Metabolic profiling of endogenous bile acids:a novel method to assess hepatoprotective effect of Tanreqing capsule on carbon-tetrachloride-induced liver injury in rats

Tanreqing(TRQ), a traditional Chinese medicine(TCM) formula, can alleviate liver injury and improve liver function. Its pharmacological mechanisms of actions are still unclear due to its complex components and multi-target natures. Metabolomic study is an effective approach to investigating drug pharmacological actions, new diagnostic markers, and potential mechanisms of actions. In the present study, a new strategy was used to evaluate the protective effect of TRQ capsule against carbon tetrachloride(CCl_4)-induced hepatotoxicity in rats, by analyzing metabolic profiling of endogenous bile acids(BAs) along with biochemical and histological analyses. BAs concentrations were determined by ultra-performance liquid chromatography coupled with quadrupole mass spectrometry(UPLC-MS). Principal component analysis and partial least squares discriminant analysis were then employed to analyze the UPLC-MS results and compare the hepatoprotective effect of TRQ capsule in different groups at the doses of 0.36, 1.44, and 2.88 g·kg^(-1) body weight, respectively. Moreover, our results suggested that taurocholic acid(TCA) and taurohyodesoxycholic acid(THDCA) were the most important biochemical markers, which were indicative of CCl_4-induced acute hepatic damage and hepatoprotective effect of TRQ capsule. Therefore, this new strategy would be an excellent alternative method for evaluating hepatoprotective effect and proposing potential mechanisms of action for other drugs as well.

Salvianolate Reduces Glucose Metabolism Disorders in Dimethylnitrosamine-Induced Cirrhotic Rats

Objective： To evaluate the preventive effect of salvianolate （Sal B） on glucose metabolism disorders of dimethylnitrosamine （DMN）-induced cirrhotic rats. Methods： Fiffy-five Wistar rats were randomly divided into a control group （n=10） and a cirrhotic group （n=45） according to a random number table. Liver cirrhosis was induced by intraperitoneal administration of DMN. The cirrhotic rats were divided into model, Sal B and metformin groups （n=15）, respectively. Rats in the model group were given saline, two treatment groups were given Sal B （50 mg/kg）, metformin （150 mg/kg） respectively for 28 consecutive days, while rats in the control group were injected 0.9% saline with same volume of vehicle. Body weight was measured everyday. Insulin sensitivity was determined by euglycemic hyperinsulinemic clamp. Organ index, glucose tolerance test （OGTT）, and fasting plasma glucose （FPG）, fasting insulin （FINS）, hepatic glycogen, hydroxyproline （HYP） and liver function were detected at the end of the treatment. Area under the curve （AUC） for OG3-1 was calculated. Liver and pancreas histology were determined by histopathological examination with hematoxylin and eosin staining （HE）, Sirius Red staining and Masson＇s trichrome staining, respectively. Hepatic expression of α-smooth muscle actin （oL-SMA） and collagen （Col I ） were evaluated by immunohistochemical staining. Results： Compared with the model group, Sal B significantly increased body and liver weight, liver-body ratio, glucose infusion rate （GIR）, FPG, FINS levels and hepatic glycogen at the end of administration （P〈0.05 or P〈0.01）. Meanwhile, Sal B significantly decreased AUC for OGI-I＇, spleen weight, spleen-body ratio, aminotransferase and HYP level （P〈0.05 or P〈0.01）. Sal B was also effective in alleviating necrosis of liver tissue, suppressing fibrosis progression and inhibiting the expression of oL-SMA and Col I in liver. Fuzheng Huayu Formula(扶正化瘀方) Prevents Rat Renal Interstitial Fibrosis Induced by HgCl2 via Antioxidative Stress and Down-regulation of Nuclear Factor-Kappa B Activity

