Deciphering gastric inflammation-induced tumorigenesis through multi-omics data and AI methods

Gastric cancer(GC), the fifth most common cancer globally, remains the leading cause of cancer deaths worldwide. Inflammation-induced tumorigenesis is the predominant process in GC development;therefore, systematic research in this area should improve understanding of the biological mechanisms that initiate GC development and promote cancer hallmarks. Here, we summarize biological knowledge regarding gastric inflammation-induced tumorigenesis, and characterize the multi-omics data and systems biology methods for investigating GC development. Of note, we highlight pioneering studies in multi-omics data and state-of-the-art network-based algorithms used for dissecting the features of gastric inflammation-induced tumorigenesis, and we propose translational applications in early GC warning biomarkers and precise treatment strategies. this review offers integrative insights for GC research, with the goal of paving the way to novel paradigms for GC precision oncology and prevention.

Active ingredients from highland barley have received considerable attention as natural products for developing treatments and dietary supplements against obesity.In practical application,the research of food combinations is more significant than a specific food component.this study investigated the lipid-lowering effect of highland barley polyphenols via lipase assay in vitro and HepG2 cells induced by oleic acid(OA).Five indexes,triglyceride(tG),total cholesterol(t-CHO),low density lipoprotein-cholesterol(LDL-C),aspartate aminotransferase(ASt),and alanine aminotransferase(ALt),were used to evaluate the lipidlowering effect of highland barley extract.We also preliminary studied the lipid-lowering mechanism by Realtime fluorescent quantitative polymerase chain reaction(q PCR).the results indicated that highland barley extract contains many components with lipid-lowering effects,such as hyperoside and scoparone.In vitro,the lipase assay showed an 18.4%lipase inhibition rate when the additive contents of highland barley extract were 100μg/m L.the intracellular lipid-lowering effect of highland barley extract was examined using 0.25 mmol/L OA-induced HepG2 cells.the results showed that intracellular tG,LDL-C,and t-CHO content decreased by 34.4%,51.2%,and 18.4%,respectively.ALt and ASt decreased by 51.6%and 20.7%compared with the untreated hyperlipidemic HepG2 cells.q PCR results showed that highland barley polyphenols could up-regulation the expression of lipid metabolism-related genes such as PPARγand Fabp4.

Catalytic transfer hydrogenation(CtH)is a green and efficient pathway for selective hydrogenation of unsaturated aldehydes and ketones.However,managing the abilities of solid catalysts to adsorb substrates and to convert them into desired products is a challenging task.Herein,we report the synthesis of carbon coated LaFe_(0.92)Pd_(0.08)O_(3) composites(LFPO-8@C)for CtH of benzaldehyde(BzH)into benzyl alcohol(BzOH),using isopropanol(IPA)as hydrogen source.the coating with carbon improves the ability to adsorb/transfer reactants from solution to active sites,and the doping of Pd2+at Fe3+site strengthens the ability of LaFeO_(3) to convert BzH into BzOH.A balanced point between them(i.e.,abilities to adsorb BzH and to convert BzH into BzOH)is obtained at LFPO-8@C,which exhibits a BzOH formation rate of 3.88 mmol·gcat^(-1)·h^(-1) at 180℃ for 3 h,which is 1.50 and 2.72 times faster than those of LFPO-8 and LaFeO_(3)@C.A reaction mechanism is proposed,in which the acidic sites(e.g.,Fe^(4+),oxygen vacancy)are used for the activation of C=O bond of BzH and O-H bond of IPA,and the basic sites(e.g.,lattice oxygen)for the activation ofα-H(O-H)bond of IPA.

Lactobacillus are considered promising therapeutic methods for nonalcoholic fatty liver disease(NAFLD).the effects of two strains of Ltmosilactobacillus mucosae on NAFLD were investigated in this study.Fourweek-old male C57BL/6J mice were divided into 4 groups(n=8 per group,Control,Model,FZJtZ26M3,FGSYC17L3).L.mucosae FZJtZ26M3 reduced the mice 's body weight,liver weight,and adipose tissue weight after 12 weeks of therapy.According to serum analysis,total cholesterol,triacylglycerol,and low-density lipoprotein cholesterol significantly decreased after L.mucosae FZJtZ26M3 intervention.Liver pathology showed that L.mucosae FZJtZ26M3 was effective to ameliorate lipid deposition in NAFLD mice.Additionally,the expression of the gene related to lipid metabolism in the liver and adipose tissue was analyzed,and the results indicated that L.mucosae FZJtZ26M3 could alleviate NAFLD by regulating lipid metabolism.Furthermore,the results of 16S rRNA gene sequencing revealed a drop in the relative abundance of Ruminococcaceae,which is linked to inflammation,but the relative abundance of a potential probiotic Akkermansia significantly increased after L.mucosae FZJtZ26M3 intervention.Generally,L.mucosae FZJtZ26M3 could be a candidate to prevent NAFLD.

