Confirming Stereochemical Structures of Strigolactones Produced by Rice and Tobacco

Major strigolactones （SLs） produced by rice （Oryza sativa L. cv. Nipponbare） and tobacco （Nicotiana tabacum L. cv. Michinoku No. 1） were purified and their stereochemical structures were determined by comparing with optically pure synthetic standards for their NMR and CD data and retention times and mass fragmentations in ESI-LC/MS and GC-MS. SLs purified from root exudates of rice plants were orobanchol, orobanchyl acetate, and ent-2＂-epi-5-deoxystr- igol. In addition to these SLs, 7-oxoorobanchyl acetate and the putative three methoxy-5~deoxystrigol isomers were detected by LC-MS/MS. The production of 7-oxoorobanchyl acetate seemed to occur in the early growth stage, as it was detected only in the root exudates collected during the first week of incubation. The root exudates of tobacco contained at least 11 SLs, including solanacol, solanacyl acetate, orobanchol, ent-2＂-epi-orobanchol, orobanchyl acetate, ent-2＇- epi-orobanchyl acetate, 5-deoxystrigol, ent-2＂-epi-5-deoxystrigol, and three isomers of putative didehydro-orobanchol whose structures remain to be clarified. Furthermore, two sorgolactone isomers but not sorgolactone were detected as minor SLs by LC-MS/MS analysis. It is intriguing to note that rice plants produced only orobanchol-type SLs, derived from ent-2＂-epi-5-deoxystrigol, but both orobanchol-type and strigol-type SLs, derived from 5-deoxystrigol were detected in tobacco plants.

A pair of unidirectional turbines(UT)can operate in oscillatory airflow without additional ***,this arrangement suffers from poor flow rectification.A fluidic diode(FD)offers variable hydrodynamic resistance based on the flow direction,and this can be coupled with UT to improve flow *** this work,a numerical investigation on the effect of FD with UT is presented using the commercial fluid dynamics software ANSYS Fluent 16.1 with k-ωSST turbulence closure *** domains of UT and FD are numerically validated individually with experimental ***,both are coupled to obtain the combined effect,and these results are compared with the analytical *** was observed that coupling FD with UT improved the unit's performance at the lower flow coefficient(<1),but its performance decreased as the flow coefficient *** to the diode's presence,fluid leaving the turbine experiences higher resistance at a higher flow coefficient,which decreases the overall performance of the combined unit.

针对p-Frobenius问题中出现的非对称性问题,利用数值半群理论以及Apery集等工具研究了p-Frobenius问题对应的p-对称数值半群。首先利用数值半群和Apery集等工具对p-Frobenius问题进行预处理,得到了对应数值半群和Apery集的各个变量;并将这些变量与对称数值半群和Frobenius问题进行对比,由此定义了p-对称数值半群,并给出了p-对称性的刻画。在此基础上,首先给出了两大类p-对称数值半群,其次研究了对称数值半群与p-对称数值半群的p化关联性。这两类p-对称数值半群给出了大量具体实例,而p化关联性则可用于处理p-Frobenius问题中出现的非对称问题。

Excited state calculations using variational quantum eigensolver with spin-restricted ansätze and automatically-adjusted constraints

The ground and excited state calculations at key geometries, such as the Frank–Condon (FC) and the conical intersection (CI)geometries, are essential for understanding photophysical properties. To compute these geometries on noisy intermediate-scalequantum devices, we proposed a strategy that combined a chemistry-inspired spin-restricted ansatz and a new excited statecalculation method called the variational quantum eigensolver under automatically-adjusted constraints (VQE/AC). Unlike theconventional excited state calculation method, called the variational quantum deflation, the VQE/AC does not require the pre-determination of constraint weights and has the potential to describe smooth potential energy surfaces. To validate this strategy,we performed the excited state calculations at the FC and CI geometries of ethylene and phenol blue at the complete active spaceself-consistent field (CASSCF) level of theory, and found that the energy errors were at most 2 kcal mol−1 even on the ibm_kawasakidevice.

Saccharomyces cerevisiae in neuroscience:how unicellular organism helps to better understand prion protein?

