Dexamethasone implant for refractory macular edema secondary to diabetic retinopathy and retinal vein occlusion

AIM:To evaluate the efficacy,timing of retreatment and safety of dexamethasone(DEX)implant on macular edema(ME)secondary to diabetic retinopathy(DME)and retinal vein occlusion(RVO-ME)patients who were refractory to anti-vascular endothelial growth factor(VEGF)***:This retrospective study included 37 eyes received at least one DEX implant treatment for DME or RVO-ME between January 1,2019,and January 1,*** refractory DME and RVO-ME cases received at least 5 anti-VEGF injections and failure to gain more than 5 letters or a significant reduction in central retinal thickness(CRT).The best corrected visual acuity(BCVA)and CRT were measured at baseline,and at 1,3,4 and 6mo post-DEX implant *** events such as elevated intraocular pressure(IOP)and cataract were ***:For RVO cases(n=22),there was a significant increase in BCVA from 0.27±0.19 to 0.35±0.20 at 6mo post-DEX injection(P<0.05)and CRT decreased from 472.1±90.6 to 240.5±39.0μm at 6mo(P<0.0001).DME cases(n=15)experienced an improvement in BCVA from 0.26±0.15 to 0.43±0.20 at 6mo post-DEX implant injection(P=0.0098),with CRT reducing from 445.7±55.7 to 271.7±34.1μm at 6mo(P<0.0001).Elevated IOP occurred in 45.9%of patients but was well-controlled with topical *** cases of cataract or other adverse events were ***:DEX implants effectively improve BCVA and reduce CRT in refractory DME and *** research with larger cohorts and longer follow-up periods is needed to confirm these findings and assess long-term outcomes.

Biotransformation of ethylene glycol by engineered Escherichia coli

There has been extensive research on the biological recycling of PET waste to address the issue of plastic waste pollution,with ethylene glycol(EG)being one of the main components recovered from this ***,finding ways to convert PET monomer EG into high-value products is crucial for effective PET waste *** this study,we successfully engineered Escherichia coli to utilize EG and produce glycolic acid(GA),expecting to facilitate the biological recycling of PET *** engineered ***,able to utilize 10 g/L EG to produce 1.38 g/L GA within 96 h,was initially ***,strategies based on overexpression of key enzymes and knock-out of the competing pathways are employed to enhance EG utilization along with GA *** engineered ***,characterized by the highest GA production titer and substrate conversion rate,was *** GA titer increased to 5.1 g/L with a yield of 0.75 g/g EG,which is the highest level in the shake flake *** level analysis and metabolomic analysis were then conducted,revealing that overexpression of key enzymes and knock-out of the competing pathways improved the metabolic flow in the EG *** improved metabolic flow also leads to accelerated synthesis and metabolism of amino acids.

P-and SV-wave dispersion and attenuation in saturated microcracked porous rock with aligned penny-shaped fractures

P-and SV-wave dispersion and attenuation have been extensively investigated in saturated poroelastic media with aligned ***,there are few existing models that incorporate the multiple wave attenuation mechanisms from the microscopic scale to the macroscopic ***,in this work,we developed a unified model to incorporate the wave attenuation mechanisms at different scales,which includes the microscopic squirt flow between the microcracks and pores,the mesoscopic wave-induced fluid flow between fractures and background(FB-WIFF),and the macroscopic Biot's global flow and elastic scattering(ES)from the *** tang's modified Biot's theory and the mixed-boundary conditions,we derived the exact frequency-dependent solutions of the scattering problem for a single penny-shaped fracture with oblique incident P-and *** then developed theoretical models for a set of aligned fractures and randomly oriented fractures using the Foldy *** results indicated that microcrack squirt flow considerably influences the dispersion and attenuation of P-and SV-wave *** coupling effects of microcrack squirt flow with the FB-WIFF and ES of fractures cause much higher velocity dispersion and attenuation for P waves than for SV *** oriented fractures substantially reduce the attenuation caused by the FB-WIFF and ES,particularly for the ES attenuation of SV *** a comparison with existing models in the limiting cases and previous experimental measurements,we validated our model.

Removal of pathogenic indicator microorganisms during partial nitrification:the role of free nitrous acid

Digested wastewater contains pathogenic microorganisms and high ammonia concentrations,which can pose a potential risk to public *** removal of pathogens and nitrogen is crucial for the post-treatment of digested *** nitrification-anammox is an energy-saving nitrogen removal *** nitrous acid(FNA),an intermediate product of partial nitrification,has the potential to inactivate ***,the efficiency and mechanisms of FNA-related inactivation in pathogens during partial nitrification remains *** this study,Enterococcus and Escherichia coli(***)were selected to investigate the efficiency and mechanisms of FNA-related inactivation in partial nitrification *** results revealed that 83%±13%and 59%±27%of *** and Enterococcus were removed,respectively,in partial nitrification process at FNA concentrations of 0.023−0.028 mg/*** the concentration of FNA increased from 0 to 0.5 mg/L,the inactivation efficiencies of *** and Enterococcus increased from 0 to 99.9%and 89.9%,*** exhibited a higher resistance to FNA attack compared to ***.3D-laser scanning microscopy(3D-LSM)and scanning electron microscopy(SEM)revealed that FNA exposure caused the surface collapse of *** and Enterococcus,as well as visible pore formation on the surface of *** cells.4',6-Diamidino-2-phenylindole dihydrochloride n-hydrate(DAPI)/propidium iodide(PI)and biomolecule leakage confirmed that inactivation of *** and Enterococcus occurred due to breakdown of cell walls and cell *** findings indicate that partial nitrification process can be used for the removal of residual pathogenic microorganisms.

