Schisandrin B Protects against Ischemic Brain Damage by Regulating PI3K/AKT Signaling in Rats

Objective To explore the effect and mechanism of schisandrin B(Sch B)in the treatment of cerebral ischemia in rats.Methods The cerebral ischemia models were induced by middle cerebral artery occlusion(MCAO)and reperfusion.Sprague-Dawley rats were divided into 6 groups using a random number table,including sham,MCAO,MCAO+Sch B(50 mg/kg),MCAO+Sch B(100 mg/kg),MCAO+Sch B(100 mg/kg)+LY294002,and MCAO+Sch B(100 mg/kg)+wortmannin groups.The effects of Sch B on pathological indicators,including neurological deficit scores,cerebral infarct volume,and brain edema,were subsequently studied.Tissue apoptosis was identified by terminal transferase-mediated dUTP nick end-labeling(tuNEL)staining.The protein expressions involved in apoptosis,inflammation response and oxidative stress were examined by immunofluorescent staining,biochemical analysis and Western blot analysis,respectively.The effect of Sch B on phosphatidylinositol 3-kinase(PI3K)/protein kinase B(AKT)signaling was also explored.Results Sch B treatment decreased neurological deficit scores,cerebral water content,and infarct volume in MCAO rats(PtuNEL staining indicated that Sch B also reduced apoptosis in brain tissues,as well as the Bax/Bcl-2 ratio and caspase-3 expression(P<0.01).Sch B regulated the production of myeloperoxidase,malondialdehyde,nitric oxide and superoxide dismutase,as well as the release of cytokine interleukin(IL)-1βand IL-18,in MCAO rats(P<0.05 or P<0.01).Sch B promoted the phosphorylation of PI3K and AKT.Blocking the PI3K/AKT signaling pathway with LY294002 or wortmannin reduced the protective effect of Sch B against cerebral ischemia(P<0.05 or P<0.01).Conclusions Sch B reduced apoptosis,inflammatory response,and oxidative stress of MCAO rats by modulating the PI3K/AKT pathway.Sch B had a potential for treating cerebral ischemia.

Combined analysis of imaging tumor capsule with imaging tumor size guides the width of resection margin for solitary hepatocellular carcinoma

Background:The optimal width of resection margin(RM)for hepatocellular carcinoma(HCC)remains controversial.This study aimed to investigate the value of imaging tumor capsule(ITC)and imaging tu-mor size(ITS)in guiding RM width for patients with HCC.Methods:Patients who underwent hepatectomy for HCC in our center were retrospectively reviewed.ITC(complete/incomplete)and ITS(≤3 cm/>3 cm)were assessed by preoperative magnetic resonance imaging(MRI).Using subgroup analyses based on ITC and ITS,the impact of RM width[narrow RM(3 cm and complete ITC,the narrow RM group exhibited comparable RFS,OS,and incidence of RM recurrence with the wide RM group(P=0.606,0.916,and 0.649,respectively).However,in patients with ITS>3 cm and incomplete ITC,the wide RM group showed better RFS and OS and a lower incidence of RM recurrence compared with the narrow RM group(P=0.037,0.018,and 0.046,respectively).Conclusions:As MRI-based preoperative markers,conjoint analysis of ITC with ITS aids in determining RM width for solitary HCC patients.Narrow RM is applicable in patients with ITS≤3 cm regardless of ITC status and in those with ITS>3 cm and complete ITC.Wide RM is preferred in those with ITS>3 cm and incomplete ITC.

Pressure evolution in shock-compacted granular media

The pressure evolution associated with the transient shock-induced infiltration of gas flow through granular media consisting of mobile particles is numerically investigated using a coupled Eulerian–Lagrangian approach.The coupling between shock compaction and interstitial flow has been revealed.A distinctive two-stage diffusing pressure field with deflection occurring at the tail of the compaction front is found,with corresponding spikes in both gaseous velocity and temperature profiles emerging within the width of the compaction front.The compaction front,together with the deflection pressure,reaches a steady state during the later period.An analytical prediction of the steady deflection pressure that considers the contributions of porosity and the non-isothermal effect is proposed.The isothermal single-phase method we developed,combining the porosity jump condition across the compaction front,shows consistent pressure evolution with the non-isothermal CMP-PIC one under weak shock strength and low column permeability.Lastly,the microscale mechanism governing the formation of not only pressure deflection but also gaseous velocity and temperature spikes within the width of the compaction front has been described.These aforementioned evolutions of the flow field are shown to arise from the nozzling effects associated with the particle-scale variations in the volume fraction.

