Secondary metabolites are important for various industrial applications. The production of secondary metabolites is often improved by the activation of substrate supply pathways for biosynthesis. However, many important pathways have remained unclear. In this study, we explored possible pathways related to substrate supply for the biosynthesis of the antifungal agent FR901469 which is a nonribosomal peptide and a fungal secondary metabolite. To clarify the unknown activated pathways, we utilized the Comprehensive Pathway Model (CPM) which was developed in our previous study. We verified that the overexpression of the hypothetical beta-alanine-aminotransferase (BAL-AT), which was included in the explored pathways, improved the FR901469 productivity. The genes encoding the BAL metabolic enzymes are considered to be important for improving the FR901469 productivity.

We observed the concentration and backscattering coefficients of aerosol particles at southwestern coast of Japan in 2011-2015. The constitution of particles in each of eight episodes, i.e. the portions of mineral dust, sea salt, sulfate and soot particles, was obtained with individual particle analysis by using an electron microscope. There was a close correlation(R2=0.76) between the BSCs and the volume concentrations of the particles when the relative humidity(RH) was lower than 0%, regardless of the difference in particle constitutions. In contrast, the BSCs normalized with aerosol concentrations differed largely even at similar concentrations of particles when the RH was larger than 70%. The BSCs of particles dominated by marine origin(sea salt) could be several times larger than those dominated by land origin(dust and soot) at 90% RH, as the latter might be slightly larger than the BSCs under the dry conditions. On the other hand, the difference of volume-size distributions of particles likely also made the BSCs largely different from each other under the humid conditions. These results indicate that the concentration of aerosol particle was the key parameter to determine the BSCs under dry conditions, while the BSCs under humid conditions were closely dependent on the content of deliquescent components and the size distributions of aerosol particles.

Nitride fuels have several advantages including high thermal conductivity and high metal density(like metallic fuels) and high melting point and isotropic crystal structure(like oxide fuels). Since the late 1990 s, the partitioning and transmutation of minor actinides(MA) has been studied to decrease the long-term radio-toxicity of high-level waste and to mitigate the burden of final disposal. Japan Atomic Energy Agency(JAEA) has proposed a dedicated transmutation cycle using an accelerator-driven system(ADS) with nitride fuels containing MA. The nitride fuel cycle we have developed includes a pyrochemical process. Our focus is on the electrolysis of nitride fuels and their refabrication from the recovered actinides; other processes are similar to the technology for metal fuel treatment and have been studied elsewhere. Here, we summarize our activity on the development of the pyrochemical treatment of spent nitride fuels.

The quantitation of serum tocilizumab using liquid chromatography tandem-mass spectrometry(LC-MS/MS)method has not been widely applied in clinical settings because of its time-consuming and costly sample *** present study aimed to develop a validated LC-MS/MS method for detecting serum tocilizumab by utilizing immobilized trypsin without an immunoglobulin G purification step and evaluate its applicability in the treatment of rheumatoid arthritis(RA)patients administered intravenously or subcutaneously with *** tocilizumab-derived signature peptide was deciphered using a nano-LC system coupled to a hybrid quadrupole-orbitrap mass *** serum tocilizumab was rapidly digested by immobilized trypsin for 30 *** chromatographic peak of the signature peptide and that of the internal standard were separated from the serum digests for a total run time of 15 *** calibration curve of serum tocilizumab concentration was linear with a range of 2-200 μg/*** intra-and inter-day accuracy and relative standard deviation(RSD)were 90.7%-109.4%and<10%,*** serum tocilizumab concentrations in the RA patients receiving intravenous and subcutaneous injections were 5.8-28.9 and 2.4-63.5 μg/mL,*** serum tocilizumab concentrations using the current method positively correlated with those using the enzyme-linked immunosorbent assay,although a systematic error was observed between these *** conclusion,a validated LC-MS/MS method with minimal sample pretreatments for monitoring serum tocilizumab concentrations in RA patients was developed.

BACKGROUND Malignant transformation(MT)of mature cystic teratoma(MCT)has a poor prognosis,especially in advanced *** chemoradiotherapy(CCRT)has an inhibitory effect on *** SUMMARY Herein,we present a case in which CCRT had a reduction effect preoperatively.A 73-year-old woman with pyelonephritis was referred to our *** tomography revealed right hydronephrosis and a 6-cm pelvic *** ultrasound-guided fine-needle biopsy(EUS-FNB)revealed squamous cell *** patient was diagnosed with MT of *** to her poor general con-dition and renal malfunction,we selected CCRT,expecting fewer adverse *** CCRT,her performance status improved,and the tumor size was reduced;surgery was *** months postoperatively,the patient developed dis-semination and lymph node *** chemotherapy was *** died 18 months after treatment *** EUS-FNB was useful in the diagnosis of MT of MCT;CCRT suppressed the disea-se and improved quality of life.
