Two novel germline mutations of MLH1 and investigation of their pathobiology in hereditary non-polyposis colorectal cancer families in China

AIM： To detect germline mutations of MLH1, and investigate microsatellite instability and expression of MLH1 in tumor tissues of hereditary non-polyposis colorectal cancer （HNPCC） with two novel germline mutations, and further investigate the pathobiology of the two novel mutations of MLH1. METHODS： RNA was extracted from the peripheral blood of 12 patients from 12 different families that fulfilled the Amsterdam 11 Criteria for HNPCC. Germline mutations of MLH1 were determined by RT-PCR, followed by cDNA sequencing analysis. PCR-GeneScan analysis was used to investigate microsatellite instability with a panel of five microsatellite markers （BAT26, BAT25, D5S346, D2S123 and mfd15）, along with immunohistochemical staining to detect the expression of MLH1 protein in two patients＇ tumor tissues with novel mutations. RESULTS： Three germline mutations were found in four patients, one of the mutations has previously been reported, but the other two, CGC→TGC at codon 217 of exon 8 and CCG→CTG at codon 581 of exon 16, have not been reported. The two patients＇ tumor tissues with novel mutations had high-frequency microsatellite instability that showed more than two unstable loci, and both tumors lost their MLH1 protein expression. CONCLUSION： The two novel germline mutations of MLH1 in HNPCC families i.e. CGC→TGC at codon 217 of exon 8 and CCG→CTG at codon 581 of exon 16, are very likely to have pathological significance.

AIM： To detect the MLH1 gene promoter germline- methylation in probands of Chinese hereditary non- polyposis colorectal cancer （HNPCC）, and to evaluate the role of methylation in MLH1 gene promoter and molecular genetics in screening for ***： The promoter germline methylation of MLH1 gene was detected by methylation-specific PCR （MSP） in 18 probands from unrelated HNPCC families with high microsatellite-instability （MSI-H） phenotype but without germline mutations in MSH2, MLH1 and MSH6 genes. At the same time, 6 kindreds were col- lected with microsatellite-stability （MSS） phenotype but without germline mutations in MSH2, MIH1 and MSH6 genes as controls. The results of MSP were confirmed by clone sequencing. To ensure the reliability of the results, family H65 with nonsense germline mutation at c.2228C 〉 A in MSH2 gene was used as the negative control and the cell line sw48 was used as the known positive control along with water as the blank control. Immunochemical staining of MIH1 protein was performed with Envision two-step method in those patients with aberrant methylation to judge whether the status of MLH1 gene methylation affects the expression of MLH1 ***： Five probands with MIH1 gene promoter methylation were detected in 18 Chinese HNPCC families with MSI-H phenotype but without germline mutations in MSH2, MLH1 and MSH6 genes. Two of the five probands from families H10 and H29 displayed exhaustive-methylation, fulfilling the Japanese criteria （JC） and the Amsterdam criteria （AC）, respectively. The other 3 probands presented part-methylation fulfilling the AC. Of the 13 probands with unmethylation phenotype, 8 fulfilled the JC and the Bethesda guidelines （BG）, 5 fulfilled the AC. The rate of aberrant methylation in MLH1 gene in the AC group （22.2%, 4/18） was higher than that in the JC/BG groups （5.6%, 1/18） in all HNPCC families with MSI-H phenotype but without germline mutations in PISH2, PIIH1 and MSH6 genes. Three novel missense germline mutations in different exons of MSH6 gene in Chinese hereditary non-polyposis colorectal cancer families

AIM: To investigate the germline mutations of MSH6 gene in probands of Chinese hereditary non-polyposis colorectal cancer (HNPCC) families fulfilling different clinical criteria. METHODS: Germline mutations of MSH6 gene were detected by PCR-based DNA sequencing in 39 unrelated HNPCC probands fulfilling different clinical criteria in which MSH2 and MLH1 mutations were excluded. To further investigate the pathological effects of detected missense mutations, we analyzed the above related MSH6 exons using PCR-based sequencing in 137 healthy persons with no family history. The clinicopathological features were collected from the Archive Library of Cancer Hospital, Fudan University and analyzed. RESULTS: Four germline missense mutations distributed in the 4th, 6th and 9th exons were observed. Of them, three were not found in international HNPCC databases and did not occur in 137 healthy controls, indicating that they were novel missense mutations. The remaining mutation which is consistent with the case H14 at c.3488A>T of exon 6 of MSH6 gene was also found in the controls, the rate was approximately 3.65% (5/137) and the type of mutation was not found in the international HNPCC mutational and SNP databases, suggesting that this missense mutation was a new SNP unreported up to date. CONCLUSION: Three novel missense mutations and a new SNP observed in the probands of Chinese HNPCC families, may play an important role in the development of HNPCC.