Enamel matrix proteins in promoting saliva lubrication

Anti-wear performance of human enamel in the mouth is closely related to the lubrication of salivary *** is well known that the inorganic hydroxyapatite(HA)of the enamel plays an important role in the adsorption and pellicle-forming of salivary proteins on the enamel,but the role of enamel matrix proteins remains *** this study,the adsorption and lubrication behavior of salivary proteins on original,heated,and deproteinated enamel surfaces was comparatively investigated using an atomic force microscopy and nano-indentation/scratch *** with that on the original enamel surface,the adsorption and lubrication behavior of salivary proteins remains almost unchanged on the heated enamel surface(where the enamel matrix proteins are denatured but the size of HA crystalline nanoparticles keeps constant)but exhibits an obvious compromise on the deproteinated enamel surface(where the enamel matrix proteins are removed and agglomeration of HA crystallites occurs).The HA agglomeration weakens the electrostatic interaction of enamel surfaces with salivary proteins to cause a distinct negative influence on the adsorption and pellicle-forming of salivary ***,the negative effect is confirmed with a quartz crystal microbalance with *** summary,by regulating enamel nanostructure for appropriate electrostatic interactions between salivary proteins and enamel surfaces,the enamel matrix proteins play an essential role in the adsorption and pellicle-forming of salivary proteins on human enamel,and then contribute to saliva lubrication,which provides the enamel with an anti-wear *** findings will promote and assist the design of enamel-inspired anti-wear materials.

Network Pharmacology and Molecular Docking Analysis of the Mechanisms of Combined Radix Bupleuri(Chai-Hu) and Radix Paeoniae Alba(Bai-Shao) Treatment in the Prevention and Treatment of Alzheimer's Disease

Objective: Radix Bupleuri and Radix Paeoniae Alba are referred to as Chai-Hu and Bai-Shao, respectively, in Chinese. We used molecular docking and network pharmacological approaches to explore the active components of this Chinese herb combination which is commonly used for treating Alzheimer's disease(AD), the Chai-Hu and Bai-Shao pair(CBP). Materials and Methods: The active compounds and action targets of Chai-Hu and Bai-Shao were identified in the Traditional Chinese Medicine Systems Pharmacology database, and the Therapeutic Target Database, Drug Bank, Gen Cards, Dis Ge NET, and Online Mendelian Inheritance in Man databases were used to identify therapeutic targets in patients with AD. To identify a common target of CBP in the treatment of AD, we used the Metascape tool to perform Gene Ontology function enrichment and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analyses. In addition, CBP-AD protein–protein interaction(PPI) and herb-component-target networks were created using Cytoscape 3.8.2. Molecular docking was verified using Auto Dock tools. Results: We identified 20 active compounds of CBP, 202 active targets, 1,323 disease targets, and 117 active compound disease intersection targets;the PPI network showed that the top eight key targets were AKT1, interleukin(IL) 6, JUN, MAPK1, tumor necrosis factor, TP53, vascular endothelial growth factor, and epidermal growth factor. These act mainly on the fluid shear stress, atherosclerosis, IL-17, hypoxia-inducible factor-1, and nonalcoholic fatty liver disease pathways, among others, which control immune inflammation, cell proliferation, and apoptosis. Based on molecular docking results, the active components of CBP bind well to its key targets. Conclusion: Our results indicated that CBP was effective for treating AD by reducing the expression of inflammatory factors, promoting cell proliferation and differentiation, and preventing nerve cell death and anti-neuronal cell apoptosis.

Examining the characteristics between time and distance gaps of secondary crashes

Understanding the characteristics of time and distance gaps between the primary(PC)and secondary crashes(SC)is crucial for preventing SC ccurrences and improving road *** previous studies have tried to analyse the variation of gaps,there is limited evidence in quantifying the relationships between different gaps and various influential *** study proposed a two-layer stacking framework to discuss the time and distance ***,the framework took random forests(RF),gradient boosting decision tree(GBDT)and eXtreme gradient boosting as the base classifiers in the first layer and applied logistic regression(LR)as a combiner in the second *** this basis,the local interpretable model-agnostic explanations(LIME)technology was used to interpret the output of the stacking model from both local and global *** SC dentification and feature selection,346 SCs and 22 crash-related factors were collected from California interstate *** results showed that the stacking model outperformed base models evaluated by accuracy,precision,and recall *** explanations based on LIME suggest that collision type,distance,speed and volume are the critical features that affect the time and distance *** volume can prolong queue length and increase the distance gap from the SCs to *** collision types,peak periods,workday,truck involved and tow away likely induce a long-distance ***,there is a shorter distance gap when secondary roads run in the same direction and are close to the primary *** speed is a significant factor resulting in a long-time gap,while the higher speed is correlated with a short-time *** results are expected to provide insights into how contributory features affect the time and distance gaps and help decision-makers develop accurate decisions to prevent SCs.
