis revealed 23 potential m6A modification sites on ZKSCAN3 mRNA.The m6A modification of ZKSCAN3 mRNA was significantly decreased in AP.Knockdown of ALKBH5 increased the modification of ZKSCAN3 mRNA,which confirmed that ALKBH5 upregulated ZKSCAN3 expression in a m6A-dependent manner.CONCLUSION ALKBH5 inhibits autophagic flux through m6A demethylation of ZKSCAN3 mRNA in AP,thereby aggravating the severity of the disease.

Identifying drivers of urban landuse changes in the Wenchuan earthquake- affected area by using night-time light data

To learn the process of urban land evolution before and after an earthquake is vital to formulate the urban reconstruction control policies and recovery measures in the earthquake-stricken areas.However,spatiotemporal evolution and its driving factors of urban land in earthquake-prone areas remains limited due to the scarcity of ground observation data.This research,leveraging night-time light remote sensing imagery and land cover data,conducted a comprehensive analysis of the long-term evolution characteristics of urban land in earthquake-prone areas.It introduced methodologies for assessing the socio-economic impact and the primary natural environmental factors driving urban land evolution in these regions.To validate the proposed methods,the 2008 Wenchuan earthquake-affected area in China was selected as a representative study area.The results indicated that the average Digital Number(DN)values in socio-economically impacted areas showed a trend of rising,falling,and then rising again after the earthquake.DN values in three types of damaged areas including Type Ⅱ,Type Ⅲ,and Type Ⅳ exceeded pre-earthquake levels.The analysis of determinative factors influencing urban land evolution revealed that slope and elevation were key elements in controlling urban land expansion before the earthquake,whereas factors such as slope,elevation,lithology,and faults had a stronger influence on urban land expansion after the earthquake.It can be seen that,in view of the differences in the natural conditions of regions for post-disaster reconstruction,the local government need to actively adjust and adapt to urban spatial planning,so as to leverage the scale effect of large-scale inputs of funds,facilities,human resources and other factors after the disaster,thus enhancing resilience and recovery efficiency in response to disaster impacts.

Rare esophageal carcinoma-primary adenoid cystic carcinoma of the esophagus: A case report

BACKGROUND Esophageal adenoid cystic carcinoma(EACC)is an exceedingly rare malignant tumor of the esophagus,posing significant challenges in the clinic.CASE SUMMARY This report detailed the case of a 72-year-old male whose diagnosis of EACC was confirmed through postoperative histopathological examination.The patient underwent thoracoscopy-assisted radical resection of the esophageal tumor,coupled with lymph node dissection.Pathological findings revealed an adenoid cystic carcinoma infiltrating the entire layer of the muscularis propria,locally extending into the outer membrane of the esophageal fiber,involving the cardia and exhibiting no lymph node metastasis.The patient’s condition was classified as primary EACC,T3N0M0,per the American Joint Committee on Cancer(2017;8th edition).One month after surgery,the patient received postoperative adjuvant radiation therapy.CONCLUSION In addressing the rarity and high potential for biopsy misdiagnosis of EACC,this study delved into its diagnostic methods and treatment.

Enhanced electromagnetic wave absorption properties of ZIF-67 modified polymer-derived SiCN ceramics by in situ construction of multiple heterointerfaces

