能谱CT影像组学预测结直肠癌MSI状态的应用研究

目的研究基于能谱CT成像静脉期单能量序列影像组学模型,用于预测结直肠癌微卫星不稳定(MSI)状态。方法回顾性收集2019年7月至2022年8月在泰州市人民医院经手术切除或穿刺活检病理确诊为结直肠癌患者97例(男性57例、女性40例)。根据免疫组化MSI表达结果进行分组:MSI组34例、微卫星稳定(MSS)组63例。以DICOM格式将静脉期1.25mm原始图像数据传至GEAW4.7后处理工作站,生成碘基物质分解图、70keV、100keV单能量图,导入ITK-SNAP开源软件,手动勾画感兴趣区(ROI),利用Pyradiomics工具进行影像组学特征提取,采用mRMR(最大相关和最小冗余)和LASSO(最小绝对收缩选择算子)算法对训练组的影像组学特征进行降维,获取关键的影像组学特征,建立预测肿瘤生物学行为的影像组学模型。采用受试者工作特征(ROC)曲线下面积(AUC)来评估模型的诊断效能。使用校正曲线对模型的校正性能进行评估,应用决策曲线评价模型在训练组中的临床实用性。结果静脉期单能量70keV序列、100keV序列、碘基物质分解图共三种模型各提取了1218个特征,其中70keV单能量序列特征分类性能最佳。结论基于能谱CT成像的单能量模型可以有效地区分结直肠癌MSI和MSS状态患者,可以作为术前预测结直肠癌患者MSI状态的重要影像生物标志物,为指导临床医生制定个体化治疗方案及评估患者预后提供新思路。

目的应用磁共振成像中的动脉自旋标记(ASL)技术来探讨乳腺癌患者化疗后脑血流灌注变化及其与认知功能之间的相关性。方法前瞻性的收集43名自2020年1月至2022年12月经过手术病理证实的女性单侧乳腺癌患者(BC组),且所有患者在术后接受标准的化疗方案进行治疗。同时招募37名健康对照组(HC组),两组均采用西门子3.0T磁共振扫描仪进行常规头颅平扫及全脑ASL扫描,采用双样本t检验对各脑区脑血流量(CBF)值进行统计学分析。之后应用spearman相关性分析,探索存在血流灌注差异脑区的CBF值与神经量表之间的相关性。结果与HC组相比,BC组左侧小脑CrusII区、双侧颞中回CBF值降低,双侧豆状壳核CBF值增加(P<0.05,FWE校正)。相关性分析结果显示左侧小脑CrusII区、左侧颞中回、右侧颞中回CBF值与神经心理学量表MMSE呈正相关(r=0.429,P<0.01;r=0.413,P<0.01;r=0.405,P<0.01)。结论乳腺癌患者化疗后大脑存在多个脑区的脑血流灌注异常且存在与化疗相关的认知功能障碍。

Postponed and reduced basal nitrogen application improves nitrogen use efficiency and plant growth of winter wheat

Excessive nitrogen(N) fertilization with a high basal N ratio in wheat can result in lower N use efficiency(NUE) and has led to environmental problems in the Yangtze River Basin, China. However, wheat requires less N fertilizer at seedling growth stage, and its basal N fertilizer utilization efficiency is relatively low; therefore, reducing the N application rate at the seedling stage and postponing the N fertilization period may be effective for reducing N application and increasing wheat yield and NUE. A 4-year field experiment was conducted with two cultivars under four N rates(240 kg N ha–1(N240), 180 kg N ha–1(N180), 150 kg N ha–1(N150), and 0 kg N ha–1(N0)) and three basal N application stages(seeding(L0), fourleaf stage(L4), and six-leaf stage(L6)) to investigate the effects of reducing the basal N application rate and postponing the basal N fertilization period on grain yield, NUE, and N balance in a soil-wheat system. There was no significant difference in grain yield between the N180 L4 and N240 L0(control) treatments, and the maximum N recovery efficiency and N agronomy efficiency were observed in the N180 L4 treatment. Grain yield and NUE were the highest in the L4 treatment. The leaf area index, flag leaf photosynthesis rate, flag leaf nitrate reductase and glutamine synthase activities, dry matter accumulation, and N uptake post-jointing under N180 L4 did not differ significantly from those under N240 L0. Reduced N application decreased the inorganic N content in the 0–60-cm soil layer, and the inorganic N content of the L6 treatment was higher than those of the L0 and L4 treatments at the same N level. Surplus N was low under the reduced N rates and delayed basal N application treatments. Therefore, postponing and reducing basal N fertilization could maintain a high yield and improve NUE by improving the photosynthetic production capacity, promoting N uptake and assimilation, and reducing surplus N in soil-wheat systems.

