The mRNA Expression and Methylation Status in Imprinting Control Region of H19 Gene Between Cattle-Yak and Their Parents

The H19 gene, which is imprinted with preferential expression from the maternal allele, was one of the first identified imprinting genes in mammals. Recent studies revealed that correct imprinting of the H19 gene plays a vital role in human spermatogenesis. To investigate whether imprinting defects were associated with the hybrid sterility of male cattle-yak, the methylation patterns of the H19 imprinting control region （ICR） and H19 mRNA expression in the testes of cattle-yak, yak, and cattle were examined. The results showed that the 3rd CCCTC-binding factor （CTCF） site of the H19 ICR was significantly hypomethylated in the testes of cattle-yak compared with yak or cattle. As expected, H19 was expressed at a significantly higher level in cattle-yak than in yak or cattle. These results suggest that imprinting defects of the CTCF- binding site in the HI9 ICR were possibly associated with disturbed spermatogenesis of male cattle-yak. Thus, we propose that disorders in H19 imprinting, resulting in an increased H19 mRNA expression, might contribute to the sterility of F1 male hybrids between cattle and yak.

Numerical simulation on gas–liquid multiphase flow behavior under coupling effects of annular gas curtain and swirling flow at tundish upper nozzle

A three-dimensional mathematical model has been established for a novel metallurgy process coupling an annular gas curtain with swirling flow at tundish upper nozzle. The discrete phase model and volume of fluid model were applied to simulate the gas–liquid multiphase flow behavior in tundish and nozzle. The effect of argon flow rate on the migration behavior of bubbles and interface behavior between steel and slag was also investigated. The presented results indicate that the novel coupling process can significantly change the flow pattern in the stream zone of a tundish, prolong the average residence time of liquid steel, and reduce the dead fraction. A complete annular gas curtain is formed around the stopper rod of tundish. Under the action of drag force of liquid steel, a part of small bubbles enter the nozzle through the swirling grooves and gather toward the center of the nozzle by centripetal force. As the argon flow rate increases, the volume fraction of argon gas entering the nozzle increases, which enhances the swirl intensity and increases the concentration of bubbles in the nozzle. To avoid the formation of slag open eye in tundish, the argon flow rate should not exceed 8 L min−1.

A water-soluble polysaccharide（named MCMP） was isolated from the mycelium with high yield mutation Cordyceps militaris by hot-water extraction, deproteinization by sevage, alcohol precipitation, anion-exchange and gel filtration chromatography CL-6B. The polysaccharide contained mannose, rhamnose, galactose and glucose in a molar ratio of 59.36：1：8.31：39.50, of which the average molecular weight is 8100. In our research, Hep-G2 cells, Hela cells and mesangial cells were chosen to determine the anti-tumor activity of the polysaccharide. The results of MTT assay show that polysaccharides of the mutant strain presented inhibitory activity on the cells proliferation after 48 h incubation.

AIM: To discuss the long-term postoperative results of bilateral lateral rectus recession(BLR) and unilateral lateral rectus recession-medial rectus resection(RR) in therapy of intermittent ***: We retrospectively analyzed 213 cases of intermittent exotropia who underwent surgery between2008 and 2010. The patients were grouped into BLR group and RR group. Motor outcomes were divided into three groups on the basis of the angle of deviation after surgery: overcorrection(esotropia/phoria >5△), orthophoria(esotropia/phoria ≤5△to exotropia/phoria ≤10△), and undercorrection/recurrence(exotropia/phoria >10△).Titmus test was used to evaluate stereoacuity, the stereoacuity 0.05) between ***, the motor outcomes at 6, 36 mo were signally different in each group, indicating the success rate in RR group at 6mo was higher than that in BLR group(83.02%vs 82.24%, P <0.05) but the result was contrary at the 3y examination(60.75% vs 43.40%, P <0.05). No statistical significance were found in the sensory outcomes between the groups at mean of 3.7y ***: The motor outcomes in RR group were better than in BLR group at 6mo after surgery, while the3 y outcomes were better in BLR group. This may be due to the recurrence rate of the BLR was lower than the RR group’s.

Aluminum-doped zinc oxide （ZnO：AI） films were deposited by direct current magnetron sputtering in incorporating hydrogen in sputtering gas at room temper- ature. The influences of hydrogen content in sputtering gas on the structural, optical, and electrical properties of ZnO：A1 films were systematically investigated. It is found that hydrogen incorporated into ZnO lattice forms shallow donors in ZnO：A1 films and plays an important role in the properties of ZnO：A1 films. The electrical conductivity and infrared （IR） reflectance are improved due to the increase of electron carrier concentration, and the average trans- mittance decreases, which is ascribed to the strong scat- tering from the hydrogen incorporated and oxygen vacancies in ZnO：A1 films. In this study, the resistivity of 5.5 × 10-4 Ω.cm is obtained, the average transmittance of the wavelength in the range of 400-900 nm is almost 86 %, and the IR reflectance reaches 75 % at 2,500 nm, which is higher than that of reported TCO films. The band gap determined by optical absorption is a result of com- petition between Burstein-Moss effect and many-body perturbation effect. However, the hydrogen content in sputtering gas is above 10 %, and the optical band gap shift is independent of hydrogen content in sputtering gas.
