Fibrous dysplasia(FD)is a rare,disabling skeletal disease for which there are no established *** evidence supports inhibiting the osteoclastogenic factor receptor activator of nuclear kappa-B ligand(RANKL)as a potential treatment *** this study,we investigated the mechanisms underlying RANKL inhibition in FD tissue and its likely indirect effects on osteoprogenitors by evaluating human FD tissue pre-and post-treatment in a phase 2 clinical trial of denosumab(NCt03571191)and in murine in vivo and ex vivo preclinical *** analysis of human and mouse tissue demonstrated increased osteogenic maturation,reduced cellularity,and reduced expression of the pathogenic Gαs variant in FD lesions after RANKL *** sequencing of human and mouse tissue supported these *** interaction between osteoclasts and mutant osteoprogenitors was further assessed in an ex vivo lesion model,which indicated that the proliferation of abnormal FD osteoprogenitors was dependent on *** results from this study demonstrated that,in addition to its expected antiosteoclastic effect,denosumab reduces FD lesion activity by decreasing FD cell proliferation and increasing osteogenic maturation,leading to increased bone formation within *** findings highlight the unappreciated role of cellular crosstalk between osteoclasts and preosteoblasts/osteoblasts as a driver of FD pathology and demonstrate a novel mechanism of action of denosumab in the treatment of bone disease.

目的探讨体素内不相干运动(intravoxel incoherent motion,IVIm)、纵向弛豫时间定量成像(t1 mapping)、横向弛豫时间定量成像(t2 mapping)等功能磁共振成像技术评估宫内发育迟缓(intrauterine growth restriction,IUGR)仔鼠的肾脏微观结构及灌注改变的价值。方法通过孕期低蛋白饮食法建立IUGR仔鼠模型。低蛋白饮食孕鼠所产仔鼠随机分为IUGR8周组、IUGR12周组,正常饮食孕鼠所产仔鼠随机分为正常8周组、正常12周组,各组n=8。比较各组仔鼠肾脏皮质、髓质的表观弥散系数(apparent diffusion coefficient,ADC)、真实扩散系数(D_(t))、伪扩散系数(D*)、灌注分数(f)、t1值、t2值,以及血肌酐、血尿素氮水平。结果IUGR 12周组肾脏髓质Dt值高于IUGR 8周组,IUGR 12周组肾脏髓质D*值低于正常12周组与IUGR 8周组(Pt1值高于皮质,IUGR 12周组肾脏髓质t1值高于IUGR 8周组与正常12周组,IUGR 12周组肾脏皮质t1值高于正常12周组(Pt2值均高于皮质(Pt2值比较差异均无统计学意义(P>0.05)。各组血肌酐及血尿素氮含量比较差异无统计学意义(P>0.05)。IUGR 8周组仔鼠肾脏可见肾小球增生肥大,未见明显纤维化改变;IUGR 12周组仔鼠肾脏可见肾小球萎缩,囊腔狭窄,间质出现炎性细胞浸润及纤维化。结论IVIm磁共振成像可用于评估及动态观察IUGR仔鼠肾脏微观结构及灌注损伤,t1 mapping磁共振成像可用于评估IUGR仔鼠肾脏损伤,t1 mapping联合t2 mapping磁共振成像可进一步分辨IUGR仔鼠的肾脏纤维化。

Purpose: Diet and eating habits are major risk factors for the health and the development of disease, such as, for example, metabolic disorder leading to cardiovascular pathology and cancer, decreased immunity exposing to infections. this study of the physico-chemical and nutritional properties of a soumara-based food broth was carried out with the aim of promoting the consumption of organic broth made from nere seeds (soumara). that is to alleviate certain metabolic diseases, which is a matter of food safety, and also to limit the risk for the health about the consumption of some cooking stocks on the market. methods: Several natural ingredients such as nere seeds (soumara), ginger, black pepper, parsley and garlic were used to create a nere-based stock. All these ingredients were freeze-dried and the powder obtained was used to make the broth, regarding their physical and chemical properties. Results: the broth had a good protein content of 17.41 ± 0.367 g/100g, a lipid content of 16.80 ± 0.08 g/100g and a fiber content of 8.66 ± 0.04 g/100g. In terms of nutritional values, the broth showed good levels of calcium 184.21 ± 0.09 mg/100g, potassium 50.04 ± 1.45 mg/100g and iron and zinc. In terms of antioxidant activity, the broth also showed good antioxidant activity. Conclusion: Regarding the properties of our food broth, whose composition is based on natural ingredients, could be recommended for consumption and, its properties, could play an important role in preventing and combating certain metabolic diseases.

