ShCTR1 Interacts with ShRBOH1 to Positively Regulate Aerenchyma Formation in Saussurea inversa through ROS Mediation

Saussurea inversa is one of the Tibetan medicine with developed aerenchyma.In this study,we investigated the function of the ShCTR1 gene related to the formation of aerenchyma.The study of the occurrence and formation of aerenchyma in S.inversa has certain theoretical significance in revealing the special mechanism of alpine plants adapting to the environment.The results of yeast two-hybrid experiments showed that S.inversa ShCTR1 interacted with the ShRBOH1,which is a key producer of reactive oxygen species(ROS),and it was hypothesized that ShCTR1 regulation of aerenchyma formation in S.inversa mainly mediated by ROS.Aerenchyma induction and inhibition experiments were carried out on S.inversa seedlings,it was shown that ROS inducer(AT)and Ethene(ETH)induced the formation of aerenchyma in S.inversa well in roots,stems,and leaves,and Ethylene inhibitor(1-MCP)and ROS inhibitor DPI(diphenylene iodonium)inhibited the formation of aerenchyma of S.inversa in different degrees,which proved that ROS and ethylene(ET)were positively correlated with the formation of aerenchyma.Meanwhile,AT and ETH well induced the expression of the ShCTR1 gene,and 1-MCP and DPI inhibited the expression of the ShCTR1 gene to different degrees,which proved that ROS and ET were also positively correlated with the expression of the ShCTR1 gene.ShCTR1 was transferred into tobacco(Nicotiana tabacum L.),then repeat the above experiment.It was shown that positive rate and porosity were lowest and highest after DPI and AT treatments,respectively,it showed a positive correlation,which proved that the ShCTR1 gene was positively regulating the formation of aerenchyma.Overexpression of the ShCTR1 gene increased the content of ROS,consequently,the regulation of aerenchyma formation by the ShCTR1 gene is mainly mediated by ROS.It suggested that the formation of aerenchyma in S.inversa was related to hypoxia in the alpine environment,and ShCTR1 interacts with ShRBOH1 to positively regulate the formation of aerenchyma mainly mediated by ROS.

BACKGROUND Targeting DNA damage response(DDR)pathway is a cutting-edge strategy.It has been reported that Schlafen-11(SLFN11)contributes to increase chemosensitivity by participating in DDR.However,the detailed mechanism is unclear.AIM To investigate the role of SLFN11 in DDR and the application of synthetic lethal in esophageal cancer with SLFN11 defects.METHODS To reach the purpose,eight esophageal squamous carcinoma cell lines,142 esophageal dysplasia(ED)and 1007 primary esophageal squamous cell carcinoma(ESCC)samples and various techniques were utilized,including methylationspecific polymerase chain reaction,CRISPR/Cas9 technique,Western blot,colony formation assay,and xenograft mouse model.RESULTS Methylation of SLFN11 was exhibited in 9.15%of(13/142)ED and 25.62%of primary(258/1007)ESCC cases,and its expression was regulated by promoter region methylation.SLFN11 methylation was significantly associated with tumor differentiation and tumor size(both Phe ATM/CHK2 signaling pathway.Epigenetic silencing of SLFN11 was found to sensitize the ESCC cells to ATM inhibitor(AZD0156),both in vitro and in vivo.CONCLUSION SLFN11 is frequently methylated in human ESCC.Methylation of SLFN11 is sensitive marker of ATM inhibitor in ESCC.

The tumor heterogeneity results in different clinical characteristics of tumor patients,so precise therapy is a popular treatment strategy clinically.Microsatellite instability(MSI)is found in 10%-15%of colorectal cancer(CRC)patients and exhibits different clinical outcomes than microsatellite stable(MSS)patients.Surprisingly,MSI patients have a better prognosis than patients with stable microsatellites.Msl patients exhibit clinical features that are not responsive to chemotherapy,however,Msl patients showed excellent responses to immunotherapy.'Although the subtype classification of MSI is beneficial to the clinical treatment of CRC patients,the mechanism of MSl remains unclear.

