Ischemic stroke can cause blood-brain barrier(BBB)injury,which worsens brain damage induced by stroke.Abnormal expression of tight junction proteins in endothelial cells(ECs)can increase intracellular space and BBB leakage.Selective inhibition of mitogen-activated protein kinase,the negative regulatory substrate of mitogen-activated protein kinase phosphatase(MKP)-1,improves tight junction protein function in ECs,and genetic deletion of MKP-1 aggravates ischemic brain injury.However,whether the latter affects BBB integrity,and the cell type-specific mechanism underlying this process,remain unclear.In this study,we established an adult male mouse model of ischemic stroke by occluding the middle cerebral artery for 60 minutes and overexpressed MKP-1 in ECs on the injured side via lentiviral transfection before stroke.We found that overexpression of MKP-1 in ECs reduced infarct volume,reduced the level of inflammatory factors interleukin-1β,interleukin-6,and chemokine C-C motif ligand-2,inhibited vascular injury,and promoted the recovery of sensorimotor and memory/cognitive function.Overexpression of MKP-1 in ECs also inhibited the activation of cerebral ischemia-induced extracellular signal-regulated kinase(ERK)1/2 and the downregulation of occludin expression.Finally,to investigate the mechanism by which MKP-1 exerted these functions in ECs,we established an ischemic stroke model in vitro by depriving the primary endothelial cell of oxygen and glucose,and pharmacologically inhibited the activity of MKP-1 and ERK1/2.Our findings suggest that MKP-1 inhibition aggravates oxygen and glucose deprivation-induced cell death,cell monolayer leakage,and downregulation of occludin expression,and that inhibiting ERK1/2 can reverse these effects.In addition,co-inhibition of MKP-1 and ERK1/2 exhibited similar effects to inhibition of ERK1/2.These findings suggest that overexpression of MKP-1 in ECs can prevent ischemia-induced occludin downregulation and cell death via deactivating ERK1/2,thereby protecting the integrity of BBB

Chronic rhinosinusitis with nasal polyps(CRSw-NP)is a multifactorial and heterogeneous disorder of the sinonasal mucosa,characterized by a high relapse rate after medical and surgical treatments[1].Endoscopic sinus surgery(ESS)is the preferred treatment for CRSwNP in patients who have failed to respond to medical therapy[2].However,the postoperative complications will impair the recovery of patients,and accompanying post-operative outpatient visits may contribute to patient discomfort[3]Suppressing postoperative mucosa edema and improving mucosa recovery is critical to reducing the disease relapse after ESS.The steroid-eluting sinus stent(SESS)represents a promising emerging therapy for CRSwNP.

Bilayer Compounded Polytetrafluoroethylene Membrane for Enhanced Oil-Water Emulsion Separation

In order to achieve efficient and durable oil-water emulsion separation,the membranes possessing high separation efficiency and mechanical strength attract extensive attention and are in great demand.In present study,a kind of polytetrafluoroethylene(PTFE)-based bilayer membrane was fabricated by electrospinning fibrous PTFE(fPTFE)on an expanded PTFE(ePTFE)substrate.The morphological observation revealed that the fibrous structure of the fPTFE layer could be tailored by controlling the formulation of spinning solution.The addition of appropriate polyoxyethylene(PEO)would make the fibers in the fPTFE layer finer and more uniform.As a result,the compounded membrane exhibited a small pore size of approximately 1.25µm and a substantial porosity nearing 80%.This led to super-hydrophobicity,characterized by a high water contact angle(WCA)of 149.8°,and facilitated rapid oil permeation.The water-in-oil emulsion separation experiment further confirmed that the compounded membrane not only had a high separation efficiency closing 100%,but such an outstanding separation capacity could be largely retained,either through multiple cycles of use or through strong acid(pH=1),strong alkali(pH=12),or high-temperature(100°C)treatment.Additionally,the mechanical behavior of the bilayer membrane was basically contributed by that of each layer in terms of their volume ratio.More significantly,the poor creep resistance of fPTFE layer was suppressed by compounding with ePTFE substrate.Hence,this study has laid the groundwork for a novel approach to create PTFE-based compounded membranes with exceptional overall characteristics,showing promise for applications in the realm of emulsion separation.

