Comparative pharmacodynamic, pharmacokinetic and tissue distribution of Dahuang-Gancao decoction in normal and experimental constipation mice

Dahuang-Gancao decoction(DGD)is a classical formula,which is commonly used for reliving constipation in Chinese *** aim of this study was to investigate the pharmacodynamic,pharmacokinetic and tissue distribution alternations of DGD in normal and constipation *** exhibited stronger purgative effect in constipation mice by the increased fecal excretion and reduced first defection time compared with normal *** Cmax,AUC0-t and MRT0-t of rhein,aloe-emodin,rhein-8-O-β-D-glucoside,sennoside A,and glycyrrhizic acid as main bio-active components in DGD were markedly increased in constipation *** tissue distribution of the analytes in constipation mice were higher than those in normal mice with rhein>rhein-8-O-β-D-glucoside>aloe-emodin>glycyrrhizic acid>emodin in liver,and glycyrrhizic acid>rhein-8-O-β-D-glucoside>liquitin>sennoside A>rhein>aloe-emodin>emodin in *** kidney concentrations of the analytes showed a descending order of rhein>rhein-8-O-β-D-glucoside>sennoside A>glycyrrhizic acid>aloe-emodin>emodin,most of them were higher while rhein was lower in constipation mice than normal *** higher exposure of the anthraquinones in plasma,liver and colon may result in the stronger purgative effect in the constipation mice than normal *** is mainly excreted through the kidney,the decreased level of rhein in constipation mice may explain the alleviated side *** of glycyrrhizic acid in colon may related with the moderate property of *** results provided the experimental basis for understanding the therapeutic effects and metabolite profile of DGD.

Diabetic kidney disease(DKD)is the primary cause of mortality among diabetic *** the increasing prevalence of diabetes,it has become a major concern around the *** therapeutic effect of clinical use of drugs is far from expected,and therapy choices to slow the progression of DKD remain ***,research on new drugs and treatments for DKD has been a hot topic in the medical *** has been found that rhein has the potential to target the pathogenesis of DKD and has a wide range of pharmacological effects on DKD,such as anti-nephritis,decreasing blood glucose,controlling blood lipids and renal *** recent years,the medical value of rhein in the treatment of diabetes,DKD and renal disease has gradually attracted worldwide attention,especially its potential in the treatment of ***,DKD can only be treated with medications from a single symptom and are accompanied by adverse effects,while rhein improves DKD with a multi-pathway and multi-target ***,this paper reviews the therapeutic effects of rhein on DKD,and proposes solutions to the limitations of rhein itself,in order to provide valuable references for the clinical application of rhein in DKD and the development of new drugs.

Efficacy and Safety of Lianhua Qingwen for Patients with COVID-19: A Systematic Review and Meta-Analysis

Background: Corona virus disease 2019(COVID-19) has spread around the world since its outbreak, and there is no ascertained effective drug up to now. Lianhua Qingwen(LHQW) has been widely used in China and overseas Chinese, which had some advantages in the treatment of COVID-19. Objective: To evaluate the efficacy and safety of LHQW for COVID-19 by conducting a systematic review with meta-analysis. Methods: A comprehensive literature search was conducted in 12 electronic databases from their establishment to October 30, 2021. Note Express 3.2.0 was used for screening of trials, and the data was independently extracted in duplicate by 2 researchers. The risk of bias of randomized controlled trials(RCTs) and retrospective studies were assessed by using the Cochrane collaboration tool and Newcastle Ottawa Scale, respectively, followed by data analysis using RevMan 5.3. The RCTs or retrospective studies to treat COVID-19 using LHQW were included. The intervention measures in the experimental group were LHQW alone or combined with chemical drugs(LCWC), and that in the control group were chemical drugs(CDs). Outcome measures included computed tomography(CT) recovery rate, disappearance rates of primary(fever, cough, fatigue), respiratory, gastrointestinal and other symptoms, exacerbation rate and adverse reaction. Subgroup analysis was conducted according to whether LHQW was combined with CDs and the different treatment methods in the control group. Results: Nine trials with 1,152 participants with COVID-19 were included. The CT recovery rates of LHQW and LCWC were 1.36 and 1.32 times of CDs, respectively(P0.05). Conclusions: LHQW was more

Effects of ferulic acid on antioxidant activity in Angelicae Sinensis Radix, Chuanxiong Rhizoma, and their combination

The present study aimed at exploring different roles of the same compound in different environment, using preparative HPLC, and the significance to investigating bio-active constituents in traditional Chinese medicine （TCM） on the basis of holism. In this study, the depletion of target component ferulic acid （FA） by using preparative HPLC followed by antioxidant activity testing was applied to investigate the roles of FA in Angelicae Sinansis Radix （DG）, Chuanxiong Rhizoma （CX） and their combination （GX）. The antioxidant activity was performed by 1, 1-diphenyl-2-picrylhydrazyl （DPPH） radical-scavenging activity testing. FA was successfully and exclusively depleted from DG, CX, and GX, respectively. By comparing the effects of the samples, it was found that FA was one of the main antioxidant constituents in DG, CX and GX, and the roles of FA were DG 〉 CX 〉 GX. Furthermore, the effects of FA varied at different doses in these herbs. This study provided a reliable and effective approach to clarifying the contribution of same compound in different TCMs to their bio-activities. The role of a constituent in different TCMs might be different, and a component with the same content might have different effects in different chemical environments. Furthermore, this study also suggested the potential utilization of preparative HPLC in the characterization of the roles of multi-ingredients in TCM.

