Chemical composition-based characterization of the anti-allergic effect of Guominkang Formula on IgE-mediated mast cells activation and passive cutaneous anaphylaxis

Guominkang(GMK),a Chinese medicine formula,has been used to treat allergic diseases in clinical settings for many years.To evaluate the antiallergic effect and molecular mechanism of action of GMK extract,RBL-2H3 cell models and passive cutaneous anaphylaxis(PCA)mouse models were established.High performance liquid chromatography(HPLC)and ultra-high performance liquid chromatography-mass spectrometry(UHPLC-MS)analyses were performed to characterize the chemical composition of GMK.A total of 94 compounds were identified or tentatively identified from GMK.Three of them,emodin,ursolic acid,and hamaudol,were identified for the first time as potential active compounds in GMK,since they inhibited the degranulation of mast cells.The anti-allergic effect of hamaudol was the first to be discovered.GMK could markedly mitigate the shade of Evans Blue extravasation and ear incrassation in PCA mouse models.Additionally,GMK significantly inhibited the degranulation of mast cells,suppressed mast cell degranulation by reducing Ca2+influx and the levels of TNF-α,IL-4,and histamine,and markedly inhibited the phosphorylation of Lyn,Syk,PLCγ1,IκBα,and NF-κB p65.Molecular docking results indicated that hamaudol and emodin had strong interaction with FcεRI and NF-κB related proteins,while ursolic acid only interacted with NF-κB associated proteins.These results suggest GMK suppresses the activation of MCs both in vivo and in vitro.The underlying mechanism of its anti-allergic activity is associated with the inhibition of FcεRI and NF-κB activation.

Efficiency and safety of acupuncture for women with premature ovarian insufficiency:study protocol for a randomized controlled trial

Acupuncture has been widely used as an alternative and complementary therapy for premature ovarian insufficiency(POI)in China.However,research to date has not shown that acupuncture is effective for POI compared with hormone replacement therapy(HRT).We will conduct a randomized,controlled,and outcome assessor-blind trial to evaluate the efficacy and safety of acupuncture on POI.Seventy-six patients with POI will be randomly assigned to two groups.The treatment group will receive twenty-eight one-hour sessions of acupuncture treatments,and the control group will receive 12-week HRT.The whole study will consist of a 12-week treatment plan and a 12-week follow-up session.The primary outcome is measured by changes in serum anti-Müllerian hormone and follicle-stimulating hormone(FSH)levels at weeks 12 and 24.Secondary outcome measures include estradiol,luteinizing hormone(LH),LH/FSH ratio,Kupperman index,and menstrual condition.This trial is expected to clarify whether or not acupuncture is effective and safe for POI compared with HRT.

Generating of rice OsCENH3-GFP transgenic plants and their genetic applications

In order to investigate rice functional centromeres, OsCENH3-GFP chimeric gene was constructed and transformed into the indica rice variety, Zhongxian 3037, mediated by Agrobacturium. The integration of the exogenous genes in the transgenic plants was confirmed by PCR and Southern blotting. The transgenic plants grow normally during their whole life time, just like Zhongxian 3037. No significant defects were detected in either mitosis or meiosis of the transgenic plants. The overlapping of GFP signals and anti-CENH3 foci in both mitotic and meiotic cells from T0 and T1 generation plants indicated that GFP had been successfully fused with CENH3, so the GFP signals can well represent the CENH3 locations on each chromosome. To evaluate the applicability of the transgenic plants to other genetic studies, fluorescence in situ hybridization (FISH) using rice centromeric tandem repetitive sequence CentO as the probe was conducted on the zygotene chromosomes of pollen mother cells (PMCs). It has been revealed that the GFP signals are overlapping with CentO FISH signals, showing that CentO is one of the key elements constituting rice functional centromeres. Immunofluorescent staining using anti-α-tublin antibody and anti-PAIR2 antibody on the chromosomes during mitosis and meiosis stages of the transgenic plants further reveals that OsCENH3-GFP transgenic plants can be widely used for studying rice molecular biology, especially for tagging functional centromeres in both living cells and tissues.