Objective：To investigate the mechanism of action of Fuzheng Huayu Formula（扶正化瘀方,FZHY）against renal interstitial fibrosis（RIF）relating to oxidative injury and nuclear factor-kappa B（NF-κB）activity.Methods：Thirty-two Sprague-Dawley rats were randomly divided into 3 groups：normal group,model group and FZHY treatment group.The RIF model was induced by oral administration of HgC l2 at a dose of 8 mg/kg body weight once a day for 9 weeks.Meanwhile,rats in FZHY treatment group orally took FZHY at a dose of4.0 g/kg rat weight for 9 weeks.The content of hydroxyproline（Hyp）and collagen deposition in kidney were observed.The activities of superoxide dismutase（SOD）and glutathione peroxidase（GSH-Px）,the content of glutathione（GSH）and malondialdehyde（MDA）of kidney were tested.The expressions of inhibitor-κappa B（IκB）,phospho-IκB（p-IκB）,tumor necrosis factor-α（TNF-α）,matrix metalloproteinase-2（MMP-2）andα-smooth muscle actin（α-SMA）were analyzed by Western blot.α-SMA expression was also observed by immunofluorescent staining.MMP-2 activity was measured by gelatin zymography.NF-κB activation was determined by electrophoretic mobility shift assay.Results：Renal interstitial fibrosis was induced by Hg Cl2,demonstrated by remarkably increased Hyp contents and excessive collagen deposition in kidney（P〈0.01）.FZHY significantly inhibited renal interstitial collagen deposition and reduced Hyp content of the Hg Cl2-treated rats（P〈0.01）.GSH content decreased obviously,and MDA content increased significantly in HgC l2-treated rats compared with that of normal rats（P〈0.01）.FZHY significantly increased GSH content and decreased MDA content in the model rats（P〈0.01）.The expressionα-SMA was increased in model rats compared with that of normal rats,FZHY significantly decreased its expression（P〈0.01）.The expressions of p-IκB and TNF-αand MMP-2,MMP-2 activity,and NF-κB activation were increased in model group compared with that in normal group（P〈0.01）,FZHY significantly decreased NF-κB activation,MMP-2 activity and p-IκB and TNF-αexpressions（P〈0.01

目的探究脓毒症合并心肌损伤患者血清组织纤溶酶原激活物-纤溶酶原激活物抑剂-1复合物(t-PAI-C)、肝素结合蛋白(HBP)、外周血高迁移率组蛋白B1(HMGB1)水平与其预后的关系。方法回顾性分析2020年3月至2023年3月蚌埠医学院第一附属医院收治的105例脓毒症合并心肌损伤患者的临床资料,依据患者治疗后28 d存活情况将其分为存活组与死亡组。比较两组临床资料(性别、年龄、体重指数、感染部位、平均动脉压、射血分数)、血清心肌肌钙蛋白I(cTnI)、t-PAI-C、HBP、HMGB1水平以及急性生理学和慢性健康状况评价Ⅱ(APACHEⅡ)评分,分析影响脓毒症合并心肌损伤患者预后的影响因素;探究t-PAI-C、HBP、HMGB1水平与cTnI、APACHEⅡ评分的相关性;绘制受试者工作特征(ROC)曲线分析t-PAI-C、HBP、HMGB1诊断脓毒症合并心肌损伤患者预后的价值。结果脓毒症合并心肌损伤患者随访期间出现死亡30例(28.57%),存活75例(71.43%)。死亡组患者的cTnI、t-PAI-C、HBP、HMGB1水平以及APACHEⅡ评分分别为(1.58±0.43)μg/L、(16.75±4.00)ng/mL、(45.68±9.25)ng/mL、(125.00±20.18)μg/L、(17.63±2.66)分,均高于存活组[(0.65±0.11)μg/L、(13.20±2.68)ng/mL、(38.00±8.63)ng/mL、(96.69±11.25)μg/L、(11.50±1.68)分],差异均有统计学意义(P<0.05)。cTnI、t-PAI-C、HBP、HMGB1以及APACHEⅡ评分均为影响脓毒症合并心肌损伤患者预后的独立危险因素(P<0.05)。t-PAI-C、HBP、HMGB1与cTnI、APACHEⅡ评分之间均呈正相关(P<0.05)。t-PAI-C、HBP、HMGB1三者联合诊断脓毒症合并心肌损伤患者预后的曲线下面积(AUC)为0.950(0.889~0.983),敏感度与特异度分别为83.33%和93.33%,诊断效能均优于单一的t-PAI-C、HBP、HMGB1指标(P<0.05)。结论 t-PAI-C、HBP、HMGB1水平与cTnI、APACHEⅡ评分相关性较好,可作为临床诊断脓毒症合并心肌损伤的潜在生物学标记物,三者联合预测脓毒症合并心肌损伤患者预后效能较好。

