Legionella dumoffii Tex-KL Mutated in an Operon Homologous to traC-traD is Defective in Epithelial Cell Invasion

Objective To understand the mechanism of invasion by Legionella dumoffii. Methods The L. dumoffii strain Tex-KL was mutated using the Tn903 derivative, Tn903 d IIlac Z. After screening 799 transposon insertion mutants, we isolated one defective mutant. We then constructed the gene-disrupted mutant, KL16, and studied its invasion of and intracellular growth in He La and A549 cells, and in A/J mice survival experiments. The structure of traC-traD operon was analyzed by RT-PCR. Results The transposon insertion was in a gene homologous to Salmonella typhi tra C, which is required for the assembly of F pilin into the mature F pilus structure and for conjugal DNA transmission. Results from RT-PCR suggested that the traC-traD region formed an operon. We found that when the tra C gene was disrupted, invasion and intracellular growth of L. dumoffii Tex-KL were impaired in human epithelial cells. When mice were infected by intranasal inoculation with a tra C deficient mutant, their survival significantly increased when compared to mice infected with the wild-type strain. Conclusion Our results indicated that the traC-traD operon is required for the invasion and intracellular growth abilities of L. dumoffii Tex-KL in epithelial cells.

The developmentof microelectromechanical systems hasresulted in the rapid development of polydimethylpolysiloxane(PDMS)microfluidic devices for drug screening *** cell functions,such as the response of endothelial cells to fluids,have beenelucdated using microuidic ***,organon-achip systems that includeorgans that are importantfor biologicalcirculation,such as the heart,liver,pancreas,kidneys,and brain,have been *** organs realize the biologicalcirculation system in a manner that cannot be reproduced by artificial organs;however,the flow channels between the organsare often artifically created by *** this study,we developeda microfluidic device consisting only of cels,by combiningcell sheet technology with microtitanium *** were placed between stacked fibroblast cellsheets,and the celisheets adhered to each other,afer which the microwires were removed leaving a luminal structure with a size approximatelyequal to the arteriolar *** lumen structure was constructed using wires with diameters of 50,100,150,and 200μm,which were approximations of the arteriole *** using a perfusion device,we successfully perfused theluminal structure created inside the *** results revealed that a aulture solution can be supplied toa cellsheet witha very high cell *** biofabrication technology proposed in this study can contribute to the development of organ-on-a-chip systems.

Electron Communication Between Electrode and Dye-Linked L-Proline Dehydrogenase in Organized Layer-by-Layer Multilayer Film

Multilayer film was fabricated on an electrode surface by alternate layer-by-layer（LBL） adsorption of polycationic redox polymer（PEI-Fc） and dye-linked L-proline dehydrogenase（L-proDH）.The electrochemistry of the PEI-Fc/L-proDH multilayer modified electrode was investigated by cyclic voltammetry,and the enzyme catalysis mediated by the redox polymer was studied in a solution containing *** was observed that electron communication between L-proDH and the electrode was achieved with the help of PEI-Fc.A mathematical expression for the current response was evaluated based on the Michaelis-Menten kinetics mode,and the calculated currents fitted well with the experimental *** kinetic analysis indicates that only a small fraction of the immobilized enzyme was efficiently electrically wired by the redox polymer.

Kinetics of thermal decomposition of benzene on lignite-derived char was investigated at 900℃ by applying a new method to continuously monitor the char surface *** vapor was continuously forced to pass through a micro fixed bed of char with residence time as short as 7.6 ms,and then detected continuously by a flame-ionization *** showed the presence of two different types of char surfaces;consumptive Type I surface and non-consumptive(sustainable)Type II *** I surface of a partially CO_(2)-gasified char had an capacity of carbon deposit from benzene over 20 wt%-char and an initial activity(represented by a first-order rate constant)as high as 160 s−*** of them decreased with increasing carbon deposit due to consumption of micropores accessible to benzene,and finally became zero leaving Type II surface that had a very stable activity with rate constant of 4 s−*** chars without gasification had capacities of Type I surfaces smaller by two orders of magnitude than the partially gasified char,while the Type II surfaces had activities similar to that of the partially gasified *** was found that Type II surface converted benzene into not only carbon deposit but also diaromatics and even greater *** of the greater aromatics was unknown because they were deposited onto the reactor wall immediately after passing through the char bed.

