Bioinspired metal-organic framework nanozyme reinforced with thermosensitive hydrogel for regulating inflammatory responses in Parkinson’s disease

Parkinson’s disease(PD)is a prevalent neurodegenerative disorder accompanied by movement disorders and neuroinflammatory injury.Anti-inflammatory intervention to regulate oxidative stress in the brain is beneficial for managing PD.However,traditional natural antioxidants have failed to meet the clinical treatment demands due to insufficient activity and sustainability.Herein,Cu-doping zeolite imidazolate framework-8(ZIF-8)nanozyme is designed to simulate Cu/Zn superoxide dismutase(SOD)by biomimetic mineralization.The nanozyme composite is then integrated into thermosensitive hydrogel(poly(lactic-co-glycolic acid)-poly(ethylene glycol)-poly(lactic-co-glycolic acid)(PLGA-PEG-PLGA))to form an effective antioxidant system(Cu-ZIF@Hydrogel).The thermosensitive hydrogel incorporating nanozymes demonstrate distinct viscoelastic properties aimed at enhancing local nanozyme adhesion,prolonging nanozyme retention time,and modulating antioxidant activity,thus significantly improving the bioavailability of nanozymes.At the cellular and animal levels of PD,we find that Cu-ZIF@Hydrogel bypass the blood-brain barrier and efficiently accumulate in the nerve cells.Moreover,the Cu-ZIF@Hydrogel significantly alleviate the PD’s behavioral and pathological symptoms by reducing the neuroinflammatory levels in the lesion site.Therefore,the hydrogel-incorporating nanozyme system holds great potential as a simple and reliable avenue for managing PD.

Spatiotemporal Changes of Snow Depth in Western Jilin,China from 1987 to 2018

Seasonal snow cover is a key global climate and hydrological system component drawing considerable attention due to glob-al warming conditions.However,the spatiotemporal snow cover patterns are challenging in western Jilin,China due to natural condi-tions and sparse observation.Hence,this study investigated the spatiotemporal patterns of snow cover using fine-resolution passive mi-crowave(PMW)snow depth(SD)data from 1987 to 2018,and revealed the potential influence of climate factors on SD variations.The results indicated that the interannual range of SD was between 2.90 cm and 9.60 cm during the snowy winter seasons and the annual mean SD showed a slightly increasing trend(P>0.05)at a rate of 0.009 cm/yr.In snowmelt periods,the snow cover contributed to an increase in volumetric soil water,and the change in SD was significantly affected by air temperature.The correlation between SD and air temperature was negative,while the correlation between SD and precipitation was positive during December and March.In March,the correlation coefficient exceeded 0.5 in Zhenlai,Da’an,Qianan,and Qianguo counties.However,the SD and precipitation were neg-atively correlated over western Jilin in October,and several subregions presented a negative correlation between SD and precipitation in November and April.

Teneligliptin mitigates diabetic cardiomyopathy by inhibiting activation of the NLRP3 inflammasome

BACKGROUND Diabetic cardiomyopathy(DCM),which is a complication of diabetes,poses a great threat to public health.Recent studies have confirmed the role of NLRP3(NOD-like receptor protein 3)activation in DCM development through the inflammatory response.Teneligliptin is an oral hypoglycemic dipeptidyl peptidase-IV inhibitor used to treat diabetes.Teneligliptin has recently been reported to have anti-inflammatory and protective effects on myocardial cells.AIM To examine the therapeutic effects of teneligliptin on DCM in diabetic mice.METHODS Streptozotocin was administered to induce diabetes in mice,followed by treatment with 30 mg/kg teneligliptin.RESULTS Marked increases in cardiomyocyte area and cardiac hypertrophy indicator heart weight/tibia length reductions in fractional shortening,ejection fraction,and heart rate;increases in creatine kinase-MB(CK-MB),aspartate transaminase(AST),and lactate dehydrogenase(LDH)levels;and upregulated NADPH oxidase 4 were observed in diabetic mice,all of which were significantly reversed by teneligliptin.Moreover,NLRP3 inflammasome activation and increased release of interleukin-1βin diabetic mice were inhibited by teneligliptin.Primary mouse cardiomyocytes were treated with high glucose(30 mmol/L)with or without teneligliptin(2.5 or 5μM)for 24 h.NLRP3 inflammasome activation.Increases in CKMB,AST,and LDH levels in glucose-stimulated cardiomyocytes were markedly inhibited by teneligliptin,and AMP(p-adenosine 5‘-monophosphate)-p-AMPK(activated protein kinase)levels were increased.Furthermore,the beneficial effects of teneligliptin on hyperglycaemia-induced cardiomyocytes were abolished by the AMPK signaling inhibitor compound C.CONCLUSION Overall,teneligliptin mitigated DCM by mitigating activation of the NLRP3 inflammasome.