Beams typically do not travel through the magnet centers because of errors in storage rings.The beam deviating from the quadrupole centers is affected by additional dipole fields due to magnetic field feed-down.Beam-based alignment(BBA)is often performed to determine a golden orbit where the beam circulates around the quadrupole center axes.For storage rings with many quadrupoles,the conventional BBA procedure is time-consuming,particularly in the commissioning phase,because of the necessary iterative process.In addition,the conventional BBA method can be affected by strong coupling and the nonlinearity of the storage ring optics.In this study,a novel method based on a neural network was proposed to determine the golden orbit in a much shorter time with reasonable accuracy.This golden orbit can be used directly for operation or adopted as a starting point for conventional BBA.The method was demonstrated in the HLS-II storage ring for the first time through simulations and online experiments.The results of the experiments showed that the golden orbit obtained using this new method was consistent with that obtained using the conventional BBA.The development of this new method and the corresponding experiments are reported in this paper.

The distribution of oil and gas resources is intricately connected to the underlying structure of the lithosphere.Therefore,investigating the characteristics of lithospheric thickness and its correlation with oil and gas basins is highly important.This research utilizes recently enhanced geological–geophysical data,including topographic,geoid,rock layer thickness,variable rock layer density,and interface depth data.Employing the principles of lithospheric isostasy and heat conduction,we compute the laterally varying lithospheric thickness in the China seas and adjacent areas.From these results,two pivotal parameters for different types of oil and gas basins were statistically analyzed:the minimum lithospheric thickness and the relative fluctuation in lithospheric thickness.A semiquantitative analysis was used to explore the connection between these parameters and the hydrocarbon abundance within the oil and gas basins.This study unveils distinct variations in lithospheric thickness among basins,with oil and gas rich basins exhibiting a thicker lithosphere in the superimposed basins of central China and a thinner lithosphere in the rift basins of eastern China.Notably,the relative fluctuations in lithospheric thickness in basins demonstrate significant disparities:basins rich in oil and gas often exhibit greater thickness fluctuations.Additionally,in the offshore basins of China,a conspicuous negative linear correlation is observed between the minimum lithospheric thickness and the relative fluctuation in lithospheric thickness.This study posits that deep-seated thermal upwelling results in lithospheric undulations and extensional thinning in oil and gas basins.Concurrently,sustained deep-seated heat influences sedimentary materials in basins,creating favorable conditions for oil and gas generation.The insights derived from this study contribute to a quantitative understanding of the intricate relationships between deep lithospheric structures and oil and gas basins.These findings provide valuable guidance for fut

Two-dimensional transition metal dichalcogenides(TMDs)are needed in highperformance piezoresistive sensors due to their strong strain-induced bandgap modification and thereby large gauge factors.However,integrating a conventional high-temperature chemical vapor deposition(CVD)-grown TMD with a flexible substrate necessitates a transfer process that inevitably degrades the sensing properties of the TMDs and increases the overall fabrication complexity.We present a high-performance piezoresistive strain sensor that employs largearea PdSe_(2) films grown directly on polyimide(PI)substrates via plasma-assisted selenization of a sputtered Pd film.The reliable strain transfer from the substrate to the PdSe_(2) film ensures an outstanding strain-sensing capability of the sensor.Specifically,the sensors have a gauge factor of up to315±2.1,a response time under 25 ms,a detection limit of 8×10^(-6),and an exceptional stability of over 104 loadingunloading cycles.By attaching the sensors to the skin surface,we demonstrate their application for measuring physiological parameters in health care monitoring,including motion,voice,and arterial pulse vibration.Furthermore,using the PdSe_(2) film sensor combined with deep learning technology,we achieved intelligent recognition of artery temperature from arterial pulse signals with only a 2%difference between predicted and actual temperatures.The excellent sensing performance,together with the advantages of low-temperature fabrication and simple device structure,make the PdSe_(2) film sensor promising for wearable electronics and health care sensing systems.