The baker's yeast Saccharomyces(S.)cerevisiae is a single-celled eukaryotic model organism widely used in research on life *** a unicellular organism,*** has some evident limitations in application to ***,yeast prions are extensively studied and they are known to share some hallmarks with mammalian prion protein or other amyloidogenic proteins found in the pathogenesis of Alzheimer's,Parkinson's,or Huntington's ***,the yeast *** has been widely used for basic research on aggregation properties of proteins in cellulo and on their ***,a yeast-based study revealed that some regions of mammalian prion protein and amyloidβ1–42 are capable of induction and propagation of yeast *** is one of the examples showing that evolutionarily distant organisms share common mechanisms underlying the structural conversion of prion proteins making yeast cells a useful system for studying mammalian prion *** has also been used to design novel screening systems for anti-prion compounds from chemical *** assays are cheap in maintenance and safe for the researcher,making them a very good choice to perform preliminary screening before further characterization in systems engaging mammalian cells infected with *** this review,not only classical red/white colony assay but also yeast-based screening assays developed during last year are *** analysis and research carried out using yeast prions force us to expect that prions are widely present in ***,the last few years brought us several examples indicating that the mammalian prion protein is no more peculiar protein–it seems that a better understanding of prion proteins nature-wide may aid us with the treatment of prion diseases and other amyloid-related medical conditions.

Circulating tumor cells and cell-free tumor DNA analyses in urothelial cancer using the LiquidBiopsy platform

Background:Emerging data suggested that liquid biopsy such as detection of circulating tumor cells(CTCs)and cell-free tumor DNA analysis augments the management of patients with urothelial cancer(UC).We presented our pilot experience of liquid biopsy using the Ion Torrent platform to detect CTCs and genomic alterations in UC.Materials and methods:Blood or urine samples from 16 patients were subjected to CTC and plasma/urine cell-free tumor DNA isolation for next generation sequencing(NGS)using the Ion S5 system to detect mutations among 50 oncogenes on the Ion AmpliSeq Cancer Hotspot Panel.Results:The Ion Torrent platform detected a higher number of CTCs than those in previous studies using the CellSearchTM ***,mutations were detected in 13/16(81.3％)patients with a median number of 18(range 12-25).NGS isolated 17 hotspot mutations from 11 genes and 41 novel genomic alterations from 24 genes,some of which are supposed to be clinically actionable.Conclusions:The Ion Torrent platform efficiently detected CTCs compared with previous *** with the present system also allowed for detection of gene alterations which are likely to be therapeutic targets and provided an attractive tool to guide personalized therapy for patients with advanced UC.

Association of antithrombotic therapy with postoperative rebleeding in patients with cerebral amyloid angiopathy

Background Cerebral amyloid angiopathy is a common cause of subcortical hemorrhage in older *** open hematoma removal may be performed for severe subcortical hemorrhage,its safety in patients with cerebral amyloid angiopathy has not been established,and postoperative rebleeding may ***,this study aimed to investigate factors associated with postoperative *** Out of 145 consecutive patients who had undergone craniotomy for surgical removal of subcortical intracerebral hemorrhage between April 2010 and August 2019 at a single institution in Japan,we examined 109 patients with subcortical hemorrhage who met the inclusion *** excluding 30 patients whose tissue samples were unsuitable for the study,the final study cohort comprised 79 *** Of the 79 patients,50(63%)were diagnosed with cerebral amyloid angiopathy(cerebral amyloid angiopathy group)and 29(37%)were not diagnosed with noncerebral amyloid angiopathy(noncerebral amyloid angiopathy group).Postoperative rebleeding occurred in 12 patients(24%)in the cerebral amyloid angiopathy group and in 2 patients(7%)in the noncerebral amyloid angiopathy *** prothrombin time-international normalized ratio and intraoperative bleeding volume were significantly associated with postoperative rebleeding in the cerebral amyloid angiopathy group(odds ratio=42.4,95%confidence interval=1.14-1578;p=0.042 and odds ratio=1.005,95%confidence interval=1.001-1.008;p=0.007,respectively).Conclusions Patients with cerebral amyloid angiopathy-related cerebral hemorrhage who are receiving antithrombotic therapy,particularly warfarin therapy,are at a high risk of postoperative *** registration Registry and Registration Number of the study:19-220,2019/12/23,retrospectively registered.
