bone-targeted therapies to reduce skeletal morbidity in prostate cancer

bone metastases are the main driver of morbidity and mortality in advanced prostate cancer. Targeting the bone microenvironment, a key player in the pathogenesis of bone metastasis, has become one of the mainstays of therapy in men with advanced prostate cancer. This review will evaluate the data supporting the use of bone-targeted therapy, including （1） bisphosphonates such as zoledronic acid, which directly target osteoclasts, （2） denosumab, a receptor activator of nuclear factor-kappa b （RANK） ligand inhibitor, which targets a key component of bone stromal interaction, and （3） radium-223, an alpha-emitting calcium mimetic, which hones to the metabolically active areas of osteoblastic metastasis and induces double-strand breaks in the DNA. Denosumab has shown enhanced delay in skeletal-related events compared to zoledronic acid in patients with metastatic castration-resistant prostate cancer （mCRPC）. Data are mixed with regard to pain control as a primary measure of efficacy. New data call into question dosing frequency, with quarterly dosing strategy potentially achieving similar effect compared to monthly dosing for zoledronic acid. In the case of radium-223, there are data for both pain palliation and improved overall survival in mCRPC. Further studies are needed to optimize timing and combination strategies for bone-targeted therapies. Ongoing studies will explore the impact of combining bone-targeted therapy with investigational therapeutic agents such as immunotherapy, for advanced prostate cancer. Future studies should strive to develop biomarkers of response, in order to improve efficacy and cost-effectiveness of these agents.

Effect of Dietary Components on the Shear bond Strength of Orthodontics brackets after Thermal Aging

Introduction: The stability of orthodontic brackets throughout orthodontic treatment plays a critical role in the treatment’s effectiveness. The present in vitro study was designed to assess the impact of various dietary components on the performance of orthodontic brackets. Methods: Metal orthodontic brackets were bonded to 66 extracted anterior teeth divided into groups based on the solution type: Milk, Gatorade, Cold Coffee, and a control group using water. Each group consisted of 20 teeth except for the control group, which included six teeth. The bracketed teeth were submerged in their respective solutions for 15 minutes three times daily at different intervals to mimic an in vivo environment and were stored in artificial saliva at room temperature (23?C). The specimens underwent artificial aging through 10,000 cycles of thermocycling (representing one clinical year) between 5?C and 55?C. Shade measurements were taken using a VITA Easy Shade device, capturing the classic shade and L*, a*, and b* values. Delta E values were calculated immediately post-bonding and after 7 days, 1 month, 1, and 2 clinical years. The shear bond strength of each bracket was measured using an ultra-tester machine. Results: After two clinical years, significant differences in ΔE color values were observed across all groups, with the most substantial change noted in teeth immersed in cold coffee. brackets submerged in milk demonstrated lower shear bond strength than other solutions, whereas the control group exhibited the highest shear bond strength (P = 0.01). Conclusion: The study indicates that dietary components significantly influence tooth color stability and the shear bond strength of orthodontic brackets, underscoring the importance of considering these factors in orthodontic treatment planning.

Pigs fed camelina meal increase hepatic gene expression of cytochrome 8b1, aldehyde dehydrogenase, and thiosulfate transferase

Comelina sotivo is an oil seed crop which can be grown on marginal lands. Camelina seed oil is rich in omega-3 fatty acids （〉35%） and y-tocopherol but is also high in erucic acid and glucosinolates. Camelina meal, is the by-product after the oil has been extracted. Camelina meal was fed to 28 d old weaned pigs at 3.7% and 7.4% unt age 56 d. The camelina meal supplements in the soy based diets, improved feed efficiency but also significantly increased the liver weights. Gene expression analyses of the livers, using intra-species microarrays, identified increased expression of phase 1 and phase 2 drug metabolism enzymes. The porcine versions of the enzymes were confirmed by real time PCR. Cytochrome 8b1 （CYPSb1）, aldehyde dehydrogenase 2 （Aldh2）, and thiosulfate transferase （TST） were all significantly stimulated. Collectively, these genes implicate the camelina glucosinolate metabolite, methyl-sulfinyldecyl isothiocyanate, as the main xeniobiotic, causing increased hepatic metabolism and increased liver weight.

A Dominant Allele of Arabidopsis Pectin-binding Wall-Associated Kinase Induces a Stress Response Suppressed by MPK6 but Not MPK3 Mutations

