AIM: To clarify the relationship between age, menopause, and nonalcoholic fatty liver disease (NAFLD) in women. METHODS: We conducted a follow-up study on non-alcoholic fatty liver disease by using abdominal ultrasonography, and investigated the relationship of age and menopause with the development of NAFLD in women. We followed 1829 women and 2572 men (response rate, 86%) selected in 2001 to represent the non-institutionalized adult population of Gifu, Japan. Data collected included self-reported medical history, lifestyle factors, and menopausal status. The postmenopausal state was defined as beginning 1 year after the cessation of menses. We diagnosed NAFLD with the aid of abdominal ultrasonography by using diagnostic criteria describedRESULTS: The prevalence of NAFLD in women increases with age, but does not alter with age in men. Furthermore, the prevalence of NAFLD in premenopausal women (6%) was lower than that in men (24%) and in postmenopausal women (15%). The associations of the postmenopausal state and hormone replacement therapy with NAFLD were statistically significant in a univariate logistic regression model. At the follow-up examination, 67 women (5%) were newly diagnosed with NAFLD. The incidence of NAFLD was 3.5% (28/802) in premenopausal women, 7.5% (4/53) in menopausal women, 6.1% (24/392) in postmenopausal women, and 5.3% (11/206) in women receiving hormone replacement therapy. The weight gain in premenopausal women was equal to that in postmenopausal women. Metabolic syndrome and weight gain were independent risk factors for NAFLD in pre-and postmenopausal women, but age was an independent risk factor in premenopausal women only. CONCLUSION: Aging is a risk factor for NAFLD in premenopausal women, independent of weight gain or influence of metabolic syndrome.

AIM:To clarify the efficiency of the criterion of metabolic syndrome to detecting non-alcoholic fatty liver disease(NAFLD).METHODS:Authors performed a cross-sectional study involving participants of a medical health checkup program including abdominal *** study involved 11 714 apparently healthy Japanese men and women,18 to 83 years of *** was defined by abdominal ultrasonography without an alcohol intake of more than 20 g/d,known liver disease,or current use of *** revised criteria of the National Cholesterol Education Program Adult Treatment PanelⅢ were used to characterize the metabolic ***:NAFLD was detected in 32.2%(95%CI:31.0%-33.5%)of men(n=1874 of 5811)and in 8.7%(95%CI:8.0%-9.5%)of women(n=514 of 5903).Among obese people,the prevalence of NAFLD was as high as 67.3%(95%CI:64.8%-69.7%)in men and 45.8%(95%CI:41.7%-50.0%)in *** NAFLD was thought of as being the liver phenotype of metabolic syndrome,the prevalence of the metabolic syndrome among subjects with NAFLD was low both in men and women.66.8%of men and 70.4%of women with NAFLD were not diagnosed with the metabolic syndrome.48.2%of men with NAFLD and 49.8%of women with NAFLD weren't overweight[body mass index(BMI)≥25 kg/m2].In the same way,68.6%of men with NAFLD and 37.9%of women with NAFLD weren't satisfied with abdominal classification(≥90 cm for men and≥80 cm for women).Next,authors defined it as positive at screening for NAFLD when participants satisfied at least one criterion of metabolic *** sensitivity of the definition"at least 1 criterion"was as good as 84.8%in men and 86.6%in *** subjects by BMI,the sensitivity was higher in obese men and women than in non-obese men and women(92.3%vs 76.8%in men,96.1%vs 77.0%in women,respectively).CONCLUSION:Authors could determine NAFLD effectively in epidemiological study by modifying the usage of the criteria for metabolic syndrome.

Efficient Production of δ-Guaiene, an Aroma Sesquiterpene Compound Accumulated in Agarwood, by Mevalonate Pathway-Engineered Escherichia coli Cells

Mevalonate pathway for isoprenoid biosynthesis was constructed in Escherichia coli cells by the transformation with a gene cluster isolated from Streptomyces sp., and farnesyl diphosphate synthase and δ-guaiene synthase genes were coexpressed in this strain. This transformant was capable of liberating an appreciable amount of δ-guaiene, an aroma sesquiterpene compound accumulated in agarwood, and its concentration was elevated to more than 30 μg/ml culture by the incubation with mevalonolactone as an isoprene precursor in a nutrient-enriched Terrific broth. Coexpression of type 1 isopentenyl diphosphate isomerase plus acetoacetyl-CoA ligase genes also enhanced δ-guaiene production, and the concentration of the compound was approximately 38 - 42 μg/ml culture in the presence of mevalonolactone or lithium acetoacetate. These results clearly indicate that mevalonate pathway-engineered E. coli cells showed an appreciable δ-guaiene producing activity in the en- riched medium in the presence of appropriate isoprene precursors.

AIM:To clarify whether nonalcoholic fatty liver disease(NAFLD)increases the risk of cardiovascular ***:We carried out a prospective observational study with a total of 1637 apparently healthy Japanese men and women who were recruited from a health check-up *** was diagnosed by abdominal *** metabolic syndrome(MS)was defined according to the modified National Cholesterol Education Program(NCEP)ATP Ⅲ *** years after the baseline evaluations,the incidence of cardiovascular disease was assessed by a self-administered ***:Among 1221 participants available for outcome analyses,the incidence of cardiovascular disease was higher in 231 subjects with NAFLD at baseline(5 coronary heart disease,6 ischemic stroke,and 1 cerebral hemorrhage)than 990 subjects without NAFLD(3 coronary heart disease,6 ischemic stroke,and 1 cerebral hemorrhage).Multivariate analyses indicated that NAFLD was a predictor of cardiovascular disease independent of conventional risk factors(odds ratio 4.12,95% CI,1.58 to 10.75,P = 0.004).MS was alsoindependently associated with cardiovascular *** simultaneous inclusion of NAFLD and MS in a multivariate model revealed that NAFLD but not MS retained a statistically significant correlation with cardiovascular ***:Although both of them were predictors of cardiovascular disease,NAFLD but not MS retained a statistically significant correlation with cardiovascular disease in a multivariate *** is a strong predictor of cardiovascular disease and may play a central role in the cardiovascular risk of MS.