Enhanced Crystallization of Poly(butylene adipate-co-terephthalate)by a Self-assembly Nucleating Agent

Poly(butylene adipate-co-terephthalate)(PBAT)is a promising biodegradable flexible polymer but suffers from slow crystallization rate,making it less attractive for some applications like the injection-molded products in comparison with low-density polyethylene(LDPE).This work aimed to accelerate the crystallization of PBAT by adding a self-assembly nucleating agent octamethylenedicarboxylic dibenzoylhydrazide(OMBH).PBAT/OMBH composites with various OMBH contents(0 wt%,0.5 wt%,0.7 wt%,1 wt%,2 wt%,3 wt%and 5 wt%)were prepared through *** effect of OMBH on the crystallization behavior,morphologies and mechanical properties of PBAT was *** highest nucleation efficiency value of 59.6%was achieved for PBAT with 0.7 wt%OMBH,much higher than that of 22.7%for PBAT with 0.7 wt%*** force microscopy results showed that OMBH formed fine fibers and induced the formation of transcrystalline layers of *** transform infrared spectroscopy(FTIR)combined with two-dimensional correlation spectra suggested that the intermolecular dipole-dipole N—H…O=C interactions but not hydrogen bond between OMBH and PBAT promoted the crystallization of PBAT in the initial period of *** presence of OMBH did not change the crystal form of PBAT but had positive contribution in enhancing its crystallinity and mechanical *** work is essential for preparing PBAT with high crystallization rate,enhancing its potential applications in injection-molded products.

Reliability ensemble averaging reduces surface wind speed projection uncertainties in the 21st century over China

Accurate prediction of future surface wind speed(SWS)changes is the basis of scientific planning for wind *** studies have projected SWS changes in the 21st century over China on the basis of the multi-model ensemble(MME)of the 6th Coupled Model Intercomparison Project(CMIP6).However,the simulation capability for SWS varies greatly in CMIP6 multi-models,so the MME results still have large *** this study,we used the reliability ensemble averaging(REA)method to assign each model different weights according to their performances in simulating historical SWS changes and project the SWS under different shared socioeconomic pathways(SSPs)in *** results indicate that REA considerably improves the SWS simulation capacity of CMIP6,eliminating the overestimation of SWS by the MME and increasing the simulation capacity of spatial *** spatial correlations with observations increased from 0.56 for the MME to 0.85 for ***,REA could eliminate the overestimation of the SWS by 33%in *** for southeastern China,the SWS generally decreases over China in the near term(2020-2049)and later term(2070-2099),particularly under high-emission *** SWS reduction projected by REA is twice as high as that by the MME in the near term,reaching-4%to-3%.REA predicts a larger area of increased SWS in the later term,which expands from southeastern China to eastern *** study helps to reduce the projected SWS uncertainties.

Thermal and hydrological processes in permafrost slope wetlands affect thermosyphon embankment stability

To ensure the long-term service performance of infrastructure such as railways,highways,airports and oil pipelines built on permafrost slope wetland sites,it is imperative to systematically uncover the long-term heat-water changes of soil in slope wetlands environment under climate *** specifically,considering valuable field data from 2001 to 2019,the long-term heat and water changes in active layers of the slope wetland site along the Qinghai-Xizang Railway(QXR)are illustrated,the effect of thermosyphon measures in protecting the permafrost environment is evaluated,and the influences of climate warming and hydrological effects on the stability of slope wetland embankments are systematically *** permafrost at the slope wetland site is rapidly degrading,demonstrating a reduction in active layer thickness of>3.7 cm per year and a permafrost temperature warming of>0.006℃ per *** thermosiphon embankment developed by QXR has a specific cooling period;thus,to mitigate the long-term impacts of climate warming on the thermal stability of permafrost foundation,it is essential to implement strengthening measures for the thermosiphon embankment,such as adding a crushed-rock layer or sunshade board on the slope of thermosiphon embankment to creating a composite cooling ***-term seasonal groundwater seepage intensifies frost damage to the slope wetland embankment,while long-term seasonal supra-permafrost water and groundwater seepage exacerbates uneven transverse deformation of slope wetland ***-term climate warming and slope effects have altered the surface water and groundwater hydrological processes of slope wetlands,potentially leading to an increased occurrence of slope embankment *** results are crucial for improving our understanding of heat and water variation processes in the active layer of slope wetland sites located in permafrost regions and ensuring long-term service safety for the QXR.