Multiphase polymer-derived ceramics have the advantages of thermal stability and adjustable dielectric properties,which exhibit significant potential for use in high-temperature microwave absorbing materials.Herein,Co-containing polymer-derived SiCN(Co-SiCN)ceramics were successfully synthesized by the physical mixing of zeolitic imidazolate framework(ZIF)-67 and polysilazane precursors and subsequent pyrolysis.The phase and chemical compositions,microstructures,dielectric properties,electromagnetic wave absorption(EWA)performance,and mechanism of the ceramics were investigated.The results showed that the introduction of ZIF-67 promoted the in situ formation of dielectric loss phases,including SiC nanocrystals,CoSi nanocrystals,and free carbon.The phase composition can be regulated by controlling the pyrolysis temperature to achieve ideal EWA properties.The Co-SiCN ceramic pyrolyzed at 1500℃demonstrated excellent EWA performance,with a maximum effective absorption band(EAB_(max))of 3.0 GHz at an ultralow thickness of 1.05 mm and minimum reflection loss(RL_(min))of-46.4 dB at a low frequency of 6 GHz.Compared with other reported SiCN-based ceramics containing magnetic metals,the ceramics prepared in this study stand out because of their low RL and high EAB at low thicknesses.The superior microwave absorption performance of the Co-SiCN ceramics is attributed to the heterointerface polarization,and impedance matching induced by the synergistic effects of their co-existing electromagnetic transparent/absorption phases.This study provides new insights into the development of high-performance SiCNbased microwave absorbers.

Corrosion evaluation and resistance study of alloys in chloride salts for concentrating solar power plants

Thermal energy storage(TES)systems based on molten salt are widely used in concentrating solar power(CSP)plants.The investigation of the corrosion behavior of alloy materials in molten salt is crucial for the correct selection of alloy materials and the design of TES systems.In this study,the corrosion behavior of 304,310S,316,and In625 alloys in molten chloride salts(27 mol%NaCl-22 mol%KCl-51 mol%MgCl,)was investigated.The evolution of mass loss of the alloy samples with corrosion time and temperature and the analysis of the experimental results by scanning electron microscopy(SEM),energy dispersive spectrometer(EDS),and X-ray diffraction(XRD)revealed the corrosion mechanism of the alloy samples in molten chloride salts.The main factors affecting the corrosion of the alloy samples were further analyzed.It was found that the loose multi-layer corrosion was formed on the surface of the corroded alloy samples with the increase in corrosion degree.Moreover,the experimental results.showed that Mo played a positive role in improving the corrosion resistance of the alloy samples because the presence of Mo could inhibit the outward diffusion of alloying element Cr.This work enriches the molten salt corrosion database and provides a reference for the selection of alloy materials for TES systems with potential application in CSP plants.

The joint knowledge reasoning model based on knowledge representation learning for aviation assembly domain

Knowledge graph technology is widely applied in the domain of general knowledge reasoning with an excellent performance.For fine-grained professional fields,professional knowledge graphs can provide more accurate information in practical industrial scenarios.Based on an aviation assembly domain-specific knowledge graph,the article constructs a joint knowledge reasoning model,which combines a named entity recognition model and a subgraph embedding learning model.When performing knowledge reasoning tasks,the two models vectorize entities,relationships and entity attributes in the same space,so as to share parameters and optimize learning efficiency.The knowledge reasoning model,which provides intelligent question answering services,is able to reduce the assembly error rate and improve the assembly efficiency.The system can accurately solve general knowledge reasoning problems in the assembly process in actual industrial scenarios of general assembly and component assembly under interference-free conditions.Finally,this paper compares the proposed knowledge reasoning model based on knowledge representation learning and the question-answering system based on large-scale pre-trained models.In the application scenario of system functional testing in general assembly,the joint model attains an accuracy rate of 95%,outperforming GPT with 78%accuracy and enhanced representation through knowledge integration with 71%accuracy.