BACKGROUND Liver grafts from donation after circulatory death(DCD)are associated with a higher risk of early graft dysfunction,determined by the warm ischemia and cold ischemia *** is essential to have precise criteria to identify this complication in order to guide therapeutic *** to validate different graft and recipient survival scores in patients undergoing liver transplantation(Lt)with DCD *** A retrospective and observational unicentric study was conducted on 65 Lt patients with grafts obtained from controlled DCD donors from November 2013 to November *** United Kingdom(UK)risk score,early allograft dysfunction(EAD)Olthoff score,and model for early allograft function(mEAF)score were used to evaluate the risk of graft and recipient survival *** survival analysis purposes,we used the Kaplan-meier method,and the differences between subgroups were compared using the log-rank(mantel-Cox)*** Sixty-five patients were included in the *** UK risk score did not demonstrate predictive capacity for recipient or graft ***,in donors aged over 70 years old(18.4%),it significantly predicted graft survival(Pto Kaplan-meier survival curves,graft survival rates at 6 months,2 years,and 5 years in the futility group dramatically decreased to 50%compared to the other groups(log-rank 8.806,Pthe EAD Olthoff and mEAF scores did not demonstrate predictive capacity for recipient or graft *** on Kaplan-meier survival curves,patients with a mEAF score≥7 had a lower graft survival rate at 6 months,2 years,and 5 years compared to patients with a lower mEAF score(log-rank 4.667,Pries,both UK DCD risk score and mEAF score showed predictive capability for graft survival.

BACKGROUND Among diverse profound impacts on patients’quality of life(QoL),end-stage renal disease(ESRD)frequently results in increased levels of depression,anxiety,and *** replacement therapies such as hemodialysis(HD)and transplantation(tX)are intended to enhance QoL,although their ability to alleviate psychological distress remains *** research posits the existence of a significant correlation between negative emotional states and QoL among ESRD patients,with varying effects observed in HD and tX *** to examine the relationship between QoL and negative emotional states(depression,anxiety,and stress)and predicted QoL in various end-stage renal replacement therapy patients with *** this cross-sectional study included HD or tX patients in the Eastern Region of Saudi *** 36-item Short Form Survey and Depression Anxiety Stress Scale(DASS)was used for data collection,and correlation and regression analyses were *** the HD and tX transplantation groups showed statistically significant inverse relationships between QoL and DASS *** patients with high anxiety levels and less education scored low on the physical component summary(PCS).In addition,the results of the mental component summary(mCS)were associated with reduced *** with older transplant patients,tX patients’PCS scores were lower,and depression,stress,and negative working conditions were highly correlated with mCS *** the findings of this study revealed notable connections between well-being and mental turmoil experienced by individuals undergoing HD and *** PCS of HD patients is affected by heightened levels of anxiety and lower educational attainment,while the mCS of transplant patients is influenced by advancing age and elevated stress *** insights will contribute to a more comprehensive understanding of patient support.

the superplastic behavior and associated deformation mechanisms of a fine-grained mg-10.1 Li-0.8Al-0.6Zn alloy(LAZ1011)with a grain size of 3.2μm,primarily composed of the BCCβphase and a small amount of the HCPαphase,were examined in a temperature range of 473 K to 623 *** microstructural refinement of this alloy was achieved by employing high-ratio differential speed *** best superplasticity was achieved at 523 K and at strain rates of 10^(-4)-5×10^(-4)s^(-1),where tensile elongations of 550±600%were *** the heating and holding stage of the tensile samples prior to tensile loading,a significant increase in grain size was observed at temperatures above 573 ***,it was important to consider this effect when analyzing and understanding the superplastic deformation behavior and *** the investigated strain rate range,the superplastic flow at low strain rates was governed by lattice diffusion-controlled grain boundary sliding,while at high strain rates,lattice diffusion-controlled dislocation climb creep was the rate-controlling deformation *** was concluded that solute drag creep is unlikely to *** the late stages of deformation at 523 K,it was observed that grain boundary sliding led to the agglomeration of theαphase,resulting in significant strain *** mechanism maps were constructed forβ-mg-Li alloys in the form of 2D and 3D formats as a function of strain rate,stress,temperature,and grain size,using the constitutive equations for various deformation mechanisms derived based on the data of the current tests.