Soluble mannose receptor as a predictor of prognosis of hepatitis B virus-related acute-on-chronic liver failure

BACKGROUND Hepatitis B virus-related acute-on-chronic liver failure(HBV-ACLF)is a syndrome with a high short-term mortality rate,and it is crucial to identify those patients at a high mortality risk clinically.AIM To investigate the clinical value of soluble mannose receptor(sMR)in predicting the 90-day mortality of HBV-ACLF patients.METHODS A total of 43 patients were diagnosed with HBV-ACLF between October 2017 and October 2018 at the Second Hospital of Anhui Medical University,and all of them were enrolled in this retrospective study.Their serum sMR levels were determined using an enzyme-linked immunosorbent assay.Demographic and clinical data,including gender,age,albumin level,total bilirubin(TBIL)level,international normalized ratio,HBV-DNA level,HBV serological markers,procalcitonin level,interleukin-6 level,and model for end-stage liver disease(MELD)score were accessed at the time of diagnosis of HBV-ACLF.A multivariate logistic regression analysis was used to analyze the independent risk factors for mortality.RESULTS Serum sMR level was significantly increased in HBV-ACLF patients compared with chronic hepatitis B patients and healthy controls(P<0.01).When compared with surviving patients,it was higher in those patients who succumbed to HBVACLF(P<0.05).Serum sMR level was positively correlated with MELD score(rs=0.533,P=0.001),HBV-DNA level(rs=0.497,P=0.022),and TBIL level(rs=0.894,P<0.001).Serum sMR level(odds ratio=1.007,95%confidence interval:1.004–1.012,P=0.001)was an independent risk factor for the 90-day mortality in the HBV-ACLF cases.The patients with HBV-ACLF were stratified into two groups in accordance with their serum sMR levels at the baseline(low risk:<99.84 pg/mL and high risk:≥99.84 pg/mL).The 90-day mortality rates were 27.3%in the low-risk group and 87.5%in the high-risk group.Furthermore,sMR level apparently improved the performance of MELD score for predicting the prognosis of patients with HBV-ACLF.CONCLUSION Serum sMR level may be a predictor of the prognosis of HBV-ACLF patients.

An experimental picosecond time-resolved X-ray ferromagnetic resonance(TR-XFMR)apparatus with a time resolution of 13 ps(RMS)or 31 ps(FWHM)was constructed and demonstrated in the 07U and 08U1A soft X-ray beamlines at the Shanghai Synchrotron Radiation Facility(SSRF)using pump-probe detection and X-ray magnetic circular dichroism(XMCD)spectroscopy.Element and time-resolved ferromagnetic resonance was excited by continuous microwave phase-locking of the bunch clock within the photon beam during synchrotron radiation and was characterized by detecting the magnetic circular dichroism signals of the elements of interest in the magnetic films.Using this equipment,we measured the amplitude of the element-specific moment precession during ferromagnetic resonance(FMR)at 2 GHz in a single Ni81Fe19layer.

Enhanced recovery after surgery protocols in functional endoscopic sinus surgery for patients with chronic rhinosinusitis with nasal polyps: a randomized clinical trial

Background:Enhanced recovery after surgery (ERAS) protocols are a series of perioperative care to optimize preoperative preparation, prevent postoperative complications, minimize stress, and speed up recovery. This study aimed to assess the impact of ERAS protocols for functional endoscopic sinus surgery (FESS) in patients with chronic rhinosinusitis with nasal polyps (CRSwNP).Methods:One hundred and two patients with CRSwNP undergoing FESS were randomly divided into the ERAS group and the control group. The outcomes of the Self-Rating Anxiety Scale (SAS), Visual Analogue Scale (VAS), Medical Outcomes Study Sleep Scale (MOS-SS) and Kolcaba Comfort Scale Questionnaire (GCQ) were determined in both groups. The serum levels of C-reactive protein (CRP) were compared preoperatively and 24 hours postoperatively.Results:The ERAS group had a significantly better SAS scores than did the control group (28 [24, 35] vs. 43 [42, 47], Z=5.968, P 0.05);However, the CRP level in 24 hours postoperatively was significantly lower in the ERAS group than that in the control group (2.5 [1.4, 3.9] vs. 6.6 [3.8, 9.0], Z = 5.027, P 0.05), hemorrhage, aspiration and tumble, were not increased in the ERAS group compared with those in the control group. The ERAS group had a significantly shorter length of hospital stay (5 [4, 5] days vs. 8 [8,9] days, Z=8.939, P<0.001) and hospitalization expenses ($ 2670 [2375, 2740] vs. $3129 [3116, 3456], Z=8.514, P<0.001).Conclusions:ERAS pro

