Celiac disease in patients with type 1 diabetes:a condition with distinct changes in intestinal immunity?

Two common chronic childhood diseases-celiac disease(CD)and type 1 diabetes(T1D)-result from complex pathological mechanisms where genetic susceptibility,environmental exposure,alterations in intestinal permeability and immune responses play central *** this study,we investigated whether these characteristics were universal for CD independently of T1D *** this purpose,we studied 36 children with normal small-bowel mucosa and 26 children with active CD,including 12 patients with *** samples from the small-bowel mucosa,we detected the lowest expression of tight junction protein 1(TJP1)mRNA in CD patients with T1D,indicating an increase in intestinal ***,these samples displayed the highest expression of forkhead box P3(FoxP3)mRNA,a marker for regulatory T cells,as compared with other patient *** the same time,serum levels of IgA antibodies specific for the CD-related antigens deamidated gliadin and tissue transglutaminase(tTG)were the highest in CD patients with *** contrast,no significant differences were found in IgA or IgG antibodies specific for bovine beta-lactoglobulin or Bifidobacterium adolescentis DSM 20083-derived *** were also no differences in the transamidating activity of serum autoantibodies between patients and control *** results show that patients with T1D and newly detected CD exhibit severely altered intestinal permeability,strong local immune activation and increased immunoregulatory mechanisms in the small *** study is required to determine whether these extreme changes in this CD subgroup are due to some specific environmental factors(virus infections),unknown genetic effects or autoimmune reactions to antigenic targets in intracellular tight junctions.

Introduction: Degenerative or traumatic causes are most common in generating sciatica syndrome, which is normally treated with well-known physical therapy methods. A relatively new way of treating sciatica problems is so-called functional magnetic stimulation (FMS), whose principle is based on electromagnetic field inducing electrical field inside the body. Electrical field triggers action potential of nerve cells and that way stimulates peripheral motor nerve system. Aim: Aim of this study is to measure and estimate the effectiveness of implementing therapy with functional magnetic stimulation in regular physical treatment of sciatica syndrome. Materials and Methods: 28 male patients aged between 30 and 55 with back problem were recruited on an outpatient basis. FMS therapy was performed with TESLA Stym? device (Iskra Medical d.o.o., Slovenia) treating lumbosacral region equally on both sides of the spine. Physical examination was performed to evaluate tree parameters: the mobility of the lumbar spine in flexion and extension, together with the straight leg raise test (Lasegue sign). We estimated patients’ progress, comparing angle values of mobility from the first examination day with other examination days. Results: In FMS treated group of patients, lumbosacral flexion, extension and Lasegue test angle were significantly higher compared to day 0 on the first physical examination day (day 3) (p < 0.05). In control group such increase of a measured angle was not noticed until a second physical examination day (day 5) or a third physical examination day (day 8) (p < 0.05). Discussion: Results in this study showed that applying FMS therapy along with other standard physical therapy methods rapidly increased effectiveness of the treatment of sciatica syndrome (lat. ischialgia). It suggests that functional magnetic therapy could be suggested as a regular physical therapy method in treating this kind of pain syndromes.

The epithelial to mesenchymal transition(EMT)is a cellular program that drives de-differentiation of cells in both physiological and pathological *** of the characteristics of cells describing an EMT is the(re)acquisition of a motility capacity that allows them to migrate through the original tissue as well as to other sites in the *** molecular mechanisms that control the EMT are rapidly emerging and here we add to the idea that the adaptation required for cells to commit to the EMT includes adjustments of the translation machinery and metabolic pathways to cope with a high demand of extracellular components.

The Pt（IV） complex cis,cis,trans-[Pt（PMSA）2Cl2（OH）2]·2H2O（1）, where PMSA = N-3-pyridinylmethanesulfonamide, has been synthesized and characterized by elemental analysis, molar electric conductivity and IR spectrum. X-ray crystallography revealed that the title compound crystallizes in the monoclinic C2/c space group with unit cell dimensions a = 16.5424（7）, b = 8.6973（3）, c = 16.6079（6） A, β = 117.185（5）° and Z = 4. Pt（IV） has an octahedral coordination geometry and the PMSA ligands are coordinated via the pyridine N atom. They are in a transoid orientation between each other and are inclined in the same direction with respect to the Pt-C1-Cli-N-Ni plane at an angle of 64.6（1）° （symmetry operation：（i） 1- x, y, 3/2-z）. The structure is stabilized by a system of hydrogen bonds involving the complex and water molecules.

