Protective effect of astrocyte-conditioned medium on neurons following hypoxia and mechanical injury

Objective： To investigate the protective effect of mouse astrocyte-conditioned medium （ACM） on hypoxic and mechanically injured neurons by a cell model in vitro, and to explore the possible mechanism. Methods： The model of hypoxic neuronal injury was caused by 3% 02 in three-gas incubator. Neurons were cultured with ordinary medium or 20% ACM respectively and randomly divided into hypoxic group （hypoxia for 4, 8, 24 h and marked as H4R0, H8R0, H24R0） and hypoxia reoxygenation group （H4R24, H8R24, H24R24）. Mechanical injury model was developed by scratching neurons cultured in 20% ACM or ordinary medium to different degrees. Neu- rons in both medium were divided into normal control group, mild, moderate and severe injury groups. The 20% ACM was added 24 h before hypoxia/reoxygenation or mechanical injury. The morphology and survival of neurons were observed and counted by trypan blue staining. The concentration of NO, lactic dehydrogenase （LDH） and membrane ATPase activity were detected by corresponding kits. Results： It was showed that 20% ACM can obviously promote the survival rate of hypoxia/reoxygenated neurons and scratched neurons as well. The morphology and num- ber of neurons exposed to hypoxia or scratch injury showed great difference between groups with or without ACM treatment. Compared with control group, the concentration of NO and LDH was much lower in hypoxic/reoxygenated neurons treated with 20% ACM, and the ATPase activity was higher. For the mechanical injury model, neurons with moderate injury also revealed a lower NO and LDH concen- tration than the control group. All the differences were sta- tistically significant （P〈0.05）. Conclusion： ACM can promote the survival and func- tional recovery of neurons following hypoxia or scratching to a certain degree. The mechanism may be associated with reducing the synthesis and release of NO and LDH as well as increasing the activity of membrane ATPase.

AIM: To evaluate the receptor protein which can specifically bind to β2GPⅠon the membrane of hepatocellular carcinoma (HCC) cell line SMMC-7721, and to study the biological function of the ***: Through β2GPⅠ-affinity chromatography column, the peptid-polysome-mRNA complex, which can specially bind to β2GPⅠ, stayed with the column and was separated from the whole polysome of liver cells, and then eluted and collected. Using cDNA synthesis kit and cDNA PCR kit, the corresponding cDNA was obtained and sequenced. RT-PCR was used to amplify annexinⅡ, and flow cytometry was used to study the competitive binding of annexinⅡ with β2GPⅠto ***: A total of 1.1 kb of the cDNA fragment of the specific binding protein of β2GPⅠon liver cell membrane was obtained. The sequence of cDNA shared high homology with human annexinⅡ (98%). AnnexinⅡ was expressed on the membrane of SMMC-7721, and could compete with β2GPⅠfor combining with ***: The receptor for β2GPⅠon membrane of SMMC-7721 cells is annexinⅡ, which might bridge HBV to infect hepatocytes.

目的:检索、评价和总结成人肝移植围手术期营养管理的相关证据,为临床制定营养策略及患者自我管理提供循证依据。方法:系统检索国内外数据库中关于成人肝移植围手术期营养管理的相关文献,根据相关文献评价标准进行评价并提取证据。结果:共纳入11篇文献,其中指南7篇、专家共识3篇、系统评价1篇,最终总结出20条证据,包括营养风险筛查与评估、骨骼肌减少症的筛查、体质量管理、术前营养管理、术后营养管理、院外随访6个方面。结论:该研究总结的成人肝移植围手术期营养管理的证据具有科学性和实用性,可为临床医护人员提供参考和借鉴,使其更有针对性、科学性地指导肝移植术后患者的自我管理。

Evaluation of mountain slope stability considering the impact of geological interfaces using discrete fractures model

The stability of rock slope is often controlled by the existing discontinuous surfaces, such as discrete fractures, which are ubiquitously distributing in a geological medium. In contrast with the traditional approaches used in soil slope with a continuous assumption, the simulation methods of jointed rock slope are different from that of in soil slope. This paper presents a study on jointed rock slope stability using the proposed discontinuous approach, which considers the effects of discrete fractures. Comparing with traditional methods to model fractures in an implicit way, the presented approach provides a method to simulate fractures in an explicit way, where grids between rock matrix and fractures are independent. To complete geometric components generation and mesh partition for the model, the corresponding algorithms were devised. To evaluate the stability state of rock slope quantitatively, the strength reduction method was integrated into our analysis framework. A benchmark example was used to verify the validation of the approach. A jointed rock slope, which contains natural fractures, was selected as a case study and was simulated regarding the workflow of our framework. It was set up in the light of the geological condition of the site. Slope stability was evaluated under different loading conditions with various fracture patterns. Numerical results show that fractures have significant contributions to slope stability, and different fracture patterns would lead to different shapes of the slip surface. The devised method has the ability to calculate a non-circular slip surface, which is different from a circular slip surface obtained by classical methods.
