Water-soluble lipopolymer delivery of N-methyl-D-aspartic acid receptor 2B siRNA relieves chronic neuropathic pain in rats

Spinal dorsal horn N-Methyl-D-aspartic acid receptor 2B （NR2B） overexpression plays an important role in the production and maintenance of neuropathic pain. Because small interfering RNA （siRNA） can inhibit NR2B expression, siRNA may provide a novel approach to treat neuropathic pain and possibly nerve injury. However, an efficient and safe vector for NR2B siRNA has not been discovered. This study shows that a water soluble lipopolymer （WSLP） comprised of low molecular weight polyethyleneimine （PEI） and cholesterol can deliver siRNA targeting NR2B for the treatment of neuropathic pain. Results show that intrathecal injection of WSLP/siRNA complexes for 3 days inhibit NR2B gene expression with reductions in mRNA and protein levels by 59% and 54%, respectively, compared with control rats （P 〈 0.01）. Injection of WSLP complexed with scrambled siRNA, or PEI with siRNA did not show this inhibitory effect. Moreover, injection of WSLP/siRNA complexes significantly relieved neuropathic pain at 3, 7, 12, and 21 days, while injection of WSLP with scrambled siRNA or PEI with siRNA did not. These results demonstrate that WSLP can efficiently deliver siRNA targeting NR2B in vivo and relieve neuropathic pain.

The article introduces a finite element procedure using the bilinear quadrilateral element or four-node rectangular element(namely Q4 element) based on a refined first-order shear deformation theory(rFSDT) and Monte Carlo simulation(MCS), so-called refined stochastic finite element method to investigate the random vibration of functionally graded material(FGM) plates subjected to the moving *** advantage of the proposed method is to use r-FSDT to improve the accuracy of classical FSDT, satisfy the stress-free condition at the plate boundaries, and combine with MCS to analyze the vibration of the FGM plate when the parameter inputs are random quantities following a normal distribution. The obtained results show that the distribution characteristics of the vibration response of the FGM plate depend on the standard deviation of the input parameters and the velocity of the moving ***, the numerical results in this study are expected to contribute to improving the understanding of FGM plates subjected to moving loads with uncertain input parameters.

With the development of satellite miniaturization and remote sensing,the establishment of microsatellite constellations is an inevitable *** to their limited size,weight,and power,spaceborne storage systems with excellent scalability,performance,and reliability are still one of the technical bottlenecks of remote sensing *** on the commercial off-the-shelf field-programmable gate array and memory devices,a spaceborne advanced storage system(SASS)is proposed in this *** work provides a dynamic programming,queue scheduling multiple-input multiple-output cache technique and a high-speed,high-reliability NAND flash controller for multiple microsatellite payload *** results show that SASS has outstanding scalability with a maximum write rate of 2429 Mb/s and preserves at least 78.53%of the performance when a single NAND flash *** scheduling technique effectively shortens the data scheduling time,and the data remapping method of the NAND flash controller can reduce the retention error by at least 50.73%and the program disturbance error by at least 37.80%.

An integrated MINLP model for multi-party coordination in downstream oil supply chain

Cooperation among enterprises can bring overall and individual performance improvement, and a smooth coordination method is indispensable. However, due to the lack of customized coordination methods, cooperation in the downstream oil supply chain cannot be carried out smoothly. This paper intends to propose a multi-party coordination method to promote cooperation between oil shippers and pipeline operator by optimizing oil transportation, oil substitution and pipeline pricing schemes. An integrated game-theoretic modeling and analysis approach is developed to characterize the operation behaviors of all stakeholders in the downstream oil supply chain. The proposed mixed integer nonlinear programming model constrains supply and demand capacity, transportation routes, oil substitution rules and pipeline freight levels. Logarithm transformation and price discretization are introduced for model linear approximation. Simulation experiments are carried out in the oil distribution system in South China. The results show that compared to the business-as-usual scheme, the new scheme saves transportation cost by 3.48%, increases pipeline turnover by 5.7%, and reduces energy consumption and emissions by 7.66% and 6.77%. It is proved that the proposed method improves the revenue of the whole system, achieves fair revenue distribution, and also improves the energy and environmental benefits of the oil supply chain.
