Objective:Recent studies have shown the important influence of various micro factors on the general biological activity and function of endothelial cells (ECs).Vascular endothelial growth factor (VEGF) and angiogenin (ANG) are classic micro factors that promote proliferation,differentiation,and migration of ECs.The underlying pathophysiological mechanisms and related pathways of these micro factors remain the focus of current research.Data sources:An extensive search was undertaken in the PubMed database by using keywords including "micro factors" and "endothelial cell." This search covered relevant research articles published between January 1,2007 and December 31,2018.Study selection:Original articles,reviews,and other articles were searched and reviewed for content on micro factors of ECs.Results:VEGF and ANG have critical functions in the occurrence,development,and status of the physiological pathology of ECs.Other EC-associated micro factors include interleukin 10,tumor protein P53,nuclear factor kappa B subunit,interleukin 6,and tumor necrosis factor.The results of Gene Ontology analysis revealed that variations were mainly enriched in positive regulation of transcription by the RNA polymerase Ⅱ promoter,cellular response to lipopolysaccharides,negative regulation of apoptotic processes,external side of the plasma membrane,cytoplasm,extracellular regions,cytokine activity,growth factor activity,and identical protein binding.The results of the Kyoto Encyclopedia of Genes and Genomes analysis revealed that micro factors were predominantly enriched in inflammatory diseases.Conclusions:In summary,the main mediators,factors,or genes associated with ECs include VEGF and ANG.The effect of micro factors on ECs is complex and multifaceted.This review summarizes the correlation between ECs and several micro factors.

传统机械卸煤方式易损坏敞车车体,敞车破损严重危害煤炭运输安全,破损敞车维修费用高昂,对比传统卸煤方法优缺点,提出使用卸煤机器人方式进行敞车卸煤。作为一个新兴的研究领域,卸煤机器人在国内外的研究尚处于起步阶段,并没有成熟的技术体系可以借鉴,因此对现有的研究进行系统的分析和总结。提出智能卸煤机器人概念,根据卸煤机器人智能化的需求提出了卸煤机器人所需突破的关键技术问题。详细探讨智能卸煤机器人自动化作业框架,对框架进行模块化处理,分成不同的子系统,对不同子系统进行了定义与阐述分析,强调了自动化作业系统的功能主要依托于关键技术。研究结果表明:今后国内中小型露天场站敞车卸煤将朝着自动化、智能化的方向发展,智能卸煤机器人为国内中小型露天场站智能化、自动化卸煤提供了一个新的思路。

Osteoarthritis is recognised to be an interactive pathological process involving the cartilage, subchondral bone and synovium. The signals from the synovium play an important role in cartilage metabolism, but little is known regarding the influence of the signalling from bone. Additionally, the collagenases and stromelysin-1 are involved in cartilage catabolism through mitogen-activated protein kinase （MAPK） signalling, but the role of the gelatinases has not been elucidated. Here, we studied the influence of osteoclastic signals on chondrocytes by characterising the expression of interleukin-1β （IL-1β）-induced gelatinases through MAPK signalling. We found that osteoclast-conditioned media attenuated the gelatinase activity in chondrocytes. However, IL-1β induced increased levels of gelatinase activity in the conditioned media group relative to the mono-cultured chondrocyte group. More specifically, IL-1β restored high levels of gelatinase activity in c-Jun N-terminal kinase inhibitor-pretreated chondrocytes in the conditioned media group and led to lower levels of gelatinase activity in extracellular signal-regulated kinase or p38 inhibitor-pretreated chondrocytes. Gene expression generally correlated with protein expression. Taken together, these results show for the first time that signals from osteoclasts can influence gelatinase activity in chondrocytes. Furthermore, these data show that IL-11~ restores gelatinase activity through MAPK inhibitors; this information can help to increase the understanding of the gelatinase modulation in articular cartilage.

Vitamin K refers to a group of structurally similar vitamins that are essential for proper blood coagulation,as well as bone and cardiovascular health.Previous studies have indicated that vitamin K may also have anti-inflammatory properties,although the underlying mechanisms of its anti-inflammatory effects remain unclear.The NLRP3 inflammasome is a multiprotein complex,and its activation leads to IL-1βand IL-18 secretion and contributes to the pathogenesis of various human inflammatory diseases.Here,we show that synthetic vitamins K3 and K4 are selective,potent inhibitors of the NLRP3 inflammasome and specifically block the interaction between NLRP3 and ASC,thereby inhibiting NLRP3 inflammasome assembly.Moreover,we show that treatment with vitamin K3 or K4 attenuates the severity of inflammation in a mouse model of peritonitis.Our results demonstrate that vitamins K3 and K4 exert their anti-inflammatory effects by inhibiting NLRP3 inflammasome activation and indicate that vitamin K supplementation may be a treatment option for NLRP3-associated inflammatory diseases.

