The biogenesis and functions of extrachromosomal circular DNA(eccDNA)have been studied for several decades.However,the heterogeneity of eccDNA is largely ignored.In this study,we purified and sequenced eccDNA and RNA using a method that simultaneously extracts DNA and RNA from cultured cells treated with iron nanoparticles.We discovered that the characteristics of eccDNA are altered by iron stress,particularly on genic and CpG islands regions,suggesting a biogenesis mechanism induced by transcription.

Four species of the genus Karstsinnectes,which live in dark,low-energy groundwater ecosystems,are currently recorded,mainly distributed in the upstream of the Pearl River,the Youjiang River and the Hongshui River.Completing biodiversity surveys is particularly difficult due to the particular cave habitats inhabited by these species.In this work,we describe a new cave fish of the genus Karstsinnectes,namely K.cehengensis luo,Zhao&Zhou,sp.nov.,collected from the Beipanjiang River,the upstream of the Hongshui River.Genetically,the four individuals of the new species form a phylogenetically separate lineage,as the sister clade of K.acridorsalis,with a 9.4–9.5%genetic difference in mitochondrial Cyt b.Morphologically,the new species can be distinguished from its congeners by a combination of the following morphological characteristics:body naked,without body color;eyes reduced,diameter 1.4–3.8%of head length;anterior and posterior nostrils adjacent,anterior nostril not elongated to a barbel-like tip;lateral line incomplete;caudal fin forked;dorsal-fin rays iii-7,pectoral-fin rays i-11–12,pelvic-fin rays i-7,anal-fin rays iii-5,with 14 branched caudal-fin rays.The discovery of this new species represents the northernmost distribution of species within the genus Karstsinnectes and the first record from Guizhou Province.All currently known species of this genus inhabited in caves,making them ideal organisms for studying historical river connectivity and cave formation process.

Piezoelectric stages use piezoelectric actuators and flexure hinges as driving and amplifying mechanisms,respectively.These systems have high positioning accuracy and high-frequency responses,and they are widely used in various precision/ultra-precision positioning fields.However,the main challenge with these devices is the inherent hysteresis nonlinearity of piezoelectric actuators,which seriously affects the tracking accuracy of a piezoelectric stage.Inspired by this challenge,in this work,we developed a Hammerstein model to describe the hysteresis nonlinearity of a piezoelectric stage.In particular,in our proposed scheme,a feedback-linearization algorithm is used to eliminate the static hysteresis nonlinearity.In addition,a composite controller based on equivalent-disturbance compensation was designed to counteract model uncertainties and external disturbances.An analysis of the stability of a closed-loop system based on this feedback-linearization algorithm and composite controller was performed,and this was followed by extensive comparative experiments using a piezoelectric stage developed in the laboratory.The experimental results confirmed that the feedback-linearization algorithm and the composite controller offer improved linearization and trajectory-tracking performance.

BACKGROUND:Prompt pathogen identification can have a substantial impact on the optimization of antimicrobial treatment.The objective of the study was to assess the diagnostic value of next-generation sequencing(NGS)for identifying pathogen and its clinical impact on antimicrobial intervention in immunocompromised patients with suspected infections.METHODS:This was a retrospective study.Between January and August 2020,47 adult immunocompromised patients underwent NGS testing under the following clinical conditions:1)prolonged fever and negative conventional cultures;2)new-onset fever despite empiric antimicrobial treatment;and 3)afebrile with suspected infections on imaging.Clinical data,including conventional microbial test results and antimicrobial treatment before and after NGS,were collected.Data were analyzed according to documented changes in antimicrobial treatment(escalated,no change,or deescalated)after the NGS results.RESULTS:The median time from hospitalization to NGS sampling was 19 d.Clinically relevant pathogens were detected via NGS in 61.7% of patients(29/47),more than half of whom suffered from fungemia(n=17),resulting in an antimicrobial escalation in 53.2% of patients(25/47)and antimicrobial de-escalation in 0.2% of patients(1/47).Antimicrobial changes were mostly due to the identification of fastidious organisms such as Legionella,Pneumocystis jirovecii,and Candida.In the remaining three cases,NGS detected clinically relevant pathogens also detected by conventional cultures a few days later.The antimicrobial treatment was subsequently adjusted according to the susceptibility test results.Overall,NGS changed antimicrobial management in 55.3%(26/47)of patients,and conventional culture detected clinically relevant pathogens in 14.9% of the patients(7/47).CONCLUSION:With its rapid identification and high sensitivity,NGS could be a promising tool for identifying relevant pathogens and enabling rapid appropriate treatment in immunocompromised patients with suspected infections.

