Do tensile and shear forces exerted on cells influence mechanotransduction through stored energy considerations?

All tissues in the body are subjected externally to gravity and internally by collagenﬁbril and cellular retractive forces that create stress and energy equilibrium required for homeostasis.Mechanotransduction involves mechanical work(force through a distance)and energy storage as kinetic and potential energy.This leads to changes in cell mitosis or apoptosis and the synthesis or loss of tissue components.It involves the application of energy directly to cells through integrin-mediated processes,cell-cell connections,stretching of the cell cytoplasm,and activation of the cell nucleus via yes-associated protein(YAP)and transcriptional coactivator with PDZ-motif(TAZ).These processes involve numerous complexes,intermediate molecules,and multiple pathways.Several pathways have been identiﬁed from research studies on vertebrate cell culture and from studies in invertebrates.These pathways involve mechanosensors and other molecules that activate the pathways.This review discusses the mitogen-activated protein kinase(MAPk)family,Hippo,Hedgehog,and Wingless-related integration site(WNT)/βcatenin signaling pathways.The mediators covered includeβcatenin,ion channels,growth factors,hormone receptors,members of the Ras superfamily,and components of the linker of nucleoskeleton and cytoskeleton(LINC)complex.However,the interrelationship among the different pathways remains to be clariﬁed.Integrin-mediated mechanotransduction involves direct tensile loading and energy applied to the cell membrane via collagenﬁbril stretching.This energy is transferred between cells by stretching the cell-cell connections involving cadherins and the WNT/βcatenin pathway.These alterations induce changes in intracellular events in the cytoskeleton and nuclear skeleton caused by the release of YAP and TAZ.These coactivators then penetrate through the nuclear pores and inﬂuence nuclear cell function.Alteration in the balance of forces and energy applied to cells and tissues is hypothesized to shift the cell-extracellular matrix mechanical equilibri

Whispered Tuning: Data Privacy Preservation in Fine-Tuning LLMs through Differential Privacy

The proliferation of Large Language Models (LLMs) across various sectors underscored the urgency of addressing potential privacy breaches. Vulnerabilities, such as prompt injection attacks and other adversarial tactics, could make these models inadvertently disclose their training data. Such disclosures could compromise personal identifiable information, posing significant privacy risks. In this paper, we proposed a novel multi-faceted approach called Whispered Tuning to address privacy leaks in large language models (LLMs). We integrated a PII redaction model, differential privacy techniques, and an output filter into the LLM fine-tuning process to enhance confidentiality. Additionally, we introduced novel ideas like the Epsilon Dial for adjustable privacy budgeting for differentiated Training Phases per data handler role. Through empirical validation, including attacks on non-private models, we demonstrated the robustness of our proposed solution SecureNLP in safeguarding privacy without compromising utility. This pioneering methodology significantly fortified LLMs against privacy infringements, enabling responsible adoption across sectors.

Photocatalytic Degradation of 4-chlorophenol by CuMoO<sub>4</sub>-doped TiO<sub>2</sub>Nanoparticles Synthesized by Chemical Route

The photocatalytic degradation of 4-chlorophenol (4-CP) in aqueous solution was studied using Cu-MoO4- doped TiO2 nanoparticles under Visible light radiation. The photocatalysts were synthesized by chemical route from TiO2 with different concentration of CuMoO4 (CuxMoxTi1-xO6;where, x values ranged from 0.05 to 0.5). The prepared nanoparticles are characterized by XRD, BET surface area, TEM, UV-vis diffuse reflectance spectra, Raman spectroscopy, XPS and EDAX spectroscopy were used to investigate the nanoparticles structure, size distribution, and qualitative elemental analysis of the composition. The CuxMoxTi1-xO6 (x = 0.05) showed high activity for degradation of 4-CP under visible light. The surface area of the catalyst was found to be 101 m2/g. The photodegradation process was optimized by using CuxMoxTi1-xO6 (x = 0.05) catalyst at a concentration level of 1 g/l. A maximum photocatalytic efficiency of 96.9% was reached at pH = 9 after irradiation for 3 hours. Parameters affecting the photocatalytic process such as catalyst loading, concentration of the catalyst and the dopant concentration, solution pH, and concentration of 4-CP have been investigated.

Evaluating Privacy Leakage and Memorization Attacks on Large Language Models (LLMs) in Generative AI Applications

The recent interest in the deployment of Generative AI applications that use large language models (LLMs) has brought to the forefront significant privacy concerns, notably the leakage of Personally Identifiable Information (PII) and other confidential or protected information that may have been memorized during training, specifically during a fine-tuning or customization process. We describe different black-box attacks from potential adversaries and study their impact on the amount and type of information that may be recovered from commonly used and deployed LLMs. Our research investigates the relationship between PII leakage, memorization, and factors such as model size, architecture, and the nature of attacks employed. The study utilizes two broad categories of attacks: PII leakage-focused attacks (auto-completion and extraction attacks) and memorization-focused attacks (various membership inference attacks). The findings from these investigations are quantified using an array of evaluative metrics, providing a detailed understanding of LLM vulnerabilities and the effectiveness of different attacks.