Dear Editor,This letter focuses on the distributed optimal containment control of continuous-time multi-agent systems(CTMASs)with respect to the minimum-energy performance index over fixed topology.To achieve this,we firstly investigate the optimal containment control problem using the inverse optimal control method,where all states of followers asymptotically converge to the convex hull spanned by the leaders while some quadratic performance indexes get minimized.A sufficient condition for existence of the distributed optimal containment control protocol is derived.By introducing the parametric algebraic Riccati equation(PARE),it is strictly proved that the global performance index can be used to approximate the standard minimumenergy performance index as the parameters tends to infinity.In consequence,the standard minimum-energy cooperative containment control can be solved by local steady state feedback protocols.

Resilient performance of self-centering hybrid rocking walls with curved interface under pseudo-static loading

Frame and rocking wall(FRW)structures have excellent resilient performance during earthquakes.However,the concrete at interfacial corners of rocking walls(RWs)is easily crushed due to local extreme compression during the rocking process.An innovative RW with a curved interface is proposed to prevent interfacial corners from producing local damage,enhancing its earthquake resilient performance(ERP).The precast wall panel with a curved interface is assembled into an integral self-centering hybrid rocking wall(SCRW)by two post-tensioned unbonded prestressed tendons.Moreover,two ordinary energy dissipation steel rebars and two shear reinforcements are arranged to increase the energy dissipation capacity and lateral resistance.Two SCRW specimens and one monolithic reinforced concrete(RC)shear wall(SW)were tested under pseudo-static loading to compare the ERPs of the proposed SCRW and the SW,focusing on studying the effect of the curved interface on the SCRW.The key resilient performance of rocking effects,failure modes,and hysteretic properties of the SCRW were explored.The results show that nonlinear deformations of the SCRW are concentrated along the interface between the SCRW and the foundation,avoiding damage within the SCRW.The restoring force provided by the prestressed tendons can effectively realize self-centering capacity with small residual deformation,and the resilient performance of the SCRW is better than that of monolithic SW.In addition,the curved interface of the SCRW makes the rocking center change and move inward,partially relieving the stress concentration and crush of concrete.The rocking range of the rocking center is about 41.4%of the width of the SCRW.

In this paper,we consider the clustering of bivariate functional data where each random surface consists of a set of curves recorded repeatedly for each subject.The k-centres surface clustering method based on marginal functional principal component analysis is proposed for the bivariate functional data,and a novel clustering criterion is presented where both the random surface and its partial derivative function in two directions are considered.In addition,we also consider two other clustering methods,k-centres surface clustering methods based on product functional principal component analysis or double functional principal component analysis.Simulation results indicate that the proposed methods have a nice performance in terms of both the correct classification rate and the adjusted rand index.The approaches are further illustrated through empirical analysis of human mortality data.

