Comparing gene expression profiles of Kashin-Beck and Keshan diseases occurring within the same endemic areas of China

In this study, differentially expressed genes in peripheral blood from patients with Kashin-Beck disease and Keshan disease were compared to further investigate the etiology and pathogenesis of both diseases, which occur in a common endemic area of China. Twenty Kashin-Beck disease patients and 12 healthy controls, and 16 Keshan disease patients and 16 healthy controls, were grouped into four pairs. Patients and controls were selected from common endemic areas for the two diseases. Total RNA was isolated from peripheral blood mononuclear cells from all patients and controls, and gene expression profiles analyzed by oligonucleotide microarrays. Sixteen genes differentially expressed in both Kashin-Beck disease and Keshan disease (versus controls) were identified, and comprised nine genes showing synchronous and seven asynchronous expression. The Comparative Toxicogenomics Database shows that expression and biological function of these genes can be affected by multiple environmental factors, including mycotoxin and selenium content, potential environmental risk factors for the two diseases. Thus, these shared differentially expressed genes may contribute to the distinct organ lesions, caused by common environmental risk factors of Kashin-Beck disease and Keshan disease.

Background Keshan disease （KD） is an endemic cardiomyopathy in China.The etiology of KD is still under debate and there is no effective approach to preventing and curing this disease.Young women of child-bearing age are the most frequent victims in rural areas.The aim of this study was to determine the differences between molecular pathogenic mechanisms in male and female KD sufferers.Methods We extracted RNA from the peripheral blood mononuclear cells of KD patients （12 women and 4 men） and controls （12 women and 4 men）.Then the isolated RNA was amplified,labeled and hybridized to Agilent human 4×44k whole genome microarrays.Gene expression was examined using oligonucleotide microarray analysis.A quantitative polymerase chain reaction assay was also performed to validate our microarray results.Results Among the genes differentially expressed in female KD patients we identified：HLA-DOA,HLA-DRA,and HLA-DQA1 associated with spontaneous autoimmunity; BMP5 and BMP7,involved in cardiomyocyte differentiation defect; and ADAMTS 8,CCL23,and TNFSF15,implicated in anti-angiogenic activities.These genes are involved in the canonical pathways and networks recognized for the female KD sufferers and might be related to the pathogenic mechanism of KD.Conclusion Our results might help to explain the higher susceptibility of women to this disease.

基于海水温度历史观测和海洋模式数值预报数据,利用长短期记忆(LSTM)神经网络开展了海水温度剖面短时预报方法研究。以南海17°46.91′N,112°03.24′E处海水温剖面预报为研究对象,利用模式预报数据和观测数据,构建了观测、预报、观测和预报混合3个样本数据集。基于LSTM神经网络模型,建立了由编码器-解码器组成的多对多海水温度剖面序列预报模型,并开展了模型的训练和性能验证分析工作。结果表明:该模型具有较高的预报精度,对小样本问题的处理具有良好的稳定性;采用深度分层预报,可有效提高模型预报精度,改善模型泛化能力;与只使用观测数据集的预报相比,混合样本集预报误差下降明显,为基于小样本观测数据的海洋环境要素预报研究提供了一种思路。

