Partial Purification and Characterization of the Rat Parotid Gland Protein Kinase Catalyzing Phosphorylation of Matured Destrin at Ser-2

Destrin, also called actin-depolymerizing factor (ADF), exists in resting parotid tissue as phosphorylated (inactive) and dephosphorylated (active) forms, and β-adrenergic stimulation of this tissue induces dephosphorylation of destrin. It is suggested that destrin dephosphorylation is involved in cortical F-actin disruption observed in parallel with β-agonist-induced amylase secretion. At present, the phosphorylation/dephosphorylation mechanism of destrin in parotid tissue is not known. We previously detected, in a crude rat parotid extract, a constitutively active protein kinase catalyzing phosphorylation of destrin;however, its identification has been hampered by difficulty in its enrichment. The purpose of this study was to explore a simple purification method(s) for this enzyme. To this end, we first developed a high-throughput dot-blot assay for the kinase with an anti-phosphodestrin antibody and then studied its purification by column chromatography on several media. We found that the kinase could be partially purified by sequential chromatography on DEAE-cellulose, phenyl-Sepharose, and hydroxyapatite columns. In each chromatography, however, the kinase could be eluted, at the cost of resolution, only by sharp increases in the elution power of the eluent;gradual increases in the elution power resulted in unacceptably poor recovery. We confirmed that enzymatic properties of the kinase were not basically altered during the purification. Further purification of the kinase was achieved by native polyacrylamide gel electrophoresis (PAGE), which resolved the kinase activity into two bands and separated the activity from most proteins (the kinase activity after PAGE was detected with destrin-coated polyvinylidene difluoride membranes and the anti-phosphodestrin antibody). Magnetic properties of soft layer/FePt-MgO exchange coupled composite perpendicular recording media

The magnetic properties of exchange coupled composite （ECC） media that are composed of perpendicular magnetic recording media FePt MgO and two kinds of soft layers have been studied by using an x-ray diffractometer, a polar Kerr magneto-optical system （PMOKE） and a vibrating sample magnetometer （VSM）. The results show that ECC media can reduce the coercivities of perpendicular magnetic recording media FePt-MgO. The ECC media with granular-type soft layers have weaker exchange couplings between magnetic grains and the magnetization process, for ECC media of this kind mainly follow the Stoner Wohlfarth model.

A Straight-bladed Vertical Axis Wind Turbine with a Directed Guide Vane Row-Effect of Guide Vane Geometry on the Performance-

The objective of this study is to show the effect of guide vane geometry on the performance. In order to overcome the disadvantages of vertical axis wind turbine, a straight-bladed vertical axis wind turbine （S-VAWT） with a directed guide vane row has been proposed and tested by the authors. According to previous studies, it was clarified that the performance of the turbine can be improved by means of the directed guide vane row. However, the guide vane geometry of S-VAWT has not been optimized so far. In order to clarify the effect of guide vanegeometry, the effects of setting angle and gap between rotor blade and guide vane on power coefticlent and start- ing characteristic were investigated in the experiments. The experimental study of the proposed wind turbine was carried out by a wind tunnel. The wind tunnel with a diameter of 1.8m is open jet type. The wind velocity is 8 m/s in the experiments. The rotor has three straight blades with a profile of NACA0018 and a chord length of 100 mm, a diameter of 0.6 m and a blade height of 0.7 m. The guide vane row consists of 3 arc plates.

Periodical shedding of cloud cavitation from a single hydrofoil in high-speed cryogenic channel flow

In order to explain criteria for periodical shedding of the cloud cavitation, flow patterns of cavitation around a piano-convex hydrofoil were observed using a cryogenic cavitation tunnel of a blowdown type. Two hydrofoils of similarity of 20 and 60 mm in chord length with two test sections of 20 and 60 mm in width were prepared. Working fluids were water at ambient temperature, hot water and liquid nitrogen. The parameter range was varied between 0.3 and 1.4 for cavitation number, 9 and 17 m/sec for inlet flow velocity, and -8° and 8° for the flow incidence angle, respectively. At incidence angle 8°, that is, the convex surface being suction surface, periodical shedding of the whole cloud cavitation was observed on the convex surface under the specific condition with cavitation number and inlet flow velocity, respectively, 0.5, 9 m/sec for liquid nitrogen at 192℃ and 1.4, 11 m/sec for water at 88℃, whereas under the supercavitation condition, it was not observable. Periodical shedding of cloud cavitation occurs only in the case that there are both the adverse pressure gradient and the slow flow region on the hydrofoil.

Influence of image classification accuracy on saliency map estimation

Saliency map estimation in computer vision aims to estimate the locations where people gaze in images. Since people tend to look at objects in images, the parameters of the model pre-trained on ImageNet for image classification are useful for the saliency map estimation. However, there is no research on the relationship between the image classification accuracy and the performance of the saliency map estimation. In this study, it is shown that there is a strong correlation between image classification accuracy and saliency map estimation accuracy. The authors also investigated the effective architecture based on multi-scale images and the up-sampling layers to refine the saliencymap resolution. The model achieved the state-of-the-art accuracy on the PASCAL-S, OSIE, and MIT1003 datasets. In the MIT saliency benchmark, the model achieved the best performance in some metrics and competitive results in the other metrics.

Effect of End Plates on the Performence of a Wells Turbine for Wave Energy Conversion

In order to improve the performance of the Wells turbine for wave energy conversion, the effect of end plates on the turbine characteristics has been investigated experimentally by model testing under steady flow conditions. The end plate attached to the tip of the original rotor blade is slightly larger than the original blade profile. The characteristics of the Wells turbine with end plates have been compared with those of the original Wells turbine, i.e., the turbine without end plate. As a result, it has been concluded that the characteristics of the Wells turbine with end plates are superior to those of the original Wells turbine and the characteristics are dependent on the size and position of end plate. Furthermore, the effect of annular plate on the turbine performance, which encircles the turbine and is attached to the tip, was investigated as an additional experiment. However, its device was not effective in improving the turbine characteristics.

Characteristic of organic aerosol in a remote area of Okinawa Island

A continuous investigation of aerosol pollutants in Cape Hedo, Japan was conducted from October 2005 to August 2006 by Aerodyne aerosol mass spectrometer （AMS）. This article focused on the analysis of long-range transport of organic aerosol from continental origin to the remote island. Based on the transport distance and air mass origin, four main air mass origins were considered, including North China, South China, Japan and Korea. Although the mass concentration and air mass origin were quite different during study period, the mass spectrum and size distribution of organic matter and oxidized organics were similar, which showed uni-modal distribution with modal diameter of around 500 nm. The loss rate of organics was （5.15 ± 0.55） × 10^-6 s^-1 obtained by plotting the concentration against the transport time. Conversion rate of SO2 to sulfate and oxidation rate of organics were （1.07 ± 0.15） × 10^-5 and （1.09 ± 0.52） × 10^-6 s^-1, respectively.
