Alkaptonuria (AKU) is an inherited disorder of tyrosine metabolism caused by lack of active enzyme homogentisate 1,2-dioxygenase (HGD). the primary consequence of HGD deficiency is increased circulating homogentisic acid (HGA), the main agent in the pathology of AKU disease. Here we report the first metabolomic analysis of AKU homozygous Hgd knockout (Hgd−/−) mice to model the wider metabolic effects of Hgd deletion and the implication for AKU in humans. Untargeted metabolic profiling was performed on urine from Hgd−/− AKU (n = 15) and Hgd+/− non-AKU control (n = 14) mice by liquid chromatography high-resolution time-of-flight mass spectrometry (Experiment 1). the metabolites showing alteration in Hgd−/− were further investigated in AKU mice (n = 18) and patients from the UK National AKU Centre (n = 25) at baseline and after treatment with the HGA-lowering agent nitisinone (Experiment 2). A metabolic flux experiment was carried out after administration of 13C-labelled HGA to Hgd−/−(n = 4) and Hgd+/−(n = 4) mice (Experiment 3) to confirm direct association with HGA. Hgd−/− mice showed the expected increase in HGA, together with unexpected alterations in tyrosine, purine and tCA-cycle pathways. Metabolites with the greatest abundance increases in Hgd−/− were HGA and previously unreported sulfate and glucuronide HGA conjugates, these were decreased in mice and patients on nitisinone and shown to be products from HGA by the 13C-labelled HGA tracer. Our findings reveal that increased HGA in AKU undergoes further metabolism by mainly phase II biotransformations. the data advance our understanding of overall tyrosine metabolism, demonstrating how specific metabolic conditions can elucidate hitherto undiscovered pathways in biochemistry and metabolism.

Flexible light-emitting fibers and fabrics serve to bridge human–machine interactions. the desire for practical applications and the commercialization of flexible light-emitting fibers has accelerated structural progress and improvements. this review focuses on the structural design of light-emitting fibers and fabrics, starting with a summary of design principles, emission mechanisms, and structural evolution of coaxial structured light-emitting fibers. Subsequently, we explore recent advances in the helical structure design strategies that boost the mechanical sensitivity of light-emitting fibers. Following that, we analyze continuous preparation processes and the development of large-area intelligent light-emitting fabrics based on interwoven structures. Examples based on stiff and rigid inorganic-based lightemitting diodes integrated into flexible systems are also presented. Finally, we discuss the current challenges and future opportunities for light-emitting applications in the field of wearable and smart devices.

It has been reported that trivalent cerium salts are effective inhibitors for corrosion of AA2024-t3 in neutral to alkaline co rrosive enviro nments,but poor in acidic *** this study,work was done for extending corrosion resistance provided by Ce(NO_(3))_(3)to acidic pHs as low as 2.5 through the addition of iodide *** this end,potentiodynamic polarization was used to ascertain the optimum Ce(Ⅲ)concentration for the inhibition of AA2024-t3 corrosion in 3.5%NaCl *** optimum concentration is found to be in a range between 0.1 and 0.3 mmol/L Ce(Ⅲ).this optimum concentration also gives a maximum width of the passive region under the pitting potential(Epit).Based on this optimization,further work was performed as a function of pH without and in the presence of 0.01 mol/L KI at five pH values(2.5,4,5,7,and 8).Without iodide the best inhibition provided by the Ce(Ⅲ)cation is at pH=7 but active corrosion is observed at pH=2.5 with an estimated inhibitor efficiency(-37.6%).the addition of iodide ions(I^(-))reverses the poor corrosion resistance whereby the inhibitor efficiency becomes 47.7%.Moreover,at pH=4 the efficiency is also increased from 54.0%to 81.0%with iodide addition providing a significant improvement over Ce(Ⅲ)cations alone under acidic *** is a synergistic effect between I-anion and Ce(Ⅲ)cation,which is determined to be 2.4 and 2.1 at pH=2.5and 4.0,*** pH=5 and 7 there is virtually no change in inhibition but at pH=8 the inhibition falls away due to a considerable amount of cerium precipitating out of solution as cerium *** protective film formed on sites in the aluminium alloy surface at different pH values was examined by both scanning electron microscopy with energy dispersive X-ray analysis and X-ray photoelectron spectroscopy to determine the role of these ions as a function of pH in surface *** is proposed that at pH=2.5 protection is provided by a bilayer adsorption of iodide with"cap