The operational stability of radial journal bearings is the bottleneck that limits the reliability of hydraulic suspension micropump. Due to self-excited vibrations, the whirl amplitude of the plain journal bearing(PJB) is large at high rotational speeds,which will accelerate wear failure. It has been proven that employing herringbone grooved journal bearing(HGJB) is an effective method to reduce the whirl amplitude and improve operational stability. However, enhancing the stability of journal bearings in micro-pumps by herringbone grooved structures has rarely been studied, and its effect needs to be verified. We validated the mechanism of the stability improvement with the CFD method and compared the dynamic characteristics of HGJB and PJB by rotor dynamics evaluation and experiment. The experimental results show that under the same conditions the whirl amplitude of the HGJB decreases by 29.61% in the x direction and by 24.09% in the y direction compared with that of the PJB. This study reveals the effect of the herringbone grooved structure on the operational stability of bearings and may provide guidance for the reliability improvement of hydraulic suspension micro-pump.

Using nanocatalysts to catalyze water electrolysis for hydrogen production is an ideal solution to address the energy crisis.The most well-adopted fabrication methods for nanocatalysts are tube furnace annealing,Hydrothermal method,etc.,hardly satisfying the trade-off among coarsening,dispersity,and particle size due to mutual restrictions.Herein,a universal,ultrafast and facile cellulose nanometer whiskers-high temperature shock（CNWHTS） method was reported for fabricating a library of ultrafine metal nanoparticles with uniform dispersion and narrow size distribution.The metal-anchor functional groups in CNW（i.e.,-OH and-COOH） and the characteristics of the HTS method for ultrafast heating and powerful quenching synergistically contribute to the successful synthesis of metal nanoparticles.As an initial demonstration,the as-prepared Pt nanocatalyst(η10 mA cm-2=51.8 mV) shows more excellent catalytic hydrogen evolution reaction（HER） performance than the Pt catalyst prepared in the tubular furnace(η10 mA cm-2=169.4 mV).This rapid and universal CNW-HTS method can pave the way for nanomanufacturing to produce high-quality metal nanoparticles,thereby expanding applications of energy conversion and electrocatalysis.

Promoted catalytic performance of Ag-Mn bimetal catalysts synthesized through reduction route

VOCs can exert great harm to both human and environment,and catalytic oxidation is believed to be an effective technique to eliminate these pollutants.In this paper,Ag-Mn bimetal catalysts with 10 wt.%of silver were synthesized using doping,impregnation,and reduction methods respectively,and then they were applied to the catalytic oxidation of benzene.Through series of characterizations it showed that the loading of silver using reduction method significantly resulted in improved physico-chemical properties of manganese oxides,such as larger surface area and pore volume,higher proportion of surface Mn~(3+)and Mn~(4+),stronger reducibility and more active of surface oxygen species,which were all beneficial to its catalytic activity.As a result,the Ag-Mn catalysts synthesized by reduction method showed a lower T_(90)value(equals to the temperature at which 90%of initial benzene was removed)of 203℃.Besides,both the used and fresh Ag-Mn catalysts synthesized by reduction method showed preferable stability in this research.

Bio-inspired Design and Inverse Kinematics Solution of an Omnidirectional Humanoid Robotic Arm with Geometric and Load Capacity Constraints

Inspired by the driving muscles of the human arm,a 4-Degree of Freedom(DOF)concentrated driving humanoid robotic arm is proposed based on a spatial double parallel four-bar mechanism.The four-bar mechanism design reduces the inertia of the elbow-driving unit and the torque by 76.65%and 57.81%,respectively.Mimicking the human pose regulation strategy that the human arm picks up a heavy object by adjusting its posture naturally without complicated control,the robotic arm features an integrated position-level closed-form inverse solution method considering both geometric and load capacity limitations.This method consists of a geometric constraint model incorporating the arm angle(φ)and the Global Configuration(GC)to avoid joint limits and singularities,and a load capacity model to constrain the feasible domain of the arm angle.Further,trajectory tracking simulations and experiments are conducted to validate the feasibility of the proposed inverse solution method.The simulated maximum output torque,maximum output power and total energy consumption of the robotic arm are reduced by up to 2.0%,13.3%,and 33.3%,respectively.The experimental results demonstrate that the robotic arm can bear heavy loads in a human-like posture,effectively reducing the maximum output torque and energy consumption of the robotic arm by 1.83%and 5.03%,respectively,while avoiding joints beyond geometric and load capacity limitations.The proposed design provides a high payload–weight ratio and an efficient pose control solution for robotic arms,which can potentially broaden the application spectrum of humanoid robots.

A technical survey on mechanism and influence factors for asphalt pavement skid-resistance

To explore the research status on mechanism of the skid-resistance for asphalt pavement, the related achievements of the rubber friction, the tire–road contact, and the influence factors were reviewed. The rubber friction was reviewed from the mechanism and rubber friction model. The tire-road contact was studied from the modeling methods, the evaluation methods, and the skid-resistance mechanisms at different conditions. The influence factors of skid-resistance were summarized from different designing processes. This review showed that the appropriate contact or evaluation model should be selected according to the required parameters. Although the finite element method is widely used, the accuracy of the model, the boundary conditions, and the load cases need to be further improved. The optimized aggregates and asphalt binders, reasonable gradation, reasonable geometric design, and strict construction quality control are necessary for good initial skid-resistance. Pavement texture as an important factor affecting skid-resistance should be considered in pavement gradation design, especially for wearing layer technology.
