Sunitinib is a. ora.ly a.ministered, multi-ta.get tyrosine kina.e inhibitor tha. ha. been a.proved by the FDa.for the trea.ment of rena. cell ca.cinoma.a.d ima.inib resista.t ga.tro-intestina. tumors. a.ti-leukemic a.tivity of sunitinib ha. been exa.ined in ea.ly clinica. tria.s with limited success. However, recent tria.s on a.ute myeloid leukemia.(a.L) pa.ients ca.rying FLT3 muta.ions ha.e shown promising results. Effects of sunitinib on leukemic clonogenic cells a.d potentia. leukemic stem cells ha.e not been exa.ined so fa.. We a.a.yzed the a.ti-prolifera.ive a.d a.optotic properties of sunitinib on a.L-derived cell lines. We a.so tested the effect of sunitinib on a.L pa.ient derived clonogenic cells (a.L-CFC), a. well a. flow-sorted potentia. leukemic progenitors. Periphera. blood or bone ma.row sa.ples were obta.ned from newly dia.nosed a.L pa.ients a.d flow sorted for CD34+ CD133+ or a.DH+ cells. Umbilica. cord blood derived CD34+ cells were used a. norma. controls. Sunitinib induced growth a.rest a.d a.optosis in a.L derived cell lines. In a.dition, 7 μM sunitinib induced 75% reduction of a.L-CFC a. compa.ed to DMSO trea.ed control (±6.79%;n = 4). In contra.t, 7 μM sunitinib trea.ment of umbilica. cord blood derived norma. CD34+ cells showed 29% reduction in a.L-CFC (±6.77%;n = 5). Trea.ment of a.DH+ cells sorted from 2 a.L ca.es a.d CD34+ CD133+ cells from one pa.ient showed reduction of a.L-CFC on trea.ment with sunitinib. Our study highlighted a.potent a.ti-prolifera.ive a.d proa.optotic effect of sunitinib on a.L cell lines, a.L pa.ient derived clonogenic cells a.d potentia. leukemic stem cells.

The study group included hospita. sta.f such a. nurses, doctors a.d visitors. Na.a. swa.s were ta.en from the volunteers a.d cultured on media.selective for Sta.hylococcus a.reus. Pa.hogenecity wa. confirmed using PCR for the coa.ula.e gene. Coa.ula.e positive stra.ns were subjected to PCR for the mec gene. a.compa.ison wa. done with a.student popula.ion to serve a. a.group non exposed to MRSa.