In recent years,researches on food components with anti-allergic effects have been gathering attention,because of the expectation for the establishment of a safe and effective treatment for food *** studies have reported that Lactiplantibacillus plantarum 22A-3 (LP22A3) inhibited degranulation of mast cells and reduced IgE *** have developed a gastrointestinal allergy system in which mice are sensitized by intraperitoneal and oral administration of OVA and then challenged by oral administration of high doses of *** a result,an increase in the amount of IgE in the blood and a decrease in the temperature of the colon were confirmed,and it was clarified that food allergy was induced by oral administration of high dose of OVA as the *** administration of LP22A3 ameliorated allergic responses significantly by reducing the amount of IgE in the blood and to recovered the decrease of rectal ***,LP22A3 did not affect the intestinal barrier *** LP22A3 significantly suppressed mRNA expression of OX40L and *** results suggested that LP22A3 suppressed Th2 differentiation and IL-4 production via downregulation of OX40L,and consequently suppressed IgE ***22A3 might provide a safe and effective treatment for allergic diseases due to ability modulating intestinal immune system.

Deep learning ferroelectric polarization distributions from STEM data via with and without atom finding

Over the last decade,scanning transmission electron microscopy(STEM)has emerged as a powerful tool for probing atomic structures of complex materials with picometer precision,opening the pathway toward exploring ferroelectric,ferroelastic,and chemical phenomena on the atomic *** to date extracting a polarization signal from lattice coupled distortions in STEM imaging rely on discovery of atomic positions from intensity maxima/minima and subsequent calculation of polarization and other order parameter fields from the atomic ***,we explore the feasibility of polarization mapping directly from the analysis of STEM images using deep convolutional neural networks(DCNNs).In this approach,the DCNN is trained on the labeled part of the image(i.e.,for human labelling),and the trained network is subsequently applied to other *** explore the effects of the choice of the descriptors(centered on atomic columns and grid-based),the effects of observational bias,and whether the network trained on one composition can be applied to a different *** analysis demonstrates the tremendous potential of the DCNN for the analysis of high-resolution STEM imaging and spectral data and highlights the associated limitations.

As an essential component of the Materials Genome Initiative aiming to shorten the period of materials research and development, combinatorial synthesis and rapid characterization technologies have been playing a more and more important role in exploring new materials and comprehensively understanding materials properties. In this review, we discuss the advantages of high-throughput experimental techniques in researches on superconductors. The evolution of combinatorial thin-film technology and several high-speed screening devices are briefly introduced. We emphasize the necessity to develop new high-throughput research modes such as a combination of high-throughput techniques and conventional methods.

Lamivudine treatment enabling right hepatectomy for hepatocellular carcinoma in decompensated cirrhosis

A 69-year-old man was admitted to our hospital in October 2003,for further examination of two liver *** was diagnosed with hepatocellular carcinoma(HCC) arising from decompensated hepatitis B virus(HBV)-related ***-term lamivudine administration improved liver function dramatically despite repeated treatment for *** Child-Pugh score was 9 points at start of lamivudine treatment,improving to 5 points after 1 *** indocyanine green at 15 min after injection test score was 48%before lamivudine treat-ment,improving to 22%after 2 years and to 5%after 4 *** ablation controlled the HCC foci and maintained his liver *** April 2009,abdominal computed tomography revealed a tumor thrombus in the right portal *** his indocyanine green test results had improved to less than 10%,we performed a right hepatectomy,which was *** our knowledge,there have been no documented reports of patients undergoing successful right hepatectomy for HCC arising from decompensated *** findings observed in our patient indicate the importance of nucleoside analogs for treating HBV-related HCC.