该文以废弃涤棉混纺织物为碳源,分别在乙二醇和硫酸溶液中通过溶剂热法合成了两种荧光碳点,实现了对溶液中Cr(Ⅵ)和Hg(Ⅱ)的检测。通过透射电镜与红外光谱表征了碳点的形貌与组成,并基于紫外吸收光谱和荧光光谱分析了碳点的电子跃迁形式及发光类型。在乙二醇体系中制得的碳点(ETCCDs)能够选择性检测Cr(Ⅵ),检出限为0.093 mg/L,其检测机理为荧光内滤效应;硫酸体系中制备的碳点(WTCCDs)能够选择性检测Hg(Ⅱ),检出限为0.018µmol/L,检测机理为能量转移。对实际水样中的Cr(Ⅵ)和Hg(Ⅱ)进行检测,验证了上述两种碳点的实用性。基于涤棉混纺织物制备的荧光碳点不仅实现了对两种重金属离子的检测,同时也为废旧纺织物的再生利用提供了借鉴意义。

针对工业无线局域网日益复杂的安全态势,开展了零信任工业无线局域网安全访问技术研究。首先,为了兼容工业网络中不具备扩展性终端,在融合无线局域网安全认证与软件定义边界(SDP)安全接入技术的基础上,提出了兼容传统无线工业终端的零信任工业无线局域网安全访问系统框架,在无线接入点中内嵌SDP透明代理,并适应性地引入了SDP网关与安全控制器;然后,设计了面向SDP与无线网络认证融合的身份体系,以及基于SDP透明代理的服务安全访问机制,同时,为了实现终端访问异常行为动态识别及访问权限动态调整,设计了基于终端访问服务的空间、时间、频率多维的信任评估模型,以及基于信任评估的动态访问控制方法。最后,通过系统实验的方法验证了所提出的系统框架及方法的安全性与有效性。

Large magnetostriction of Nd_(0.2)Tb_(0.3)Dy_(0.5)Fe_(1.93)/epoxy composites in a wide temperature range

We report the magnetostrictive performance of the Nd_(0.2)Tb_(0.3)Dy_(0.5)Fe_(1.93)/epoxy composites.Measurement of M?ssbauer spectra and the XRD results indicate that the easy magnetic direction of the Nd_(0.2)Tb_(0.3)Dy_(0.5)Fe_(1.93)compound is alongdirection with spontaneous magnetostrictionλ111 of1900 ppm.The-oriented Nd_(0.2)Tb_(0.3)Dy_(0.5)Fe_(1.93)/epoxy composites with particle volume fractions of20 vol%-55 vol%were prepared.Magnetization measurements demonstrate that the composite is highly anisotropic.The raw material cost of Nd_(0.2)Tb_(0.3)Dy_(0.5)Fe_(1.93)is about 85%of that of the Terfenol-D(Tb_(0.3)Dy_(0.7)Fe_(2))composition.Nd_(0.2)Tb_(0.3)Dy_(0.5)Fe_(1.93)/epoxy composites possess comparable magnetostriction with those composites made of Tb_(0.3)Dy_(0.7)Fe_(2)particles.At room temperature,the magnetostrictionλ_Ⅱof the Nd_(0.2)Tb_(0.3)Dy_(0.5)Fe_(1.93)/epoxy composite reaches 420 ppm at a low field of 80 kA/m and about1000 ppm at 239 kA/m.In addition,the composite exhibits a large saturation magnetostrictionλ_Ⅱover1200 ppm from 300 to 100 K,suggesting its potential applications as a low-cost magnetostrictive material in a wide temperature range.Therefore,the current study offers an opportunity to widen the operating temperature range of the magnetostrictive composite.

Weight Analysis of Impact Factors of Interbedded Anti-Inclined Slopes Block-Flexure Toppling Based on Support Vector Regression

Block-flexure toppling failure is frequently encountered in interbedded anti-inclined rock(IAR)slopes,and seriously threatens the construction of hydropower infrastructure.In this study,we first investigated the Lean Reservoir area’s geological setting and the Linda landslide’s characteristics.Then,uniform design and random design were used to design 110 training datasets and 31 testing datasets,respectively.Afterwards,the toppling response was obtained by using the discrete element code.Finally,support vector regression was used to obtain the influence weights of 21 impact factors.The results show that the influence weight of the slope angle and rock formation dip angle on the toppling deformation among tertiary impact factors is 25.96%and 17.28%,respectively,which are much greater than the other 19 impact factors within the research range.For the primary impact factors,the influence weight is sorted from large to small as slope geometry parameters,joints parameters,and rock mechanics parameters.Joints parameters,especially the geometric parameters,cannot be ignored when evaluating the stability of IAR slopes.Through numerical simulation,it was qualitatively determined that failure surfaces of slopes were controlled by cross joints and that the rocks in the slope toe play a role in preventing slope deformation.