The plant cell wall is composed of a matrix of cellulose fibers, flexible pectin polymers, and an array of as- sorted carbohydrates and proteins. The receptor-like Wall-Associated Kinases （WAKs） of Arabidopsis bind pectin in the wall, and are necessary both for cell expansion during development and for a response to pathogens and wounding. Mitogen Activated Protein Kinases （MPKs） form a major signaling link between cell surface receptors and both transcrip- tional and enzyme regulation in eukaryotes, and Arabidopsis MPK6 and MPK3 indeed have important roles in develop- ment and the response to stress and pathogens. A dominant allele of WAK2 requires kinase activity and activates a stress response that includes an increased ROS accumulation and the up-regulation of numerous genes involved in pathogen resistance, wounding, and cell wall biogenesis. This dominant allele requires a functional pectin binding and kinase domain, indicating that it is engaged in a WAK signaling pathway. A null mutant of the major plasma membrane ROS-producing enzyme complex, rbohd/f does not suppress the WAK2cTAP-induced phenotype. A mpk6, but not a mpk3, null allele is able to suppress the effects of this dominant WAK2 mutation, thus distinguishing MPK3 and MPK6, whose activity previously was thought to be redundant. Pectin activation of gene expression is abated in a wak2-null, but is tempered by the WAK-dominant allele that induces elevated basal stress-related transcript levels. The results suggest a mechanism in which changes to the cell wall can lead to a large change in cellular responses and help to explain how pathogens and wounding can have general effects on growth.

Introduction:biological soil crusts(bSCs)can dominate surface cover in dry lands worldwide,playing an integral role in arid land biogeochemistry,particularly in N fertilization through fixation and *** is a characteristic and universal N transformation in bSCs that becomes important for the export of N beyond the microscopic bounds of the crust *** contribution of ammonia-oxidizing bacteria(AOb)in bSCs has been shown,but the role and extent of the recently discovered ammonia-oxidizing archaea(AOA)have ***:We sampled various types of crusts in four desert regions across the western United States and characterized the composition and size of ammonia-oxidizing communities using clone libraries and quantitative PCR targeting the amoA gene,which codes for the ammonia monooxygenase enzyme,universally present in ammonia-oxidizing ***:All archaeal amoA sequences retrieved from bSCs belonged to the Thaumarchaeota(Nitrososphaera associated Group I.1b).Sequences from the Sonoran Desert,Colorado Plateau,and Great basin were indistinguishable from each other but distinct from those of the Chihuahuan *** on amoA gene abundances,archaeal and bacterial ammonia oxidizers were ubiquitous in our survey,but the ratios of archaeal to bacterial ammonia oxidizers shifted from bacterially dominated in northern,cooler deserts to archaeally dominated in southern,warmer ***:Archaea are shown to be potentially important biogeochemical agents of biological soil crust N *** associated with different types of bSCs and biogeographical factors reveal a niche differentiation between AOA and AOb,possibly driven by temperature.

High stability supercontinuum generation in lead silicate SF57 photonic crystal fibers

We report supercontinuum （SC） generation in a lead silicate SF57 photonic crystal fiber by using a 1550 nm pump source. The effective nonlinear coefficient of the SF57 fiber is simulated to be 111.5 W-1 .km- 1 at 1550 nm. The fiber also shows ultraflat dispersion from 1700 nm to 2100 nm. Our results reveal that with an increase of the average power of the incident pulse from 10 mW to 90 mW, the SC of the SF57 photonic crystal fiber is generated from 1300 nm to 1900 nm with high stability and without significant change in spectral broadening.

Sleep Duration and Its Links to Psychological Distress, Health Status, Physical Activity and body Mass Index among a Large Representative General Population Sample

This study aimed to investigate associations between: (a) psychological distress, self-perceived health status and sleep duration among a large representative general population sample;and (b) patterns of sleep duration, physical activity and body Mass Index among a subgroup of participants who self-reported as being in good health with low psychological distress. Data collected from six waves of the Nation Health Interview Surveys (NHISs) was employed. The results indicated that both psychological distress and self-perceived health status were strong predictors of sleep duration. Participants with high serious psychological distress scores reported sleeping 7 - 8 hours less often than those in low or moderate psychological distress and were also most likely to sleep for less than 6 hours or 9 or more hours. Similar patterns were observed for sleep duration by self-reported health status. Subgroup analysis including only participants in self-reported excellent or very good physical health with low mental distress scores showed that participants who engaged in higher frequencies of vigorous and strengthening exercises were more likely to sleep less than six hours, and participants with a bMI of 25 or higher were also more likely to sleep less than six hours.

A meta-analysis of 1-year outcomes of transcatheter versus surgical aortic valve replacement in low-risk patients with severe aortic stenosis

background Transcatheter aortic valve replacement(TAVR)for the treatment symptomatic severe aortic stenosis(AS)is indicated in patients with intermediate or higher surgical *** trials showed TAVR,and surgical aortic valve replacement(SAVR)perform similarly at 1-year for the composite outcomes of mortality,stroke and *** performed a comprehensive meta-analysis to compare individual outcomes at 1-year for TAVR compared to SAVR in low-risk *** Pub Med,Embase,and Cochrane central were searched for all the randomized controlled trials(RCTs)that reported 1-year comparative outcomes of TAVR and surgical aortic valve replacement(SAVR).Our conclusions are based upon the random-effects model using Der Simonian-Laird *** Data from 4 trials and 2887 randomized patients showed that TAVR had lower rates of all-cause mortality,cardiovascular mortality,and atrial fibrillation compared to SAVR at 1-year follow-up(Pbility of clinical benefits in TAVR over SAVR in low-risk patients.