Design and Implementation of Hand Gesture Detection System Using HM Model for Sign Language Recognition Development

Gesture detection is the primary and most significant step for sign language detection and sign language is the communication medium for people with speaking and hearing disabilities. This paper presents a novel method for dynamic hand gesture detection using Hidden Markov Models (HMMs) where we detect different English alphabet letters by tracing hand movements. The process involves skin color-based segmentation for hand isolation in video frames, followed by morphological operations to enhance image trajectories. Our system employs hand tracking and trajectory smoothing techniques, such as the kalman filter, to monitor hand movements and refine gesture paths. Quantized sequences are then analyzed using the Baum-Welch Re-estimation Algorithm, an HMM-based approach. A maximum likelihood classifier is used to identify the most probable letter from the test sequences. Our method demonstrates significant improvements over traditional recognition techniques in real-time, automatic hand gesture recognition, particularly in its ability to distinguish complex gestures. The experimental results confirm the effectiveness of our approach in enhancing gesture-based sign language detection to alleviate the barrier between the deaf and hard-of-hearing community and general people.

Incidence and risk factors for vitreous loss in residents performing manual small-incision cataract surgery

AIM: To assess the incidence of vitreous loss and associated risk factors in residents performing manual small-incision cataract surgery(MSICS). METHODS: The present retrospective record review study was performed on 490 patients who underwent MSICS performed between November 2018 and December 2019 by 7 third-year postgraduate residents. The study group comprised of patients having intraoperative vitreous prolapse. All the surgeries were performed under supervision of a trained assistant. RESULTS: The mean age of the participants at the time of surgery was 68.42±2.05 y. Of the 490 patients, 250 patients were male, and 240 patients were female(P=0.23). A total of 215(43.9%) eyes had mature white cataract, 185(37.8%) eyes had brown cataract, and 90(18.3%) eyes had immature senile cataract. The incidence of intraoperative vitreous loss among residents was 2%(10/490). Vitreous loss occurred during hydrodissection [1/10(10%)], nucleus delivery [3/10(30%)], irrigation and aspiration [5/10(50%)], and intraocular lens insertion [1/10(10%)]. Multivariate stepwise Logistic regression analysis confirmed immature senile cataract [odds ratio(OR)=3.99;P=0.02], irrigation and aspiration of cortical material(OR=3.07;P=0.03), and anterior capsular extension(OR=3.22, P=0.03) as independent risk factors for vitreous loss. CONCLUSION: Immature senile cataract, irrigation and aspiration of cortical material, and anterior capsular extension are independent risk factors for vitreous loss. Our findings may serve as a guide for future trainers or residents learning MSICS.

A Chicken's Egg as a Reaction Vessel to Explore Biomineralization

Natural composites, formed through biomineralization, have highly ordered structures which have been aptly explored for functional applications. Though the role of organic phases has been well understood in biomineralization, not enough attention has been paid to the role of bio-membranes which are often found encapsulating the chamber in which mineralization occurs. We have used the natural protein and semi-permeable membrane of chicken eggs to grow different materials such as ceramics, semi-metals and metals to understand the role of bio-membranes in biomineralization. We here report the successful biomimetic synthesis of calcite, cadmium sulphide, and silver having homogeneous morphologies. We have found that the membrane operates like a tuned gateway, playing a significant role in controlling the morphology of the inorganic crystals formed during biomineralization.

Power distribution systems are profoundly inclined to disturbances like untimely switching of breakers & relays, sympathetic tripping, and uncertainties regarding fault location. Thus, system stability and reliability are greatly affected. In this way, situational awareness and system integrity are the crucial factors in developing power system security, as it empowers successful decision making & timely reaction by the operators to any disturbance and also maintaining continuity of power supply. This paper focuses on the enhancement of situational awareness by fault location through fault passage indicators (FPI) to improve nominal impedance-based methods in distribution networks. Also, the proposed method is validated by comparing it with Intelligent Electronic Device (IED) based fault location method. Further, simultaneous reconfiguration of the system is incorporated to maintain the continuity of supply. The analysis has been tested on IEEE 33 bus distribution system.

Optimal reactive power dispatch(ORPD)is a complex and non-linear problem,and is one of the sub-problems of optimal power flow(OPF)in a power system.ORPD is formulated as a single-objective problem to minimize the active power loss in a transmission system.In this work,power from distributed generation(DG)is integrated into a conventional power system and the ORPD problem is solved to minimize transmission line power loss.It proves that the application of DG not only contributes to power loss minimization and improvement of system stability but also reduces energy consumption from the conventional sources.A recently proposed meta-heuristic algorithm known as the JAYA algorithm is applied to the standard IEEE 14,30,57 and 118 bus systems to solve the newly developed ORPD problem with the incorporation of DG.The simulation results prove the superiority of the JAYA algorithm over others.The respective optimal values of DG power that should be injected into the four IEEE test systems to obtain the minimum transmission line power losses are also provided.

Factors Affecting Vulnerability of Ready-Made Garment Factory Buildings in Bangladesh: An Assessment Under Vertical and Earthquake Loads

The importance of workplace safety in the ready-made garment(RMG) industry in Bangladesh came to the forefront after a series of disastrous events in recent years. In order to reduce the loss of lives and to ensure sustainable development, an in-depth understanding of the determining factors governing structural vulnerability in the RMG industry is needed. This research explores the key factors influencing the vulnerability of factory buildings under both vertical and earthquake loads. For this purpose,an ordered probit model was applied to 3746 RMG factory buildings to determine the key factors that influenced their vertical load vulnerability. A smaller subset of the original sample, 478 buildings, was examined by the same modeling method in greater detail to assess the key factors that influenced their earthquake load vulnerability. This research reveals that column capacity, structural system,and construction materials are the most influential factors for both types of vulnerabilities. Among other factors, soil liquefaction and irregular internal frame affect earthquake load vulnerability significantly. These findings are expected to enable factory owners and designers to better weigh the appropriate vulnerability factors in order to make informed decision that increase workplace safety. Theresearch findings will also help the designated authorities to conduct successful inspections of factory buildings and take actions that reduce vulnerability to both vertical and earthquake loads.