A Systematic Review of the Genetic Predisposition to Injury in Football

Purpose Genetic research in football is currently in it’s infancy but is growing ***,the practical application of genetic testing in football and the views concerning its use are ***,the purpose of this study was to assess the current practical application of genetic testing in professional football and provide an insight into the perspectives of key stakeholders(i.e.,coaches,practitioners,players).Methods In total,122 participants completed an online anonymous *** consisted of 21 multiple choice and Likert scale questions,with the option of providing an explanation for each *** Findings revealed genetic testing is rarely utilised by key stakeholders(10%)or their respective organisations(14%).However,three quarters(75%)had the opinion that genetic testing will have great utility in the *** majority(72%)believed genetic testing should be used for athlete development and injury risk,whilst 35%believed that genetic testing should be utilised for talent identification ***,most key stakeholders viewed their own(89%)and their col-leagues’(79%)knowledge related to genetic testing as insufficient;mainly due to ineffective current communication methods(91%).Most believed educational workshops are required(71%),whilst nearly all(91%)were interested in developing their expertise on the utility of genetic *** Genetic testing is rarely used within professional football,although key stakeholders anticipate that it will be utilised more in the *** such,educational support may prove valuable in improving key stakeholder knowledge and the practical application of genetic testing in professional football.

the vascular network of the circulatory system plays a vital role in maintaining homeostasis in the human *** this paper,a novel modular microfluidic system with a vertical two-layered configuration is developed to generate largescale perfused microvascular networks in *** two-layer polydimethylsiloxane(PDMS)configuration allows the tissue chambers and medium channels not only to be designed and fabricated independently but also to be aligned and bonded *** method can produce a modular microfluidic system that has high flexibility and scalability to design an integrated platform with multiple perfused vascularized tissues with high *** medium channel was designed with a rhombic shape and fabricated to be semiclosed to form a capillary burst valve in the vertical direction,serving as the interface between the medium channels and tissue *** and anastomosis at the vertical interface were successfully achieved by using different combinations of tissue chambers and medium *** large-scale microvascular networks were generated and quantified in terms of vessel length and *** leakage of the perfused 70-kDa FItC-dextran confirmed the lumenization of the microvascular networks and the formation of tight vertical interconnections between the microvascular networks and medium channels in different structural *** platform enables the culturing of interconnected,large-scale perfused vascularized tissue networks with high density and scalability for a wide range of multiorgan-on-a-chip applications,including basic biological studies and drug screening.

Karst habitats are hotspots of diversity and endemism. their naturally fragmented distributions across broad geographic landscapes have led to the complex array of smaller evolutionary ecosystems that present unique challenges from a conservation *** assessments of karst habitats have revealed that these ecosystems contain an almost unparalleled level of endemism, and many site-restricted species remain undescribed, thus posing considerable challenges for effective conservation *** rock-dwelling species, such as geckos, may be particularly prone to such isolation. In this paper, we discuss one such genus, i.e.,Hemiphyllodactylus, and explore its diversity across karst landforms in Yunnan Province, southwestern China. Based on morphological and genetic data, we describe two new species of Hemiphyllodactylus from karst habitats in Simao District and Yanshan County. A phylogenetic tree for Hemiphyllodactylus was constructed using 1?039 base pairs(bp) of the mitochondrial NADH dehydrogenase subunit 2 gene(ND2). the Simao and Yanshan specimens can be distinguished from all other congeners within their respective subclades based on uncorrected genetic pairwise distances greater than 6.3% and 4.3%respectively, as well as significant morphological differences. the discovery and description of these two new species brings the total number of described Hemiphyllodactylus species in China to 14and indicates many more undescribed species from unsurveyed karst regions await *** findings suggest that karst ecosystems in Yunnan support a higher diversity of Hemiphyllodactylus than previously *** study also highlights the importance of karst ecosystems as refugia for sitespecific endemic species and the need for heightened conservation efforts.