Synergistic oxidation-reforming of biomass for high quality syngas production based on a bifunctional catalyst

Conventional O_(2)gasification for low-rank biomass/sludge conversion is prone to high CO_(2)concentrations in the syngas because of its high O content and low calorific value.This study establishes a synergistic oxidationreforming reaction route for the conversion of low-rank carbon-containing resources into high-quality syngas.The efficient oxidation-reforming reaction is based on the bifunctional catalyst NiO-Fe_(2)O_(3)/Al_(2)O_(3),which includes Fe_(2)O_(3) oxidation sites and NiO reforming sites.Hydrogen temperature-programmed reduction,together with X-ray diffraction and X-ray photoelectron spectroscopy experiments,demonstrated that the two functional active sites have strong interactions with the support,leading to efficient cooperation between the oxidation reaction and reforming reaction with regards to both the reaction sequence and C/H/O element balance.Syngas produced from biomass/sludge based on oxidation-reforming reactions has an extremely low CO_(2)concentration of approximately 3%,and the valid gas(CO,H_(2))concentration exceeds 95%.The valid gas yield of walnut shell reached 1452.9 mL/g,the total gas yield was 1507.2 mL/g,and the H_(2)/CO ratio was 1.02,which are all very close to the theoretical maximum values of 1553.1 mL/g and 1.01,respectively,demonstrating that the inherent CO_(2)/H_(2)O along with CH4/tar species were efficiently converted to H_(2)and CO through oxidation-reforming reactions.During a 60-cycle test,NiO-Fe_(2)O_(3)/Al_(2)O_(3) exhibited good redox stability.

ransforming growth factor(TGF)-βis a multifunctional cytokine expressed by almost every tissue and cell type.The signal transduction of TGF-βcan stimulate diverse cellular responses and is particularly critical to embryonic development,wound healing,tissue homeostasis,and immune homeostasis in health.The dysfunction of TGF-βcan play key roles in many diseases,and numerous targeted therapies have been developed to rectify its pathogenic activity.In the past decades,a large number of studies on TGF-βsignaling have been carried out,covering a broad spectrum of topics in health,disease,and therapeutics.Thus,a comprehensive overview of TGF-βsignaling is required for a general picture of the studies in this field.In this review,we retrace the research history of TGF-βand introduce the molecular mechanisms regarding its biosynthesis,activation,and signal transduction.We also provide deep insights into the functions of TGF-βsignaling in physiological conditions as well as in pathological processes.TGF-β-targeting therapies which have brought fresh hope to the treatment of relevant diseases are highlighted.Through the summary of previous knowledge and recent updates,this review aims to provide a systematic understanding of TGF-βsignaling and to attract more attention and interest to this research area.

The significant increase in the demand for biomass waste treatment after garbage classification has led to housefly larvae treatment becoming an attractive treatment option.It can provide a source of protein while treating biomass waste,which means that nutrients can be returned to the natural food chain.However,the performance of this technology in terms of its environmental impacts is still unclear,particularly with regards to global warming potential(GWP).This study used a life cycle assessment(LCA)approach to assess a housefly larvae treatment plant with a treatment capacity of 50 tons of biomass waste per day.The LCA results showed that the 95% confidence intervals for the GWP in summer and winter were determined to be 24.46-32.81 kg CO_(2) equivalent(CO_(2)-eq)/ton biomass waste and5.37-10.08 kg CO_(2)-eq/ton biomass waste,respectively.The greater GWP value in summer is due to the longer ventilation time and higher ventilation intensity in summer,which consumes more power.The main GWP contributions are from(1)electricity needs(accounting for 78.6% of emissions in summer and 70.2%in winter)and(2)product substitution by mature housefly larvae and compost(both summer and winter accounting for 96.8% of carbon reduction).