硒对肿瘤坏死因子-α诱导的AC16心肌细胞炎性损伤的影响

目的探讨硒对肿瘤坏死因子-α(TNF-α)诱导的AC16心肌细胞炎性损伤的影响。方法将体外培养的AC16心肌细胞分为对照组、硒预处理组(100 μg/L亚硒酸钠预处理4 h)、50 μg/L TNF-α组、硒预处理+ 50 μg/L TNF-α组、100 μg/L TNF-α组、硒预处理+ 100 μg/L TNF-α组。各组分别培养24 h后, 收集细胞培养液和细胞进行检测。Griess法检测细胞培养液一氧化氮(NO)含量;荧光倒置显微镜观察细胞Hoechst 33342核染色, 分析核固缩比例;实时荧光定量PCR检测B淋巴细胞瘤-2基因(Bcl-2)、B淋巴细胞瘤-2相关X蛋白(Bax)及诱导型一氧化氮合酶(iNOS)mRNA表达水平, 蛋白质印迹法检测核因子-kappa B(NF-κB)p65、NF-κB抑制蛋白-α(IκB-α)蛋白表达水平。结果对照组、硒预处理组、50 μg/L TNF-α组、硒预处理+ 50 μg/L TNF-α组、100 μg/L TNF-α组和硒预处理+ 100 μg/L TNF-α组细胞培养液NO含量分别为(10.58 ± 2.32)、(9.07 ± 0.73)、(15.53 ± 3.97)、(12.05 ± 1.11)、(30.65 ± 4.16)、(19.02 ± 3.72)μmol/L。硒、TNF-α对细胞培养液NO含量均存在主效应(F = 37.71、99.07, 均P < 0.001), 且二者间存在交互效应(F = 11.80, P < 0.001)。硒、TNF-α对核固缩比例, Bcl-2、Bax mRNA表达水平均存在主效应(F = 56.37、462.81, 18.04、32.85, 18.38、170.77, 均P < 0.05), 且硒、TNF-α对核固缩比例, Bax mRNA表达水平均存在交互效应(F = 21.48、19.96, 均P < 0.001)。硒、TNF-α对iNOS mRNA表达水平均存在主效应(F = 129.98、1 051.76, 均P < 0.001), 且二者间存在交互效应(F = 53.28, P < 0.001)。硒、TNF-α对NF-κB p65、IκB-α蛋白表达水平均存在主效应(F = 73.65、145.49, 710.20、105.66, 均P < 0.001), 且二者间均存在交互效应(F = 26.73、197.59, 均P < 0.001)。结论硒可能通过NF-κB信号系统抑制iNOS表达, 保护心肌细胞免受TNF-α诱导的炎性损伤。

研学旅行是新时代教育的重要抓手之一,高校所具有的厚重历史文化资源、所集聚的名师名家与学生等人力资源、所拥有的不同专业学科及其实验(训)室和图书馆等设施设备资源,使高校彰显出开展研学旅行的优势,众多知名院校打开了研学旅行发展的新局面,而普通地方院校研学旅行发展则不尽如人意。其中,研学旅行课程的开发与设计是制约普通地方院校研学旅行发展的重要因素。借鉴在教育教学设计领域具有广泛实用性和适用性的ADDIE模型,改进优化后形成ADIERI模型,并以上饶师范学院为例,提出包括前端分析、系统设计、科学实施、评价总结的地方院校研学旅行课程开发方案,以期对地方院校研学教学实践及其应用推广提供参考。

Intrinsic vacancy in 2D defective semiconductor In_(2)S_(3) for artificial photonic nociceptor

It is crucial to develop an advanced artificially intelligent optoelectronic information system that accurately simulates photonic nociceptors like the activation process of a human visual nociceptive pathway.Visible light reaches the retina for human visual perception,but its excessive exposure can damage nearby tissues.However,there are relatively few reports on visible light–triggered nociceptors.Here,we introduce a two-dimensional natural defectiveⅢ–Ⅵsemiconductorβ-In_(2)S_(3)and utilize its broad spectral response,including visible light brought by intrinsic defects,for visible light–triggered artificial photonic nociceptors.The response mode of the device,under visible light excitation,is very similar to that of the human eye.It perfectly reproduces the pain perception characteristics of the human visual system,such as‘threshold,’‘relaxation,’‘no adaptation’,and‘sensitization’.Its working principle is attributed to the mechanism of charge trapping associated with the intrinsic vacancies in In_(2)S_(3)nanosheets.This work provides an attractive material system(intrinsic defective semiconductors)for broadband artificial photonic nociceptors.

