Fixed-point fast sweeping WENO methods are a class of efficient high-order numerical methods to solve steady-state solutions of hyperbolic partial differential equations(PDEs).The Gauss-Seidel iterations and alternating sweeping strategy are used to cover characteristics of hyperbolic PDEs in each sweeping order to achieve fast convergence rate to steady-state solutions.A nice property of fixed-point fast sweeping WENO methods which distinguishes them from other fast sweeping methods is that they are explicit and do not require inverse operation of nonlinear local systems.Hence,they are easy to be applied to a general hyperbolic system.To deal with the difficulties associated with numerical boundary treatment when high-order finite difference methods on a Cartesian mesh are used to solve hyperbolic PDEs on complex domains,inverse Lax-Wendroff(ILW)procedures were developed as a very effective approach in the literature.In this paper,we combine a fifthorder fixed-point fast sweeping WENO method with an ILW procedure to solve steadystate solution of hyperbolic conservation laws on complex computing regions.Numerical experiments are performed to test the method in solving various problems including the cases with the physical boundary not aligned with the grids.Numerical results show highorder accuracy and good performance of the method.Furthermore,the method is compared with the popular third-order total variation diminishing Runge-Kutta(TVD-RK3)time-marching method for steady-state computations.Numerical examples show that for most of examples,the fixed-point fast sweeping method saves more than half CPU time costs than TVD-RK3 to converge to steady-state solutions.

Chemical vapor deposition on metal substrates using gaseous hydrocarbon as carbon feedstock has proven to be a feasible way for synthesis of large-area and uniform graphene films. Meanwhile, rearrangement of amorphous carbon species extracted from solid carbon sources into crystalline graphene not only offers an alternative route for graphene growth but also provides a powerful tool to excavate the underlying mechanisms of graphene formation. In this review, we discuss the processes of extracting carbon species from various solid carbon sources to form gra- phene. We summarize the typical steps of solid carbon sources to graphene transition and evaluate the resultant graphene film quality with regard to electrical performance. Unveiling the detailed solid carbon to graphene transition process paves the way for a reproducible production of graphene with controlled layer number, defect type and density, toward the final end of graphene＇s commercial utilizations with low cost.

BACKGROUND Gastric cancer(GC)is the fifth most common type of cancer and has the fourth highest death rate among all cancers.There is a lack of studies examining the impact of liver metastases on the effectiveness of immunotherapy in individuals diagnosed with GC.AIM To investigate the influence of liver metastases on the effectiveness and safety of immunotherapy in patients with advanced GC.METHODS This retrospective investigation collected clinical data of patients with advanced stomach cancer who had immunotherapy at our hospital from February 2021 to January 2023.The baseline attributes were compared using either the Chi-square test or the Fisher exact probability method.The chi-square test and Kaplan-Meier survival analysis were employed to assess the therapeutic efficacy and survival duration in GC patients with and without liver metastases.RESULTS The analysis comprised 48 patients diagnosed with advanced GC,who were categorized into two groups:A liver metastasis cohort(n=20)and a non-liver metastatic cohort(n=28).Patients with liver metastasis exhibited a more deteriorated physical condition compared to those without liver metastasis.The objective response rates in the cohort with metastasis and the cohort without metastasis were 15.0%and 35.7%(P>0.05),respectively.Similarly,the disease control rates in these two cohorts were 65.0%and 82.1%(P>0.05),respectively.The median progression-free survival was 5.0 months in one group and 11.2 months in the other group,with a hazard ratio of 0.40 and a significance level(P)less than 0.05.The median overall survival was 12.0 months in one group and 19.0 months in the other group,with a significance level(P)greater than 0.05.CONCLUSION Immunotherapy is less effective in GC patients with liver metastases compared to those without liver metastasis.

Dysregulation of histone deacetylases(HDACs) is closely related to tumor development and progression. As promising anticancer targets, HDACs have gained a great deal of research interests and two decades of effort has led to the approval of five HDAC inhibitors(HDACis). However, currently traditional HDACis, although effective in approved indications, exhibit severe off-target toxicities and low sensitivities against solid tumors, which have urged the development of next-generation of HDACi. This review investigates the biological functions of HDACs, the roles of HDACs in oncogenesis, the structural features of different HDAC isoforms, isoform-selective inhibitors, combination therapies, multitarget agents and HDAC PROTACs. We hope these data could inspire readers with new ideas to develop novel HDACi with good isoform selectivity, efficient anticancer effect, attenuated adverse effect and reduced drug resistance.

Protective effects of IRG1/itaconate on acute colitis through the inhibition of gasdermins-mediated pyroptosis and inflammation response