Xuebijing improves intestinal microcirculation dysfunction in septic rats by regulating the VEGF-A/PI3K/Akt signaling pathway

BACKGROUND:This study aims to explore whether Xuebijing(XBJ) can improve intestinal microcirculation dysfunction in sepsis and its mechanism.METHODS:A rat model of sepsis was established by cecal ligation and puncture(CLP).A total of 30 male SD rats were divided into four groups:sham group,CLP group,XBJ + axitinib group,and XBJ group.XBJ was intraperitoneally injected 2 h before CLP.Hemodynamic data(blood pressure and heart rate) were recorded.The intestinal microcirculation data of the rats were analyzed via microcirculation imaging.Enzyme-linked immunosorbent assay(ELISA) kits were used to detect the serum levels of interleukin-6(IL-6),C-reactive protein(CRP),and tumor necrosis factor-α(TNF-α) in the rats.Histological analysis and transmission electron microscopy were used to analyze the injury of small intestinal microvascular endothelial cells and small intestinal mucosa in rats.The expression of vascular endothelial growth factor A(VEGF-A),phosphoinositide 3-kinase(PI3K),phosphorylated PI3K(p-PI3K),protein kinase B(Akt),and phosphorylated Akt(p-Akt) in the small intestine was analyzed via Western blotting.RESULTS:XBJ improved intestinal microcirculation dysfunction in septic rats,alleviated the injury of small intestinal microvascular endothelial cells and small intestinal mucosa,and reduced the systemic inflammatory response.Moreover,XBJ upregulated the expression of VEGF-A,p-PI3K/total PI3K,and p-Akt/total Akt in the rat small intestine.CONCLUSION:XBJ may improve intestinal microcirculation dysfunction in septic rats possibly through the VEGF-A/PI3K/Akt signaling pathway.

A survey of energy-efficient strategies for federated learning in mobile edge computing

With the booming development of fifth-generation network technology and Internet of Things,the number of end-user devices(EDs)and diverse applications is surging,resulting in massive data generated at the edge of networks.To process these data eficiently,the innovative mobile edge computing(MEC)framework has emerged to guarantee low latency and enable eficient computing close to the user traffic.Recently,federated learning(FL)has demonstrated its empirical success in edge computing due to its privacy-preserving advantages.Thus,it becomes a promising solution for analyzing and processing distributed data on EDs in various machine learning tasks,which are the major workloads in MEC.Unfortunately,EDs are typically powered by batteries with limited capacity,which brings challenges when performing energy-intensive FL tasks.To address these challenges,many strategies have been proposed to save energy in FL.Considering the absence of a survey that thoroughly summarizes and classifies these strategies,in this paper,we provide a comprehensive survey of recent advances in energy-efficient strategies for FL in MEC.Specifically,we first introduce the system model and energy consumption models in FL,in terms of computation and communication.Then we analyze the challenges regarding improving energy efficiency and summarize the energy-efficient strategies from three perspectives:learning-based,resource allocation,and client selection.We conduct a detailed analysis of these strategies,comparing their advantages and disadvantages.Additionally,we visually illustrate the impact of these strategies on the performance of FL by showcasing experimental results.Finally,several potential future research directions for energy-efficient FL are discussed.

Game-theoretical Model for Dynamic Defense Resource Allocation in Cyber-physical Power Systems Under Distributed Denial of Service Attacks

Electric power grids are evolving into complex cyber-physical power systems(CPPSs)that integrate advanced information and communication technologies(ICTs)but face increasing cyberspace threats and attacks.This study considers CPPS cyberspace security under distributed denial of service(DDoS)attacks and proposes a nonzero-sum game-theoretical model with incomplete information for appropriate allocation of defense resources based on the availability of limited resources.Task time delay is applied to quantify the expected utility as CPPSs have high time requirements and incur massive damage DDoS attacks.Different resource allocation strategies are adopted by attackers and defenders under the three cases of attack-free,failed attack,and successful attack,which lead to a corresponding consumption of resources.A multidimensional node value analysis is designed to introduce physical and cybersecurity indices.Simulation experiments and numerical results demonstrate the effectiveness of the proposed model for the appropriate allocation of defense resources in CPPSs under limited resource availability.
