Gut barrier failure biomarkers are associated with poor disease outcome in patients with primary sclerosing cholangitis

To assess the prevalence of a panel of serologic markers that reflect gut barrier dysfunction in a mixed cohort of pediatric and adult primary sclerosing cholangitis (PSC) patients. METHODSSera of 67 PSC patients [median age (range): 32 (5-79) years, concomitant IBD: 67% and cirrhosis: 20%] were assayed for the presence of antibodies against to F-actin (AAA IgA/IgG) and gliadin (AGA IgA/IgG)] and for serum level of intestinal fatty acid-binding protein (I-FABP) by ELISA. Markers of lipopolysaccharide (LPS) exposure [LPS binding protein (LBP)] and various anti-microbial antibodies [anti-OMP Plus IgA and endotoxin core IgA antibody (EndoCAb)] were also determined. Poor disease outcome was defined as orthotopic liver transplantation and/or liver-related death during the follow-up [median: 99 (14-106) mo]. One hundred and fifty-three healthy subjects (HCONT) and 172 ulcerative colitis (UC) patients were the controls. RESULTSA total of 28.4%, 28.0%, 9% and 20.9% of PSC patients were positive for AAA IgA, AAA IgG, AGA IgA and AGA IgG, respectively. Frequencies of AAA IgA and AAA IgG (P AAA IgA posvsneg: 365 vs 166 pg/mL, P = 0.011), but not with serum LBP level. Modification of tooth development by heat shock protein 60

several heat shock proteins have been investigated in relation to tooth development, no available information is available about the spatial and temporal expression pattern of heat shock protein 60 （Hsp 60）. To characterize Hsp 60 expression in the structures of the developing tooth germ, we used Western blotting, immunohistochemistry and in situ hybridization. Hsp 60 was present in high amounts in the inner and outer enamel epithelia, enamel knot （EK） and stratum intermedium （SI）. Hsp 60 also appeared in odontoblasts beginning in the bell stage. To obtain data on the possible effect of Hsp 60 on isolated lower incisors from mice, we performed in vitro culturing. To investigate the effect of exogenous Hsp 60 on the cell cycle during culturing, we used the 5-bromo-2- deoxyuridine （BrdU） incorporation test on dental cells. Exogenously administered Hsp 60 caused bluntness at the apical part of the 16.5-day-old tooth germs, but it did not influence the proliferation rate of dental cells. We identified the expression of Hsp 60 in the developing tooth germ, which was present in high concentrations in the inner and outer enamel epithelia, EK, SI and odontoblasts. High concentration of exogenous Hsp 60 can cause abnormal morphology of the tooth germ, but it did not influence the proliferation rate of the dental cells. Our results suggest that increased levels of Hsp 60 may cause abnormalities in the morphological development of the tooth germ and support the data on the significance of Hsp during the developmental processes.

Effect of hydrocele on appendix testis in children

The purpose of this study was to investigate the effects of an elevated hydrostatic pressure of hydrocele on the structural integrity and steroid receptor expression pattern of the appendix testis in children. Twenty-six testicular appendages were obtained from boys （aged between 13 and 79 months, mean 40 months） who underwent surgical exploration because of hydrocele or congenital inguinal hernia. The tissue sections of testicular appendages were stained with hematoxylin-eosin. Immunohistochemistry and immunofluorescence laser microscopy were performed using monoclonal mouse anti-human receptors against androgen and estrogen receptors. Patients were divided into three groups： group A （n = 8） represented patients with groin hernia without hydrocele, who served as control group; group B （n = 7） represented patients with communicating hydrocele; and group C （n = 11） represented patients with noncommunicating hydrocele. The tissue sections of appendix testis expressed both androgen and estrogen receptors in all patients in groups A and B, and epithelial destruction was not present. The presence of androgen receptor （two of 11, P 〈 0.001） and estrogen receptor （four of 11, P = 0.006） was lower and the number of appendix testes with epithelial destruction was higher （eight of 11, P-- 0.001） in group C. We demonstrated that groin hernia and communicating hydrocele did not influence the receptor expression pattern and the anatomic structure of testicular appendages, whereas noncommunicating hydrocele caused damage as indicated by the absence of steroid receptors and destruction of the epithelial surface. A better understanding of the physiological role of testicular appendages may change the indications of surgical treatment in patients with noncommunicating hydrocele.