Objective:Analyzing the symptom characteristics of Coronavirus Disease 2019(COVID-19)to improve control and prevention.Methods:Using the Baidu Index Platform(http://***)and the website of Chinese Center for Disease Control and Prevention as data resources to obtain the search volume(SV)of keywords for symptoms associated with COVID-19 from January 1 to February 20 in each year from 2017 to 2020 and the epidemic data in Hubei province and the other top 9 impacted provinces in China.Data of 2020 were compared with those of the previous three years.Data of Hubei province were compared with those of the other 9 provinces.The differences and characteristics of the SV of COVID-19-related symptoms,and the correlations between the SV of COVID-19 and the number of newly confirmed/suspected cases were analyzed.The lag ef-fects were discussed.Results:Comparing the SV from January 1,2020 to February 20,2020 with those for the same period of the previous three years,Hubei’s SV for cough,fever,diarrhea,chest tightness,dys-pnea,and other symptoms were significantly increased.The total SV of lower respiratory symptoms was significantly higher than that of upper respiratory symptoms(P<0.001).The SV of COVID-19 in Hubei province was significantly correlated with the number of newly confirmed/suspected cases(rconfirmed Z 0.723,rsuspected Z 0.863,both p<0.001).The results of the distributed lag model suggested that the patients who searched relevant symptoms on the Internet may begin to see doctors in 2e3 days later and be confirmed in 3e4 days later.Conclusion:The total SV of lower respiratory symptoms was higher than that of upper respira-tory symptoms,and the SV of diarrhea also increased significantly.It warned us to pay atten-tion to not only the symptoms of the lower respiratory tract but also the gastrointestinal symptoms,especially diarrhea in patients with *** search behavior had a pos-itive correlation with the number of newly confirmed/suspected cases,suggesting that big data has an important role in the early warning of

In this study,the Jinzhou 9-3 CEPD float-over installation project was investigated.During the undocking condition,the water depth of the motion path of the working barge gradually changed from 10.31 m to 9.41 m.The undocking clearance of the HYSY 228 is smaller than 1 m;therefore,the barge shows highly nonlinear hydrodynamic characteristics,and it is difficult to be accurately simulated by numerical analysis.Thus,it is necessary to obtain the hydrodynamic characteristics and laws of the float-over barge at different water depths by using tank model test,to provide some reference and guidance for float-over operations in shallow water.

A simple method to extract glacier length based on Digital Elevation Model and glacier boundaries for simple basin type glacier

Glacier length is a key morphological element that has many glaciological applications; however, it is often difficult to determine, especially for glaciers that cover larger spatial areas or those that exhibit frequent temporal change. In this paper, we describe a new Arc GIS-based method that can derive glacier flow lines for determining glacier length based on digital elevation model and glacier outlines. This method involves(1) extraction of the highest and lowest points on a glacier,(2) calculation of 10-m contour lines on the glacier from 10 m to 100 m height, and(3) connection of the midpoints of each contour line with the highest and the lowest points in order to create a flow line, which is subsequently smoothed. In order to assess the reliability of this method, we tested the algorithm's results against flow lines calculated using field measurements, analysing data from the Chinese Glacier Inventory, and manual interpretation. These data showed that the new automated method is effective in deriving glacier flow lines when contour lines are relatively large; in particular, when they are between 70 m and 100 m. Nonetheless, a key limitation of the algorithm is the requirement to automatically delete repeated and closed curves in the pre-treatment processes. In addition to calculating glacier flow lines for derivation of glacier length, this method also can be used to effectively determine glacier terminus change.