To elucidate the response of oxidative metabolism,triggered by elevated ammonium(NH_(4)^(+))concentrations,on root growth of wheat seedlings,Yumai 49(NH_(4)^(+)-tolerant)and Lumai 15(NH_(4)^(+)-sensitive)cultivars were supplied with either 5.0 mmol L^(–1)NH_(4)^(+)-N(EAC)or 5.0 mmol L–1 NO_(3)–-N(CON)under hydroponic *** growth in both cultivars was significantly reduced under EAC,and the negative effect was greater in Lumai *** enhanced the activities of monodehydroascorbate reductase and dehydroascorbate reductase in the roots of both cultivars,while it decreased ascorbic acid(ASA)content and GDP-mannose pyrophosphorylase(GMPase)activity at the 12 th day after treatment in Lumai 15 by 62.0 and 71.4%;and in Yumai 49 by 38.8 and 62.2%,respectively,indicating that the regeneration of ASA was increased,but the biosynthesis of ASA was reduced under EAC ***,EAC increased DHA/ASA,reactive oxygen species(ROS),and malondialdehyde contents,as well as antioxidant enzyme activities in the roots of both *** greater increases in ROS and soluble sugar,and lower antioxidant enzyme activities in Lumai 15 indicate severe disruption of oxidative metabolism when compared to Yumai *** reveal that the reduction of ASA biosynthesis via decreased GMPase activity under the EAC condition probably acts as a trigger for accumulated ROS and imbalanced redox status,resulting in root growth inhibition during wheat seedling growth *** 49,being an NH_(4)^(+)-tolerant cultivar,had the stronger capacity to protect itself from oxidative stress,which allowed it to retain a lower DHA to ASA ratio by maintaining a better redox homeostasis than could be maintained in the NH_(4)^(+)-sensitive cultivar Lumai 15.

Microplastics(MPs)have become a significant concern for their potential ***,the correlation between the size of plastic particles and their toxicity remains ***,we investigate the toxic effects of different sizes(80 nm,800 nm,8μm and 80μm)polystyrene MPs(PS-MPs)on the model organism Nile tilapia(Oreochromis niloticus).The results of bioluminescent imaging indicate that the 80 nm PS-MPs are more likely to invade the body.H&E staining shows severe damage on the intestinal villi and distinct hepatic steatosis in the 80 nm *** U labeling shows that the proliferation activity of intestinal and liver cells reduces significantly in the 80 nm *** gut microbiome analysis shows a severe imbalance of gut microbiota homeostasis in the 80 nm *** analysis of liver transcriptomics and metabolomics shows that the liver lipid metabolism is disordered in the 80 nm *** conclusion,this study confirms that the 80 nm PS-MPs are more likely to induce intestinal and liver *** the above lay the foundation for further study on the pathological damage of MPs to other organisms.

How to effectively evaluate the firing precision of weapon equipment at low cost is one of the core contents of improving the test level of weapon system.A new method to evaluate the firing precision of the MLRS considering the credibility of simulation system based on Bayesian theory is proposed in this *** of all,a comprehensive index system for the credibility of the simulation system of the firing precision of the MLRS is constructed combined with the group analytic hierarchy process.A modified method for determining the comprehensive weight of the index is established to improve the rationality of the index weight *** Bayesian posterior estimation formula of firing precision considering prior information is derived in the form of mixed prior distribution,and the rationality of prior information used in estimation model is discussed *** the simulation tests,the different evaluation methods are compared to validate the effectiveness of the proposed ***,the experimental results show that the effectiveness of estimation method for firing precision is improved by more than 25%.

We study dark localized waves within a nonlinear system based on the Boussinesq approximation,describing the dynamics of shallow water *** symbolic calculus,we apply the Hirota bilinear method to transform an extended Boussinesq system into a bilinear form,and then use the multiple rogue wave method to obtain its dark rational *** the first-and second-order dark solutions,we examine the conditions under which these localized solutions exist and their spatiotemporal *** the selection of various parameters and by utilizing different visualization techniques(intensity distributions and contour plots),we explore the dynamical properties of dark solutions found:in particular,the first-and second-order dark rogue *** also explore the methods of their *** findings presented here not only deepen the understanding of physical phenomena described by the(1+1)-dimensional Boussinesq equation,but also expand avenues for further *** method can be extended to other nonlinear systems,to conceivably obtain higher-order dark rogue waves.