前列腺癌组织中Plexin-B1的表达及其与AR表达的相关性

目的探讨前列腺癌组织中Plexin-B1的表达与临床病理学特征、预后的关系,及其与AR表达的相关性。方法采用免疫组化EnVision法检测82例前列腺癌及46例良性前列腺组织中Plexin-B1和AR的表达,应用qRT-PCR检测20对前列腺癌和良性前列腺组织中Plexin-B1 mRNA的表达。结果前列腺癌组织中Plexin-B1阳性率(74.39%)明显高于良性前列腺组织(10.87%,P<0.01)。Plexin-B1表达与前列腺癌临床分期、WHO/ISUP分组以及患者总生存期相关(P<0.01,P<0.05)。qRT-PCR检测显示Plexin-B1 mRNA在前列腺癌组织中高表达(267.15±57.06)%(P<0.05)。Kaplan-Meier生存分析显示,Plexin-B1高表达组累积生存率(55.74%)低于低表达组(85.71%),差异有统计学意义(P<0.05)。Spearman等级相关性分析显示,前列腺癌中Plexin-B1表达与AR表达呈正相关(r=0.275,P=0.012)。结论Plexin-B1在前列腺癌组织中过表达,与前列腺癌组织临床分期、WHO/ISUP分组密切相关,有望成为判断前列腺癌预后的分子标志物。

A review of space-object collision probability computation methods

The collision probability computation of space objects plays an important role in space situational awareness,particularly for conjunction assessment and collision avoidance.Early works mainly relied on Monte Carlo simulations to predict collision probabilities.Although such simulations are accurate when a large number of samples are used,these methods are perceived as computationally intensive,which limits their application in practice.To overcome this limitation,many approximation methods have been developed over the past three decades.This paper presents a comprehensive review of existing space-object collision probability computation methods.The advantages and limitations of different methods are analyzed and a systematic comparison is presented.Advice regarding how to select a suitable method for different short-term encounter scenarios is then provided.Additionally,potential future research avenues are discussed.

针对传统水闸无法实现渠道自动清污和清淤的问题,提出了一种集拦污栅、清淤器和液压伺服控制系统于一体的清污水闸。首先,利用SolidWorks软件设计了一体化清污水闸的三维机械结构,并对其清淤、清污动作进行了模拟仿真;然后,对其核心受力部件液压缸进行了受力分析,通过ANSYS Workbench软件对液压缸和底座进行了校核测试,并分析了液压缸和伺服电机的选型过程和方法;最后,利用MATLAB软件对液压伺服测试平台模型进行了仿真,得到了系统的动态性能曲线,并对比分析了PID控制和非PID控制系统的响应情况。研究结果表明:一体化清污水闸不仅能实现清污和清淤功能,而且符合受力和校核要求;同时,采用PID算法能在0.38 s左右使系统达到稳定,且稳态误差趋向于0,可保证拦污栅和清淤器快速准确定位,满足农业灌区渠道的无人化管理需求。

Low expression of CDK5RAP3 and DDRGK1 indicates a poor prognosis in patients with gastric cancer

AIM To investigate the effects of different levels of expression of CDK5RAP3 and DDRGK1 on long-term survival of patients undergoing radical gastrectomy.METHODS The expression of CDK5RAP3 and DDRGK1 was detected by immunohistochemistry in 135 patients who received standard gastrectomy were enrolled in the study. Western Blot was used to detect the expression of CDK5RAP3 and DDRGK1 in gastric cancer and its adjacent tissues and cell lines. The correlations between the expression of CDK5RAP3 and DDRGK1 and clinicopathological factors were analyzed, and the value of each parameter to the prognosis of the patients was compared. Receiver operating characteristic analysis was used to compare the accuracy of the prediction of clinical outcome by the parameters.RESULTS CDK5RAP3 and DDRGK1 expression was downregulated in the gastric cancer compared to its respective adjacent non-tumor tissues. The expression of CDK5RAP3 was closely related to the age of the patients(P = 0.035) and the T stage of the tumor(P = 0.017). The expression of DDRGK1 was correlated with the sex of the patients(P = 0.080), the degree of tumor differentiation(P = 0.036), the histological type(P = 0.036) and the N stage of the tumor(P = 0.014). Low expression CDK5RAP3 or DDRGK1 is a poor prognostic factor for gastric cancer patients. Prognostic analysis showed that the co-expression of CDK5RAP3 and DDRGK1 was an independent prognostic factor correlating with the overall survival of gastric cancer patients. Combined expression analysis of CDK5RAP3 and DDRGK1 may provide a more accurate prognostic value for overall survival.

Mainchain Semifluorinated Polymers with Ultra High Molecular Weight via Reaction-enhanced Reactivity of Intermediate(RERI) Mechanism

Achieving the linear polymers with high molecular weight via step-growth polymerization of A2 and B2 monomers is significantly limited by the requirement of strict stoichiometry of two monomers when the reactivity of A and B groups are not changed during the polymerization.Herein,a unique step-growth polymerization based on copper-catalyzed azide-alkyne cycloaddition(CuAAC)with reactionenhanced reactivity of intermediate(RERI)mechanism was developed for the preparation of mainchain semifluorinated polymers with high molecular weight.The CuAAC polymerization of bis-alkynyl-terminated fluorinated monomers(A2)and 2,2-bis(azidomethyl)propane-1,3-diyl bis(2-methylpropanoate)(BiAz,B2)with RERI effect at different stoichiometric ratio was systematically investigated.The results indicated that the semifluorinated polymers with ultrahigh molecular weight,Mw,MALLS>106 g/mol,could be efficiently synthesized by using excess molar of BiAz monomers.The resultant high-molecular-weight semifluorinated polymers show good thermostability and high hydrophobicity.In addition,the glass transition temperature(Tg)of these mainchain semifluorinated polymers could be tuned conveniently due to the bis-alkynyl-terminated comonomers could be consumed completely when excessive BiAz monomers were used in this this step-growth polymerization.