A Self-Healing Optoacoustic Patch with High Damage Threshold and Conversion Efficiency for Biomedical Applications

Compared with traditional piezoelectric ultrasonic devices,optoacoustic devices have unique advantages such as a simple preparation process,anti-electromagnetic interference,and wireless long-distance power supply.However,current optoacoustic devices remain limited due to a low damage threshold and energy conversion efficiency,which seriously hinder their widespread applications.In this study,using a self-healing polydimethylsiloxane(PDMS,Fe-Hpdca-PDMS)and carbon nanotube composite,a flexible optoacoustic patch is developed,which possesses the self-healing capability at room temperature,and can even recover from damage induced by cutting or laser irradiation.Moreover,this patch can generate high-intensity ultrasound(>25 MPa)without the focusing structure.The laser damage threshold is greater than 183.44 mJ cm^(-2),and the optoacoustic energy conversion efficiency reaches a major achievement at 10.66×10^(-3),compared with other carbon-based nanomaterials and PDMS composites.This patch is also been successfully examined in the application of acoustic flow,thrombolysis,and wireless energy harvesting.All findings in this study provides new insight into designing and fabricating of novel ultrasound devices for biomedical applications.

Multispectral point cloud superpoint segmentation

The multitude of airborne point clouds limits the point cloud processing efficiency.Superpoints are grouped based on similar points,which can effectively alleviate the demand for computing resources and improve processing efficiency.However,existing superpoint segmentation methods focus only on local geometric structures,resulting in inconsistent spectral features of points within a superpoint.Such feature inconsistencies degrade the performance of subsequent tasks.Thus,this study proposes a novel Superpoint Segmentation method that jointly utilizes spatial Geometric and Spectral Information for multispectral point cloud superpoint segmentation(GSI-SS).Specifically,a similarity metric that combines spatial geometry and spectral information is proposed to facilitate the consistency of geometric structures and object attributes within segmented superpoints.Following the formation of the primary superpoints,an intersuperpoint pointexchange mechanism that maximizes feature consistency within the final superpoints is proposed.Experiments are conducted on two real multispectral point cloud datasets,and the proposed method achieved higher recall,precision,F score,and lower global consistency and feature classification errors.The experimental results demonstrate the superiority of the proposed GSI-SS over several state-of-the-art methods.

Tunable VO_(2) cavity enables multispectral manipulation from visible to microwave frequencies

Optical materials capable of dynamically manipulating electromagnetic waves are an emerging field in memories,optical modulators,and thermal management.Recently,their multispectral design preliminarily attracts much attention,aiming to enhance their efficiency and integration of functionalities.However,the multispectral manipulation based on these materials is challenging due to their ubiquitous wavelength dependence restricting their capacity to narrow wavelengths.In this article,we cascade multiple tunable optical cavities with selective-transparent layers,enabling a universal approach to overcoming wavelength dependence and establishing a multispectral platform with highly integrated functions.Based on it,we demonstrate the multispectral(ranging from 400 nm to 3 cm),fast response speed(0.9 s),and reversible manipulation based on a typical phase change material,vanadium dioxide.Our platform involves tandem VO_(2)-based Fabry–Pérot(F-P)cavities enabling the customization of optical responses at target bands independently.It can achieve broadband color-changing capacity in the visible region(a shift of~60 nm in resonant wavelength)and is capable of freely switching between three typical optical models(transmittance,reflectance,and absorptance)in the infrared to microwave regions with drastic amplitude tunability exceeding 0.7.This work represents a state-of-art advance in multispectral optics and material science,providing a critical approach for expanding the multispectral manipulation ability of optical systems.

Causal roles of gut microbiota in cholangiocarcinoma etiology suggested by genetic study

BACKGROUND Cholangiocarcinoma(CCA)is a highly malignant biliary tract cancer with poor prognosis.Previous studies have implicated the gut microbiota in CCA,but evidence for causal mechanisms is lacking.AIM To investigate the causal relationship between gut microbiota and CCA risk.METHODS We performed a two-sample mendelian randomization study to evaluate potential causal associations between gut microbiota and CCA risk using genome-wide association study summary statistics for 196 gut microbial taxa and CCA.Genetic variants were used as instrumental variables.Multiple sensitivity analyses assessed result robustness.RESULTS Fifteen gut microbial taxa showed significant causal associations with CCA risk.Higher genetically predicted abundance of genus Eubacteriumnodatum group,genus Ruminococcustorques group,genus Coprococcus,genus Dorea,and phylum Actinobacteria were associated with reduced risk of gallbladder cancer and extrahepatic CCA.Increased intrahepatic CCA risk was associated with higher abundance of family Veillonellaceae,genus Alistipes,order Enterobacteriales,and phylum Firmicutes.Protective effects against CCA were suggested for genus Collinsella,genus Eisenbergiella,genus Anaerostipes,genus Paraprevotella,genus Parasutterella,and phylum Verrucomicrobia.Sensitivity analyses indicated these findings were reliable without pleiotropy.CONCLUSION This pioneering study provides novel evidence that specific gut microbiota may play causal roles in CCA risk.Further experimental validation of these candidate microbes is warranted to consolidate causality and mechanisms.