Turning depths of internal tides in the South China Sea inferred from profile data

Theoretically,propagating internal tides in the ocean may reflect at turning depths,where buoyancy frequencies equal tidal frequencies,before colliding with the air-sea interface or rugged bottom topography.Globally,the internal tide lower turning depths(ITLTDs)in the open ocean have been mapped;however,knowledge of the presence of ITLTDs in the South China Sea(SCS)is lacking.In this study,2125 high-quality temperature-salinity profiles(including 58 deep-sea hydrographic measurements with observational depths exceeding 3000 m)are collected and analyzed to investigate the existence of ITLTDs in the SCS.Furthermore,the concept of the upper turning depth is first introduced in the context of internal tides,and internal tide upper turning depths(ITUTDs)are also investigated.ITLTDs are found to exist at several abyssal stations;these stations are distributed mostly in the southern part of the SCS basin,possibly due to the greater water depths there.Fewer locations show the presence of ITLTDs for K_(1) versus M_(2) tidal frequencies because of the lower tidal frequency.The distance between ITLTDs and the seafloor ranged from 270 m to more than 1200 m,implying the possible existence of multiple internal wave evanescent regions in the abyssal bottom.ITUTDs of tens of meters are ubiquitous in the SCS;stations with the presence of ITUTDs are located mainly in the northeastern SCS due to the intensive observations there.However,the calculated ITUTDs have large uncertainties;they are sensitive to the selected bin values.The horizontal propagation directions of internal tides in the SCS change dramatically,and as a result,the estimated turning depths under the full Coriolis force definition are different compared to that under the traditional approximation.

目的探讨静息状态下双相障碍三种临床相边缘系统网络脑区间功能连接及脑区内低频振荡振幅(amplitude of low-frequency fluctuation,ALFF)是否有共性改变。方法于2019年7月至2021年12月纳入中南大学湘雅二医院精神卫生研究所确诊为双相障碍的患者107例(双相障碍组)以及与之年龄、性别、受教育程度等相匹配的健康对照者46名(对照组)。根据杨氏躁狂评定量表(Young Mania Rating Scale,YMRS)及17项汉密尔顿抑郁量表(Hamilton Depression Rating Scale,HAMD17)评分将双相障碍患者分为双相躁狂组、双相抑郁组、双相缓解组。采集所有受试者静息态功能磁共振成像数据,采用基于种子点功能连接和ALFF的方法分析其边缘系统网络脑区间功能连接及脑区内功能活动状态,使用单因素方差分析及事后检验的方法比较组间功能连接值和ALFF值的差异。采用Pearson或Spearman对组间差异有统计学意义的脑区功能连接值和ALFF值与临床量表总分及各因子分进行相关分析。结果与对照组相比,双相障碍三种临床相边缘系统网络中前扣带回皮质与海马区的功能连接显著降低(t=-2.60~-2.07,Bonferroni校正,P<0.05),且三种临床相前扣带回皮质ALFF也显著降低(t=-4.41~-3.51,FDR校正,P<0.05)。双相抑郁组左侧前扣带回与左侧海马功能连接值与HAMD17中工作兴趣因子分呈负相关(r=-0.406,P=0.021)。结论双相障碍边缘系统网络中前扣带回的失连接和功能活动降低可能是双相障碍三种临床相情绪调节障碍的共性改变。

Person-independent expression recognition based on person-similarity weighted expression feature

A new method to extract person-independent expression feature based on higher-order singular value decomposition （HOSVD） is proposed for facial expression recognition. Based on the assumption that similar persons have similar facial expression appearance and shape, the person-similarity weighted expression feature is proposed to estimate the expression feature of test persons. As a result, the estimated expression feature can reduce the influence of individuals caused by insufficient training data, and hence become less person-dependent. The proposed method is tested on Cohn-Kanade facial expression database and Japanese female facial expression （JAFFE） database. Person-independent experimental results show the superiority of the proposed method over the existing methods.