Neutrophil-mimicking therapeutic nanoparticles for targeted chemotherapy of pancreatic carcinoma

Due to the critical correlation between inflammation and carcinogenesis, a therapeutic candidate with anti-inflammatory activity may find application in cancer therapy. Here, we report the therapeutic efficacy of celastrol as a promising candidate compound for treatment of pancreatic carcinoma via na?ve neutrophil membrane-coated poly(ethylene glycol) methyl ether-block-poly(lactic-co-glycolic acid)(PEG-PLGA) nanoparticles. Neutrophil membranecoated nanoparticles(NNPs) are well demonstrated to overcome the blood pancreas barrier to achieve pancreas-specific drug delivery in vivo. Using tumor-bearing mice xenograft model, NNPs showed selective accumulations at the tumor site following systemic administration as compared to nanoparticles without neutrophil membrane coating. In both orthotopic and ectopic tumor models,celastrol-loaded NNPs demonstrated greatly enhanced tumor inhibition which significantly prolonged the survival of tumor bearing mice and minimizing liver metastases. Overall, these results suggest that celastrol-loaded NNPs represent a viable and effective treatment option for pancreatic carcinoma.

Rheumatoid arthritis(RA)is a chronic inflammatory and destructive arthropathy with a high deformity rate.Despite numerous studies and clinical trials,no curative treatment is available for large weight-bearing joints.Intra-articular(IA)injections could deliver high concentrations of drug to the afflicted joint and improve the drug efficacy while reducing systemic toxicity.However,free drugs are rapidly cleared from synovial fluid and do not significantly halt the progression of joint disease.Herein,a phospholipids-based controlledrelease gel was prepared for sustained IA delivery of celastrol(CEL)and the therapeutic efficiencywas evaluated in a rheumatoid arthritis rabbitmodel.The CEL-loaded gel(CEL-gel)contained up to 70%phospholipids yetwas easy to inject.After injecting into the joint cavity,CEL-gel achieved sol to gel phase transition without special stimuli and gelling agent.In vitro release and in vivo pharmacokinetic studies evidenced the stable and sustained release action of CEL-gel.A single IA injection of CEL-gel could maintain therapeutic efficiency for about 25 d and showed much better anti-arthritic efficacy compared to repeated injections of free drug solution(CEL-sol).Furthermore,the IA injection of CEL-gel greatly reduced the systemic toxicity of CEL.With good biocompatibility and biodegradability,CEL-gel might be a promising IA drug delivery system.

The cancer stem cell(CSC)theory is gaining increasing attention from researchers and has become an important focus of cancer research.According to the theory,a minority population of cancer cells is capable of self-renewal and generation of differentiated progeny,termed cancer stem cells(CSCs).Understanding the properties and characteristics of CSCs is key to future study on cancer research,such as the isolation and identification of CSCs,the cancer diagnosis,and the cancer therapy.Standard oncology treatments,such as chemotherapy,radiotherapy and surgical resection,can only shrink the bulk tumor and the tumor tends to relapse.Thus,therapeutic strategies that focus on targeting CSCs and their microenvironmental niche address the ineffec-tiveness of traditional cancer therapies to eradicate the CSCs that otherwise result in therapy resistance.The combined use of traditional therapies with targeted CSC-specific agents may target the whole cancer and offer a promising strategy for lasting treatment and even cure.

Characteristics of oral methicillin-resistant Staphylococcus epidermidis isolated from dental plaque

The oral microbial community is widely regarded as a latent reservoir of antibiotic resistance genes. This study assessed the molecular epidemiology, susceptibility profile, and resistance mechanisms of 35 methicillin-resistant Staphylococcus epidermidis(MRSE) strains isolated from the dental plaque of a healthy human population. Broth microdilution minimum inhibitory concentrations(MICs) revealed that all the isolates were nonsusceptible to oxacillin and penicillin G. Most of them were also resistant to trimethoprim(65.7%) and erythromycin(54.3%). The resistance to multiple antibiotics was found to be largely due to the acquisition of plasmid-borne genes. The mec A and dfr A genes were found in all the isolates, mostly dfr G(80%), aac A-aph D(20%), aad D(28.6%), aph A3(22.9%), msr A(5.7%), and the erm C gene(14.3%). Classical mutational mechanisms found in these isolates were mainly efflux pumps such as qac A(31.4%), qac C(25.7%), tet K(17.1%), and nor A(8.6%). Multilocus sequence type analysis revealed that sequence type 59(ST59) strains comprised 71.43% of the typed isolates, and the e BURST algorithm clustered STs into the clonal complex 2-II(CC2-II). The staphyloccoccal cassette chromosome mec(SCCmec) type results showed that 25(71.43%) were assigned to type IV. Moreover, 88.66% of the isolates were found to harbor six or more biofilm-associated genes. The aap, atl E, embp, sdr F, and IS256 genes were detected in all 35 isolates. This research demonstrates that biofilm-positive multipleantibiotic-resistant ST59-SCCmec IV S. epidermidis strains exist in the dental plaque of healthy people and may be a potential risk for the transmission of antibiotic resistance.