Inflammatory bowel disease(IBD)is a chronic relapsing gastrointestinal disorder,while the treatment effect is not satisfactory.Immune responsive gene 1(IRG1)is a highly ex-pressed gene in macrophage in response to inflammatory response and catalyzes the production of itaconate.Studies have reported that IRG1/itaconate has a significant antioxidant effect.This study aimed to investigate the effect and mechanism of IRG1/itaconate on dextran sulfate so-dium(DSS)-induced colitis in vivo and in vitro.In vivo experiments,we found IRG1/itaconate ex-erted protective effects against acute colitis by increasing mice weight,the length of colon,reducing disease activity index and colonic inflammation.Meanwhile,IRG1 deletion aggravated the macrophages/CD4+/CD8+T-cell accumulation,and increased the release of interleukin(IL)-1b,tumor necrosis factor-a(TNF-a),IL-6,the activation of nuclear factor-kB(NF-kB)/mitogen-activated protein kinase(MAPK)signaling pathway,and gasdermin D(GSDMD)mediated pyrop-tosis.Four-octyl itaconate(4-OI),a derivative of itaconate,attenuated these changes,therefore relieved DSS-induced colitis.In vitro experiment,we found 4-OI inhibited the reactive oxygen species production,thereby inhibiting the activation of MAPK/NF-kB signaling pathway in RAW264.7 and murine bone-marrow-derived macrophages.Simultaneously,we found 4-OI inhib-ited caspase1/GSDMD-mediated pyroptosis to reduce the release of cytokines.Finally,we found anti-TNF-a agent reduced the severity of DSS-induced colitis and inhibited gasdermin E(GSDME)-mediated pyroptosis in vivo.Meanwhile,our study revealed that 4-OI inhibited caspase3/GSDME-mediated pyroptosis induced by TNF-a in vitro.Taken together,IRG1/itaconate exerted a pro-tective role in DSS-induced colitis by inhibiting inflammatory response and GSDMD/GSDME-medi-ated pyroptosis,which could be a promising candidate for IBD therapy.

Many years of fundamental studies on viral genome packaging motors have led to fruitful applications.The double-stranded DNA(dsDNA)viruses package their genomes into preformed protein shells via nanomotors including several elegant and meticulous coaxial modules.The motor is geared by the hexameric RNA ring.An open washer displayed as hexametric string of phi29 motor ATPase has been reported.The open washer linked into a filament as a queue with left-handed chirality along the dsDNA chain.It was found that a free 5′-and 3′-dsDNA end is not required for one gp16 dimer and four monomers to assemble into the hexametric washer on dsDNA.The above studies have inspired several applications in nanotechnology and nanomedicine.These applications include:(i)studies on the precision motor channels have led to their application in the single pore sensing;(ii)investigations into the hand-in-hand integration of the hexametric pRNA ring have resulted in the emergence of the new field of RNA nanotechnology;and(iii)the studies on the motor stoichiometry of homologous multi-subunits that subsequently have inspired the discovery of new methods in highly potent drug development.This review focuses on the structure and function of the viral DNA packaging motors and describes how fundamental studies inspired various applications.Given these advantages,more nanotechnological and biomedical applications using bacteriophage motor components are expected.

Aero-optic imaging deviation research is carried out for infrared-guided vehicle with cone-head side window,with a focus on the propagation characteristics of light in an aero-optic flow field.When the light entering the aero-optic flow field from the free-stream should be close to the normal,numerous data indicate that the light is refracted away from the normal.This paper divides the aero-optic flow field into two parts and uses the gas density distribution in the aero-optic flow field to propose the hypothesis that there are two modes of refraction when light propagates through the flow field.The results show that light propagates from the optically denser medium to the optically thinner medium after passing through the shock wave and eventually leads to refraction away from the normal when the light enters the aero-optic flow field.

BACKGROUND Acute gastrointestinal bleeding is an emergency condition that can lead to significant morbidity and mortality.Embolization is considered the preferred therapy in the treatment of lower gastrointestinal bleeding when it is unrealistic to perform the surgery or vasopressin infusion in this population.Treatment of acute lower gastrointestinal(GI)bleeding(any site below the ligament of Treitz)using this technique has not reached a consensus,because of the belief that the risk of intestinal infarction in this condition is extremely high.The purpose of the study is to evaluate the effectiveness and safety of this technique in a retrospective group of patients who underwent embolization for acute lower GI bleeding.AIM To evaluate the efficacy and safety of super-selective arterial embolization in the management of acute lower GI bleeding.METHODS A series of 31 consecutive patients with angiographically demonstrated small intestinal or colonic bleeding was retrospectively reviewed.The success rate and complication rate of super-selective embolization were recorded.RESULTS Five out of thirty-one patients(16.1%)could not achieve sufficiently selective catheterization to permit embolization.Initial control of bleeding was achieved in 26 patients(100%),and relapsed GI bleeding occurred in 1 of them at 1 wk after the operation.No clinically apparent bowel infarctions were observed in patients undergoing embolization.CONCLUSION Super-selective embolization is a safe therapeutic method for acute lower GI bleeding,and it is suitable and effective for many patients suffering this disease.Importantly,careful technique and suitable embolic agent are essential to the successful operation.

Conotoxins are marine peptide toxins from marine cone snails.Theα-conotoxin RegIIA can selectively act on human(h)α3β4 nicotinic acetylcholine receptor(nAChR),and is an important lead for drug development.The high-resolution cryo-electron microscopy structure of theα3β4 nAChR demonstrates several carbohydrates are located near the orthosteric binding sites,which may affectα-conotoxin binding.Oligosaccharide chains can modify the physical and chemical properties of proteins by changing the conformation,hydrophobicity,quality and size of the protein.The purpose of this study is to explore the effect of oligosaccharide chains on the binding modes and activities of RegIIA and its derivatives at hα3β4 nAChRs.Through computational simulations,we designed and synthesized RegIIA mutants at position 14 to explore the importance of residue H14 to the activity of the peptide.Molecular dynamics simulations suggest that the oligosaccharide chains affect the binding of RegIIA at the hα3β4 nAChR through direct interactions with H14 and by affecting the C-loop conformation of the binding sites.Electrophysiology studies on H14 analogues suggest that in addition to forming direct interactions with the carbohydrates,the residue might play an important role in maintaining the conformation of the peptide.Overall,this study further clarifies the structure–activity relationship ofα-conotoxin RegIIA at the hα3β4 nAChR and,also provides important experimental and theoretical basis for the development of new peptide drugs.