Synchronous condensers(SCs)are generally used at the receiving-end stations of ultra-high-voltage direct current(UHVDC)transmission systems due to their strong reactive power support and flexible regulation of reactive power according to the interconnected grids operating conditions.In this paper,different starting control schemes of a SC integrated power grid are investigated providing four main contributions:1)The principle of reactive power support of the SC on the interconnected power grid is analytically studied,providing the establishment of mathematical models.2)Four different starting control schemes are developed for the initialization and SC integration,i.***.in Scheme 1,a preset initial falling speed is directly utilized without initialization;in Scheme 2,a black start sequential control approach with a static frequency converter(SFC)is proposed;in Scheme 3,PI/PD/PID controllers are respectively applied for the excitation device at the speed-falling stage;in Scheme 4,a pre-insertion approach of an energy absorption component with R/L/RL is utilized to suppress the surges at the SC integration instant.3)The dynamic behaviors of four different starting schemes at specific operating stages are evaluated.4)The success rate of SC integration is analyzed to evaluate starting control performance.Performance of the SC interconnected system with four different starting control schemes is evaluated in the timedomain simulation environment PSCAD/EMTDC^(TM).The results prove the superiority of the proposed starting control approach in Scheme 4.

Circadian disturbance induces erectile dysfunction by impairing endothelial function

In order to explore the impact of circadian disturbance on erectile function,we randomly divided 24 adult male rats into groups of control(light on at 8:00 a.***.and off at 8:00 p.m.),dark/dark(DD;constant dark),light/light(LL;constant light),and shift dark/light(DL;light off at 8:00 a.***.and on at 8:0o p.m.).Four weeks later,erectile function was measured and corpora cavernosa were harvested for analysis.The maximum intracavernous pressure(mlcP)and mCP/mean arterial pressure(MAP)ratio in the DD,LL,and DL groups were significantly lower than that in the control group.The LL and DL groups showed significantly attenuated endothelial nitric oxide synthase(eNOS),while DD,LL,and DL showed reduced neuronal nitric oxide synthase(nNOS)at both mRNA and protein levels.The production of nitric oxide(NO)and cyclic guanosinemonophosphate(cGMP)was inhibited by altered light/dark cycles to varying degrees.Circadian disturbance impaired endothelial function and contributed to erectile dysfunction.For the core circadian elements,mRNA expression of circadian locomotor output cycles kaput(Clock)and brain/muscle aryl-hydrocarbon receptor nuclear translocator-like protein 1(Bmal1)was elevated in the DL group,but their protein expression was not significantly changed.DD,LL,and DL increased period 1(Per1)and Per3 levels,while LL and DL increased PER1 levels.No significant difference was found for Per2levels,and PER2 and PER3 concentrations were not significantly changed.Moreover,LL and DL significantly increased cryptochrome-1(CRY1)and CRY2 at both mRNA and protein levels.The altered light/dark rat model showed that circadian disturbance contributed to erectile dysfunction probably by impairing endothelial function.Meanwhile,the core circadian elements were detected in the corpora cavernosa,but these were disrupted.However,which circadian element regulates erectile function and how it works need further analysis.

The limited infiltration and persistence of chimeric antigen receptor(CAR)-T cells is primarily responsible for their treatment deficits in solid tumors. Here, we present a three-dimensional scaffold, inspired by the physiological process of T-cell proliferation in lymph nodes. This scaffold gathers the function of loading,delivery, activation and expansion for CAR-T cells to enhance their therapeutic effects on solid tumors. This porous device is made from poly(lactic-co-glycolic acid) by a microfluidic technique with the modification of T-cell stimulatory signals, including anti-CD3, anti-CD28 antibodies, as well as cytokines. This scaffold fosters a 50-fold CAR-T cell expansion in vitro and a 15-fold cell expansion in vivo. Particularly, it maintains long-lasting expansion of CAR-T cells for up to 30 days in a cervical tumor model and significantly inhibits the tumor growth. This biomimetic delivery strategy provides a versatile platform of cell delivery and activation for CAR-T cells in treating solid tumors.