Sedimentary paleoenvironment and its control on organic matter enrichment in the Mesoproterozoic hydrocarbon source rocks in the Ordos Basin, southern margin of the North China Craton

The black shale of the Mesoproterozoic Cuizhuang Formation in the Changcheng System in Yongji city,North China Craton, is a potential source rock. Understanding the organic matter enrichment mechanism is crucial for evaluating source rock resources and understanding oil and gas accumulation *** this study, we evaluated the sedimentary paleoenvironment and organic matter enrichment mechanisms of shale using thin section observations, mineral composition analysis, organic geochemistry, and elemental geochemistry. We found significant differences in the sedimentary paleoenvironment and organic matter enrichment mechanisms between the lower Cuizhuang Formation and the Beidajian Formation shale. The Cuizhuang Formation was deposited in a late-stage, restricted basin environment during the rift phase, and elemental and geochemical indicators showed that the Cuizhuang Formation was in a suboxic-anoxic water environment, that was influenced by a warm and humid paleoclimate and submarine hydrothermal activities, which promoted the accumulation of organic matter. However, the enrichment of organic matter in the Cuizhuang Formation was mainly controlled by redox *** formation of suboxic-anoxic water environments may be closely related to the warm and humid paleoclimate and submarine hydrothermal activities. Warm conditions promote continental weathering and increase marine productivity, thereby consuming oxygen in the bottom water. Moreover, acidic hydrothermal activity also helps to establish an anoxic environment. Our results reveal the effects controlling various coupled mechanisms dominated by redox conditions, which may explain the development of source rocks in the Cuizhuang Formation.

Luminescence properties and optical sensing behaviors of Sr_(2)GdSbO_(6):Eu^(3+) phosphors

Commonsense question answering(CQA)requires understanding and reasoning over QA context and related commonsense knowledge,such as a structured Knowledge Graph(KG).Existing studies combine language models and graph neural networks to model ***,traditional knowledge graph are mostly concept-based,ignoring direct path evidence necessary for accurate *** this paper,we propose MRGNN(Meta-path Reasoning Graph Neural Network),a novel model that comprehensively captures sequential semantic information from concepts and *** MRGNN,meta-paths are introduced as direct inference evidence and an original graph neural network is adopted to aggregate features from both concepts and paths *** conduct sufficient experiments on the CommonsenceQA and OpenBookQA datasets,showing the effectiveness of ***,we conduct further ablation experiments and explain the reasoning behavior through the case study.

Yolk free egg substitute improves the serum phospholipid profile of mice with metabolic syndrome based on lipidomic analysis

In this study,the impacts of egg consumption on mice model of metabolic syndrome(Met S)were comparatively *** were divided into five groups(n=8):normal diet group(ND),high-fat diet group(HFD),HFD with whole egg group(WE),HFD with free-yolk egg substitute group(YFES),and HFD with lovastatin group(Lov).Main biochemical indexes and a non-targeted lipidomic analysis were employed to insight the lipid profile changes in *** was revealed that WE could significantly improve serum biochemical indexes by reducing body weight,low-density lipoprotein cholesterol(LDL-C)and total cholesterol(TC),while increasing high-density lipoprotein *** exhibited remarkably better performance in increasing phosphatidylglycerol and phosphatidic acids,while decreasing phosphatidylinositol than WE.A total of 50 differential lipids biomarkers tightly related to glycerophospholipids metabolism were screened *** C18:2 and C12:1,SM(d18:0/12:0),and SM(d18:1/14:1)were significantly upregulated in YFES compared to *** reduced expression of SREBP-1c and Cpt1a,while did not affect the expression of PPAR-α.Sphingomyelin biomarkers were positively related to the TC(|r|>0.6),while PPAR-αwas negatively correlated with triglyceride and LDL-C *** sum up,YFES attenuated HFD-induced Met S by improving the serum phospholipids,which account for its modulation of glycerophospholipid metabolism.