AIM:To evaluate weight loss and surgical outcomes of Roux-en-Y gastric bypass(RYGB)and laparoscopic adjustable gastric band(LAGB).METHODS:Data relating to changes in body mass index(BMI)and procedural complications after RYGB(1995-2009;n=609;116M:493F;42.4±0.4 years)or LAGB(2004-2009;n=686;131M:555F;37.2±0.4years)were extracted from prospective ***:Pre-operative BMI was higher in RYGB than LAGB patients(46.8±7.1 kg/m2vs 40.4±4.2 kg/m2,P<001);more patients with BMI<35 kg/m2underwent LAGB than RYGB(17.1%vs 4.1%,P<0.0001).BMI decrease was greater after *** were direct relationships between weight loss and pre-operative BMI(P<0.001).Although there was no difference in weight loss between genders during the first 3-year post-surgery,male LAGB patients had greater BMI reduction than females(-8.2±4.3 kg/m2vs-3.9±1.9kg/m2,P=0.02).Peri-operative complications occurred more frequently following RYGB than LAGB(8.0%vs0.5%,P<0.001);majority related to wound *** had more long-term complications requiring corrective procedures than RYGB(8.9%vs 2.1%,P<0.001).Conversion to RYGB resulted in greater BMI reduction(-9.5±3.8 kg/m2)compared to removal and replacement of the band(-6.0±3.0 kg/m2).Twelve months post-surgery,fasting glucose,total cholesterol and low density lipoprotein levels were significantly lower with the magnitude of reduction greater in RYGB ***:RYGB produces substantially greater weight loss than *** peri-operative complications are greater after RYGB,long-term complication rate is higher following LAGB.

The objective of the study was to develop a univariate model for analyzing energy balance data from lactating goats at mid lactation and determine maintenance requirements and partial efficiencies of energy utilization. Energy balance data from eight studies involving lactating Murciano-Granadina goats fed total mixed diets, which accounted for a variation in metabolizable energy (MEI) intake, milk energy output (EI), and tissue energy balance, were used. The database included records obtained by indirect calorimetry. Data were adjusted with a mixed model that included the study as a random effect. Then, two multivariate linear models were obtained: metabolizable and net energy models. The metabolizable model was MEI=β0+β2E1+β3Tg+β4T1+ε and the net energy model was E1=β0+β1MEI+β2Tg+β3T1+ε;where β0, β1, β2 and β3 were the parameters, Tg was tissue energy retention and Tl the milk energy derived from body stores. For a better fitted proposed model, net energy for maintenance (NEm) was 283 kJ/kg of Body Weight 0.75 (BW) per day, and the efficiency of utilization of ME for lactation (k1), body weight gain (kg) and body tissue mobilization for milk production (kt) were 62%, 83% and 78%, respectively. Maintenance requirements and partial efficiencies for milk production and tissue energy mobilization were similar to the values proposed by INRA (2018). The increase in the efficiency of utilizing dietary energy for gain, compared with other feeding systems, was partially attributed to the stage of lactation, due to that goats were feeding at mid lactation.

Natural killer （NK） ceils are innate immune cells with the ability to identify and eliminate transformed cells. However, within tumors, many studies have described NK cells as non-functional. The developmental stage of tumor-associated NK cells and how this may relate to functionality has not been explored. We examined the developmental state of NK cells from polyoma middle T antigen （pyMT） transgenic mouse （MMTV-pMT） breast tumors. In pyMT tumors, NK cells were immature as evidenced by their decreased expression of DX5 and their CD27~~WCD1 lbI^w phenotype. These immature NK cells also had increased expression of NKG2A and expressed low levels of NKp46, perforin, and granzyme B. In contrast, splenic NK cells isolated from the same mice maintained their maturity and their expression of activation markers. To delineate whether the tumor microenvironment directly alters NK cells, we adoptively transferred labeled NK cells and followed their activation status in both the spleen and the tumor. NK cells that arrived at the tumor had half the expression of NKp46 within three days of transfer in comparison to those which arrived at the spleen. In an effort to modify the tumor microenvironment and assess the plasticity of intratumoral NK cells, we treated pyMT tumors with IL-12 and anti-TGF-β. After one week of treatment, the maturity of tumor-associated NK cells was increased; thus, indicating that these cells possess the ability to mature and become activated. A better understanding of how NK cells are modified by the tumor microenvironment will held to develop strategies aimed at bolstering immune resoonses against tumors.
