The key role of myostatin b in somatic growth in fishes derived from distant hybridization

The basic mechanism of heterosis has not been systematically and completely *** previous studies,we obtained three economically important fishes that exhibit rapid growth,WR(WCC♀×RCC♂),WR-Ⅱ(WR♀×WCC♂),and WR-Ⅲ(WR-Ⅱ♀×4nAU♂),through distant ***,the mechanism underlying this rapid growth remains *** this study,we found that WR,WR-Ⅱ,and WR-Ⅲshowed muscle hypertrophy and higher muscle protein and fat contents compared with their parent species(RCC and WCC).Candidate genes responsible for this rapid growth were then obtained through an analysis of 12 muscle ***,the mRNA level of mstnb(myostatin b),which is a negative regulator of myogenesis,was significantly reduced in WR,WR-Ⅱ,and WR-Ⅲcompared with the parent *** verify the function of mstnb,a mstnb-deficient mutant RCC line was generated using the CRISPR-Cas9 *** average body weight of mstnb-deficient RCC at 12 months of age was significantly increased by 29.57%compared with that in wild-type ***,the area and number of muscle fibers were significantly increased in mstnb-deficient RCC,indicating hypertrophy and ***,the muscle protein and fat contents were significantly increased in mstnb-deficient *** molecular regulatory mechanism of mstnb was then revealed by transcription profiling,which showed that genes related to myogenesis(myod,myog,and myf5),protein synthesis(PI3K-AKT-mTOR),and lipogenesis(pparγand fabp3)were highly activated in hybrid fishes and mstnb-deficient *** study revealed that low expression or deficiency of mstnb regulates somatic growth by promoting myogenesis,protein synthesis,and lipogenesis in hybrid fishes and mstnb-deficient RCC,which provides evidence for the molecular mechanism of heterosis via distant hybridization.

Alzheimer’s disease (AD) remains the foremost cause of dementia and represents a significant unmet healthcare need globally. The complex pathogenesis of AD, characterized by various pathological and physiological events, has historically challenged the development of anti-AD drugs. However, recent breakthroughs in AD drug development, including the approvals of aducanumab, lecanemab, and sodium oligomannate (GV-971), have ended a nearly two-decade hiatus in the introduction of new AD drugs. These developments have addressed long-standing challenges in AD drug development, marking a substantial shift in the therapeutic landscape of AD. Moreover, natural products (NPs) have shown promise in AD drug research, with several currently under clinical investigation. Their distinct properties and mechanisms of action offer new avenues to complement and enhance existing AD treatment approaches. This review article aims to provide an overview of the recent advancements and prospects in AD therapeutics, focusing on both NPs and approved drugs.

Interface engineering via temperature-dependent self-transformation on SnS2/SnS for enhanced piezocatalysis

Heterojunction has been widely used in vibration-driven piezocatalysis for enhanced charges separation,while the weak interfaces seriously affect the efficiency during mechanical deformations due to prepared by traditional step-by-step ***,the intimate contact interfaces with shared S atoms are ingeniously constructed in SnS_(2)/SnS anchored on porous carbon by effective interface engineering,which is in-situ derived from temperature-dependent self-transformation of SnS_(2).Benefiting from intimate contact interfaces,the piezoelectricity is remarkably improved due to the larger interfacial dipole moment caused by uneven distribution of ***,vibration-induced piezoelectric polarization field strengthens the interfacial electric field to further promote the separation and migration of *** dynamic charges then transfer in porous carbon with high conductivity and adsorption for significantly improved piezocatalytic *** degradation efficiency of bisphenol A(BPA)is 6.3 times higher than SnS_(2) and H2 evolution rate is increased by 3.8 *** with SnS_(2)/SnS prepared by two-step solvothermal method,the degradation efficiency of BPA and H2 evolution activity are increased by 3 and 2 times,*** provides a theoretical guidance for developing various multiphase structural piezocatalyst with strong interface interactions to improve the piezocatalytic efficiency.

Ultrasound-assisted isolation:A new method to isolate stromal vascular fraction

Background:The stromal vascular fraction(SVF),a cluster of stem and progenitor cells isolated from adipose tissue,holds significant promise for application in regenerative ***,the existing methods for SVF isolation are time-consuming and ***,in this study,we explored a new method of SVF extrac-tion-ultrasound-assisted SVF isolation(USASI)-and compared the viability and characteristics of SVF isolated using different ***:SVF extraction methods using different combinations of ultrasound power,ultrasound time,collagenase dosage,and collagenase digestion time were compared with those of the control group(collagenase digestion method).The cell yield and vitality of the SVF were evaluated via cell counting and trypan blue *** cell components and immunophenotypes of freshly isolated SVF were analyzed using flow *** prolifer-ative capacity and differentiation potential of the SVF were also ***:Ultrasonication at 95 W-20 kHz for 30 s followed by digestion with 0.15%collagenase for 30 min was identified as the most suitable parameter for the USASI method in isolating SVF,as recommended based on the evaluation of various tested *** USASI method significantly reduced the collagenase dosage and shortened the digestion *** to the collagenase digestion method,the USASI method had a higher cell yield and cell viability,with no adverse effects on cell components,proliferative capacity,or multipotential differentiation ***:With reduced processing time,lower collagenase dosage,and increased cell yield without impairing the viability and characteristics of SVF,USASI holds the potential to emerge as a time-saving and cost-effective method for future clinical applications.