Expediting redox kinetics of sulfur species on conductive scaffolds with limited charge accessible surface is considered as an imperative approach to realize energy-dense and power-intensive lithium-sulfur(Li-S)batteries.In this work,the concept of concurrent hetero-/homo-geneous electrocatalysts is proposed to simultaneously mediate liquid-solid conversion of lithium polysulfides(LiPSs)and solid lithium disulfide/sulfide(Li_(2)S_(2)/Li_(2)S)propagation,the latter of which suffers from sluggish reduction kinetics due to buried conductive scaffold surface by extensive deposition of Li_(2)S_(2)/Li_(2)S.The selected model material to verify this concept is a two-in-one catalyst:carbon nanotube(CNT)scaffold supported iron-cobalt(Fe-Co)alloy nanoparticles and partially carbonized selenium(C-Se)component.The Fe-Co alloy serves as a heterogeneous electrocatalyst to seed Li_(2)S_(2)/Li_(2)S through sulphifilic active sites,while the C-Se sustainably releases soluble lithium polyselenides and functions as a homogeneous electrocatalyst to propagate Li_(2)S_(2)/Li_(2)S via solution pathways.Such bi-phasic mediation of the sulfur species benefits reduction kinetics of LiPS conversion,especially for the massive Li_(2)S_(2)/Li_(2)S growth scenario by affording an additional solution directed route in case of conductive surface being largely buried.This strategy endows the Li-S batteries with improved cycling stability(836 mA h g^(-1)after 180 cycles),rate capability(547 mA h g^(-1)at 4 C)and high sulfur loading superiority(2.96 mA h cm^(-2)at 2.4 mg cm^(-2)).This work hopes to enlighten the employment of bi-phasic electrocatalysts to dictate liquid-solid transformation of intermediates for conversion chemistry batteries.

On Robust Cross-view Consistency in Self-supervised Monocular Depth Estimation

Remarkable progress has been made in self-supervised monocular depth estimation (SS-MDE) by exploring cross-view consistency, e.g., photometric consistency and 3D point cloud consistency. However, they are very vulnerable to illumination variance, occlusions, texture-less regions, as well as moving objects, making them not robust enough to deal with various scenes. To address this challenge, we study two kinds of robust cross-view consistency in this paper. Firstly, the spatial offset field between adjacent frames is obtained by reconstructing the reference frame from its neighbors via deformable alignment, which is used to align the temporal depth features via a depth feature alignment (DFA) loss. Secondly, the 3D point clouds of each reference frame and its nearby frames are calculated and transformed into voxel space, where the point density in each voxel is calculated and aligned via a voxel density alignment (VDA) loss. In this way, we exploit the temporal coherence in both depth feature space and 3D voxel space for SS-MDE, shifting the “point-to-point” alignment paradigm to the “region-to-region” one. Compared with the photometric consistency loss as well as the rigid point cloud alignment loss, the proposed DFA and VDA losses are more robust owing to the strong representation power of deep features as well as the high tolerance of voxel density to the aforementioned challenges. Experimental results on several outdoor benchmarks show that our method outperforms current state-of-the-art techniques. Extensive ablation study and analysis validate the effectiveness of the proposed losses, especially in challenging scenes. The code and models are available at https://***/sunnyHelen/RCVC-depth.

Histone modification and nucleosome assembly play important roles in chromatin-related processes.Histone chaperones form different complexes and coordinate histone transportation and assembly.Various histone chaperone complexes have been identified in different organisms.The ciliate protozoa(ciliates)have various chromatin structures and different nuclear morphology.However,histone chaperone components and functions of different subunits remain unclear in ciliates.Tet-rahymema thermophila contains a transcriptionally active macronucleus(MAC)and a transcriptionally inactive micronu-cleus(MIC)which exhibit multiple replication and various chromatin remodeling progresses during vegetative growth and sexual developmental stages.Here,we found histone chaperone RebL1 not only localized evenly in the transcriptionally active MAC but also dynamically changed in the MIC during vegetative growth and sexual developmental stages.REBL1 knockdown inhibited cellular proliferation.The macronuclear morphology became bigger in growing mutants.The abnormal macronuclear structure also occurred in the starvation stage.Furthermore,micronuclear meiosis was disturbed during sexual development,leading to a failure to generate new gametic nuclei.RebL1 potentially interacted with various factors involved in histone-modifying complexes and chromatin remodeling complexes in different developmental stages.REBL1 knockdown affected expression levels of the genes involved in chromatin organization and transcription.Taken together,RebL1 plays a vital role in maintaining macronuclear structure stability and gametogenesis in T.thermophila.