Production of Sucrose Crystals of Uni-Modal Size Distribution by Seeded Batch Cooling Crystallization

From sucrose aqueous solution of high viscosity, sucrose was crystallized by seeded cooling in a batch crystallizer. At low seed loadings, the product crystal size distribution (CSD) was wide-spread because of enormous secondary and primary nucleation. However, at high seed loadings, it became uni-modal, where the crystallization was dominated by seed growth with practically no secondary nucleation. Enough seeding was thus effective in suppressing nucleation even during batch crystallization with high viscosity solution. The volume mean size of the product crystals obtained at high seed loadings agreed with that calculated by a simple mass balance assuming growth-dominated crystallization with no change in the number of crystals.

Petrology and SHRIMP zircon geochronology of granulites from Vesleknausen, Lützow-Holm Complex, East Antarctica: Neoarchean magmatism and Neoproterozoic high-grade metamorphism

We report new petrological data and geochronological measurements of granulites from Vesleknausen in the highest-grade section of the L＆#252;tzow-Holm Complex, part of the Gondwana-assembling collisional orogen in East Antarctica. The locality is dominated by felsic to intermediate orthogneiss （charnockite and minor biotite gneiss）, mafic orthogneiss, and hornblende-pyroxene granulite, with deformed and undeformed dykes of metagranite and felsic pegmatite. Pseudosection analysis of charnockite in the system NCKFMASHTO, supported by geothermometry of mafic orthogneiss, was used to infer peak metamorphic temperatures of 750e850 ？C, approximately 150 ？C lower than those estimated for met-asedimentary gneisses from Rundv？gshetta, 6 km to the northeast. SHRIMP U-Pb analysis of zircons from feldspar-pyroxene gneiss, which corresponds to a partially molten patch around mafic orthogneiss, yielded a Concordia upper intercept ages of 2507.9 ？ 7.4 Ma, corresponding to the time of formation of the magmatic protolith to the orthogneiss. Partial melting during peak metamorphism probably took place between 591 and 548 Ma, as recorded in rims overgrew around magmatic zircon. Our results suggest that Rundv？gshetta-Vesleknausen-Strandnibba region in southwestern L＆#252;tzow-Holm Bay, where orthogneisses are dominant, consists of a single crustal block, possibly formed by ca. 2.5 Ga arc mag-matism. The Neoarchean magmatic terrane was tectonically mingled with other fragments （such as metasedimentary units in northern L＆#252;tzow-Holm Bay） by subduction/collision events during the as-sembly of Gondwana supercontinent, and subsequently underwent w850 ？C granulite-facies meta-morphosed during Neoproterozoic to Cambrian final collisional event.

AIM: To evaluate the neuroprotective effect of a dietary supplement(ClearVision EX~?;CV) against glutamateinduced excitotoxicity in ***: We evaluated the protective effects CV on glutamate-induced cell toxicity of an immortalized mouse hippocampal cell line(HT-22) in vitro and N-methyl-Daspartate(NMDA) induced retinal injury in vivo. Once-daily oral administration of CV or vehicle(5% Arabic gum) was started the day before the NMDA injection and continued until the end of the study. Electroretinograms(ERGs) were recorded to evaluate the retinal function at 2 d after NMDA injection. Furthermore, a histological evaluation, Western blot analysis, and immunohistochemistry were performed for assessing the signal transduction pathway. RESULTS: HT-22 cell death was induced by the addition of glutamate and co-incubation with CV protected against it. Oral administration of CV inhibited the decrease in scotopic threshold response amplitudes induced by the intravitreal injection of NMDA and those of the thickness of the inner retinal layer in the histological evaluation. The increased phosphorylated levels of extracellular signalregulated kinase(ERK) but not cAMP response element binding protein(CREB) or Akt were observed 1 h after NMDA injection in both the vehicle-and CV-treated rats;however, p ERK activation was no more upregulated at 3 h after NMDA injection. pERK upregulation was observed in Müller ***: CV shows a protective effect against both glutamate-induced HT-22 cell death and NMDAinduced retinal damage. pERK upregulation in the Müller cells plays a key role in the protective effect of CV against glutamate-induced retinal toxicity.

AIM To perform automatic gastric cancer risk classificationusing photofluorography for realizing effective mass screening as a preliminary study. METHODS We used data for 2100 subjects including X-ray images, pepsinogen Ⅰ and Ⅱ levels, PGⅠ/PGⅡ ratio, Helicobacter pylori (H. pylori) antibody, H. pylori eradication history and interview sheets. We performed two-stage classification with our system. In the first stage, H. pylori infection status classification was performed, and H. pylori-infected subjects were automatically detected. In the second stage, we performed atrophic level classification to validate the effectiveness of our *** Sensitivity, specificity and Youden index(YI) of H. pylori infection status classification were 0.884, 0.895 and 0.779, respectively, in the first stage. In the second stage, sensitivity, specificity and YI of atrophic level classification for H. pylori-infected subjects were 0.777, 0.824 and 0.601, respectively. CONCLUSION Although further improvements of the system are needed, experimental results indicated the effectiveness of machine learning techniques for estimation of gastric cancer risk.