Anti-obesity and Gut Microbiota Modulation Effect of Astragalus Polysaccharides Combined with Berberine on High-Fat Diet-Fed Obese Mice

Objective To investigate whether astragalus polysaccharides(APS)combined with berberine(BBR)can reduce high-fat diet(HFD)-induced obesity in *** Except for normal mice,32 HFD-induced obese mice were randomized into HFD,APS(1,000 mg/kg APS),BBR(200 mg/kg BBR),and APS plus BBR(1,000 mg/kg APS plus 200 mg/kg BBR)groups,*** 6-week treatment(once daily by gavage),the obesity phenotype and pharmacodynamic effects were evaluated by histopathological examination of epididymal fat,liver,and colon using hematoxylin-eosin staining and serum biochemical analyses by an automated chemistry *** feces were collected at the 12 th week,and taxonomic and functional profiles of gut microbiota were analyzed by 16S ribosomal ribonucleic acid(16S rRNA)*** Compared with HFD group,the average body weight of APS plus BBR group was decreased(P<0.01),accompanied with the reduced fat accumulation,enhanced colonic integrity,insulin sensitivity and glucose homeostasis(P<0.05 or P<0.01).Importantly,APS combined with BBR treatment was more effective than APS or BBR alone in improving HFD-induced insulin resistance(P<0.05 or P<0.01).16S rRNA sequence-based analysis of fecal samples demonstrated that APS combined with BBR treatment exhibited a better impact on HFD-induced gut microbiota dysbiosis,exclusively via the enriched abundances of Bacteroides,which corresponded to the large increase of predicted bacterial genes involved in carbohydrate *** APS combined with BBR may synergistically reduce obesity and modulate the gut microbiota in HFD-fed mice.

Quercetin-3-O-β-D-glucopyranosyl-(4→1)-α-L-rhamnoside metabolites in the rat using UPLC-Q-TOF/MS

Ultraperformance liquid chromatography/quadrupole-time-of-flight mass spectrometry(UPLC-Q-TOF/MS) and the MetabolynxTM software, combined with mass defect filtering, were applied to identity the metabolites of quercetin-3-O-β-D-glucopyranosyl-(4→1)-α-L-rhamnoside(QGR) in rats after intravenous administration. MSE was used for simultaneous acquisition of precursor ion information and fragment ion data at high and low collision energy in one analytical run, which facilitated the rapid structural characterization of eight metabolites in rat plasma, urine and bile. The results indicated that methylation and glucuronidation were the major metabolic pathways of QGR in vivo. The present study provided important information about the metabolism of QGR which will be useful for fully understanding the mechanism of action of this compound. Furthermore, this work demonstrated the potential of the UPLC-Q-TOF/MS approach using Metabolynx for rapid and automated research of the metabolites of natural products.

Ds-echinoside A,a new triterpene glycoside derived from sea cucumber,exhibits antimetastatic activity via the inhibition of NF-κB-dependent MMP-9 and VEGF expressions

Ds-echinoside A (DSEA),a non-sulfated triterpene glycoside,was isolated from the sea cucumber Pearsonothuria *** vitro and in vivo investigations were conducted on the effects of DSEA on tumor cell adhesion,migration,invasion,and *** this study,we found that DSEA inhibited the proliferation of human hepatocellular liver carcinoma cells Hep G2,with a half-maximal inhibitory concentration (IC50) of 2.65 μmol/L,and suppressed Hep G2 cell adhesion,migration,and invasion in a dose-dependent *** also reduced tube formation of human endothelial cells ECV-304 on matrigel in vitro and attenuated neovascularization in the chick embryo chorioallantoic membrane (CAM) assay in *** analysis revealed that DSEA significantly decreased the expression of matrix metalloproteinase-9 (MMP-9),which plays an important role in the degradation of basement membrane in tumor metastasis and *** also increased the protein expression level of tissue inhibitor of metalloproteinase-1 (TIMP-1),an important regulator of MMP-9 *** the results of Western blotting,the expressions of nuclear factor-kappa B (NF-κB) and vascular endothelial growth factor (VEGF) were found to be remarkably reduced by *** findings suggest that DSEA exhibits a significant antimetastatic activity through the specific inhibition of NF-κB-dependent MMP-9 and VEGF expressions.