树木是虚拟森林景观的重要要素,也是自然景观可视化中较难建模与表达的要素之一。传统的树木建模方法无法用简单规律描述形态复杂的树木,且生成的树木往往也忽略生物学意义。因此限制了其实用性。针对这些问题提出了一种能较为全面反映生物学形态结构特征的L系统树木建模方法,并以热带水果种类之一的芒果树为模型对该方法进行了初步的设计与实现,结果表明这种方法能生成植物学形态特征的、具有不同树龄的三维逼真芒果树。

流芳传世的建筑作品中,不少都是教堂建筑,世人用憧憬、尊敬的眼神望向为神明构建的房屋,也望向设计的精华。除去那些体型恢弘、结构复杂的地标性建筑,最接近我们身边的祷告处可能就是那些朴素简单的教会,体量不大却功能齐全。来自SAGRA Architects的建筑师们在匈牙利索尔诺克为当地的基督教会修建了一座教堂。这个建筑复合体主要包含了三部分:一座教堂、一座带有牧师工作室的教会场所和一座钟塔。至现在,教会场所和钟塔已经建设完毕,教堂部分因为一些财政问题还待修建。

目的　研究瘦素（ｌｅｐｔｉｎ）对罗猴下丘脑促性腺激素释放激素脉冲发生器（ＧｎＲＨＰＧ）的多单位电脉冲频率骤然增加（ＭＵＡｖｏｌｌｅｙ）的影响。方法　在基础及受吗啡抑制状态时脑室内输注瘦素，记录植入下丘脑内侧基底部电极的电脉冲释放，计算两次ＭＵＡｖｏｌｌｅｙ之间的间期（ＭＶＩ）。结果瘦素可显著减轻吗啡使ＭＶＩ延长的抑制效应，ＭＶＩ在注射吗啡后为对照的２９０％，如脑室内输注瘦素则可使其减少至１６９％（瘦素１．５μｇ／ｋｇ）和１５１％（瘦素７．５μｇ／ｋｇ），Ｐ＜０．０１。结论　瘦素可部分保护ＧｎＲＨＰＧ少受吗啡的抑制。

AIM:To assess the combined effect of disease phenotype, smoking and medical therapy [steroid, azathioprine(AZA), AZA/biological therapy] on the probability of disease behavior change in a Caucasian cohort of patients with Crohn's disease(CD).METHODS:Three hundred and forty well-characterized, unrelated, consecutive CD patients were analyzed(M/F:155/185, duration:9.4 ± 7.5 years) with a complete clinical *** records including disease phenotype according to the Montreal classification, extraintestinal manifestations, use of medications and surgical events were analyzed *** were interviewed on their smoking habits at the time of diagnosis and during the regular follow-up ***:A change in disease behavior was observed in 30.8% of patients with an initially non-stricturing, non-penetrating disease behavior after a mean diseaseduration of 9.0 ± 7.2 *** a logistic regression analysis corrected for disease duration, perianal disease, smoking, steroid use, early AZA or AZA/ biological therapy use were independent predictors of disease behavior *** a subsequent Kaplan-Meier survival analysis and a proportional Cox regression analysis, disease location(P = 0.001), presence of perianal disease(P < 0.001), prior steroid use(P = 0.006), early AZA(P = 0.005) or AZA/biological therapy(P = 0.002), or smoking(P = 0.032) were independent predictors of disease behavior ***:Our data suggest that perianal disease, small bowel disease, smoking, prior steroid use, early AZA or AZA/biological therapy are all predictors of disease behavior change in CD patients.