信息系统通常会借助数据管理系统来进行数据管理,其中SQL凭借良好的易用性和灵活性一直作为数据管理的主流查询语言,用户将编写的SQL语句交由数据管理系统执行后便可得到查询结果.执行模型的高效与否决定了系统能否快速响应用户的查询请求,现有执行模型主要采用解释执行和编译执行2种方式.解释执行具有良好的拓展性、可维护性等因而被大多数系统采用.不同于解释执行,编译执行为原本需要解释执行的查询生成高效的定制化代码来加速查询,带来的显著性能提升吸引了一众数据管理系统开始实现相应技术.然而,如何针对查询生成其对应的定制化代码是一个复杂的过程,在实现时需要考虑诸多方面,甚至在某些情况下,采用编译执行的查询性能可能还不及传统的火山模型.从概念、技术等角度系统地综述了编译执行技术的研究进展.首先,概述了编译执行的基本概念,对相关术语和背景知识进行了介绍;其次,分别从中间代码生成、中间表示、机器码生成与运行3个角度介绍了相关技术;最后,结合当前数据管理系统的研究趋势以及近期研究工作展望了编译执行未来的发展方向.

Ferroptosis, a unique type of non-apoptotic cell death resulting from iron-dependent lipid peroxidation, has a potential physiological function in tumor suppression, but its underlying mechanisms have not been fully elucidated. Here, we report that the long non-coding RNA(lncRNA) LncFASA increases the susceptibility of triple-negative breast cancer(TNBC) to ferroptosis. As a tumor suppressor, LncFASA drives the formation of droplets containing peroxiredoxin1(PRDX1), a member of the peroxidase family, resulting in the accumulation of lipid peroxidation via the SLC7A11-GPX4 axis. Mechanistically, LncFASA directly binds to the Ahpc-TSA domain of PRDX1, inhibiting its peroxidase activity by driving liquid-liquid phase separation, which disrupts intracellular ROS homeostasis. Notably, high LncFASA expression indicates favorable overall survival in individuals with breast cancer, and LncFASA impairs the growth of breast xenograft tumors by modulating ferroptosis. Together, our findings illustrate the crucial role of this lncRNA in ferroptosis-mediated cancer development and provide new insights into therapeutic strategies for breast cancer.

Spin order and dynamics in the topological rare-earth germanide semimetals

The REAl（Si,Ge）(RE = rare earth) family, known to break both the inversion-and time-reversal symmetries, represents one of the most suitable platforms for investigating the interplay between correlated-electron phenomena and topologically nontrivial ***, we report on systematic magnetic, transport, and muon-spin rotation and relaxation（μSR） measurements on（Nd,Sm）AlGe single crystals, which exhibit antiferromagnetic（AFM） transitions at TN=6.1 and 5.9 K, respectively. In addition, NdAlGe undergoes also an incommensurate-to-commensurate ferrimagnetic transition at 4.5 K. Weak transverse-field μSR measurements confirm the AFM transitions, featuring a ～90% magnetic volume fraction. Zero-field（ZF） μSR measurements reveal a more disordered internal field distribution in NdAlGe than in SmAlGe, reflected in a larger transverse muon-spin relaxation rate λTat T■TN. This may be due to the complex magnetic structure of NdAlGe, which undergoes a series of metamagnetic transitions in an external magnetic field, while SmAlGe shows only a robust AFM order. In NdAlGe, the topological Hall effect（THE） appears between the first and the second metamagnetic transitions for H‖c, while it is absent in SmAlGe. Such THE in NdAlGe is most likely attributed to the field-induced topological spin textures. The longitudinal muon-spin relaxation rate λL, diverges near the AFM order, followed by a clear drop at T < T＿N. In the magnetically ordered state, spin fluctuations are significantly stronger in NdAlGe than in SmAlGe. In general, our longitudinal-field μSR data indicate vigorous spin fluctuations in NdAlGe, thus providing valuable insights into the origin of THE and of the possible topological spin textures in REAl（Si,Ge） Weyl semimetals.

作为一种有效解决传统全波形反演(FWI)中周波跳跃问题的方法,基于2-Wasserstein距离的全波形反演(OT-FWI)通常需要采用适当的数据规则化方法。为此,将Sigmoid函数作为数据规则化方法引入OT-FWI,形成基于Sigmoid的OT-FWI方法。Sigmoid函数可以限制数据的取值范围,将小于0的部分映射到接近0的值。与常用的数据规则化方法如仿射尺度规则化和指数归一化规则化方法相比,Sigmoid函数可以更好地利用地震数据中小于0的相位信息,从而进一步提高反演精度。1D Ricker子波测试结果表明,基于Sigmoid数据规则化的2-Wasserstein全波形反演方法可以有效提高目标函数的凸性,并增加共轭震源中的低频信息。将该方法应用于胜利模型数据和中国东部某勘探区实际地震数据进行测试,结果表明,相对于传统的数据规则化方法,基于Sigmod数据规则化的2-Wasserstein全波形反演方法可以进一步提高模型反演的精度。