Background:Bitter taste receptors(tas2rs)are generally considered to sense various bitter compounds to escape the intake of toxic substances.Bitter taste receptors have been found to widely express in extraoral tissues and have important physiological functions outside the gustatory system in vivo.Methods:to investigate the physiological functions of the bitter taste receptor cluster tas2r106/tas2r104/tas2r105/tas2r114 in lingual and extraoral tissues,multiple tas2rs mutant mice and Gnat3 were produced using CRISPR/Cas9 gene-editing technique.A mixture containing Cas9 and sgRNA mRNAs for tas2rs and Gnat3 gene was microinjected into the cytoplasm of the zygotes.then,t7EN1 assays and sequencing were used to screen genetic mutation at the target sites in founder mice.Quantitative real-time polymerase chain reaction(qRt-PCR)and immunostaining were used to study the expression level of taste signaling cascade and bitter taste receptor in taste buds.Perception to taste substance was also studied using twobottle preference tests.Results:We successfully produced several tas2rs and Gnat3 mutant mice using the CRISPR/Cas9 technique.Immunostaining results showed that the expression of GNAt3 and PLCB2 was not altered in tas2rs mutant mice.But qRt-PCR results revealed the changed expression profile of m tas2rs gene in taste buds of these mutant mice.With two-bottle preference tests,these mutant mice eliminate responses to cycloheximide due to genetic mutation of tas2r105.In addition,these mutant mice showed a loss of taste perception to quinine dihydrochloride,denatonium benzoate,and cucurbitacin B(CuB).Gnat3-mediated taste receptor and its signal pathway contribute to CuB perception.Conclusions:these findings implied that these mutant mice would be a valuable means to understand the biological functions of tAS2Rs in extraoral tissues and investigate bitter compound-induced responses mediated by these tAS2Rs in many extraoral tissues.

Systemic immune monitoring is a crucial clinical tool for disease early diagnosis,prognosis and treatment planning by quantitative analysis of immune cells.However,conventional immune monitoring using flow cytometry faces huge challenges in large-scale sample testing,especially in mass health screenings,because of time-consuming,technical-sensitive and high-cost features.However,the lack of high-performance detection platforms hinders the development of high-throughput immune monitoring technology.to address this bottleneck,we constructed a generally applicable DNA framework signal amplification platform(DSAP)based on post-systematic evolution of ligands by exponential enrichment and DNA tetrahedral framework-structured probe design to achieve high-sensitive detection for diverse immune cells,including CD4+,CD8+t-lymphocytes,and monocytes(down to 1/100μl).Based on this advanced detection platform,we present a novel high-throughput immune-cell phenotyping system,DSAP,achieving 30-min one-step immune-cell phenotyping without cell washing and subset analysis and showing comparable accuracy with flow cytometry while significantly reducing detection time and cost.As a proof-of-concept,DSAP demonstrates excellent diagnostic accuracy in immunodeficiency staging for 107 HIV patients(AUC>0.97)within 30 min,which can be applied in HIV infection monitoring and screening.therefore,we initially introduced promising DSAP to achieve high-throughput immune monitoring and open robust routes for point-of-care device development.

舌鳞状细胞癌患者非常规淋巴结转移分析

目的分析舌鳞状细胞癌(简称鳞癌)颈部淋巴结转移的特点。方法采用回顾性研究方法,纳入2010年6月至2018年3月间在中南大学湘雅二医院口腔颌面外科就诊,行原发灶及颈淋巴清扫(简称颈清)的连续整块切除手术的舌鳞癌患者329例,其中男性283例,女性46例,年龄26~80岁。均采用新颈清术式:与经典颈清的主要区别在于非常规淋巴结的处理,彻底清扫甲状腺上动脉、面动脉基部、颈外动脉各分支旁淋巴脂肪组织,将舌动脉及其沿线组织及口底淋巴结与原发灶一并切除。计数资料的比较采用χ2检验,多因素分析采用线性回归模型。结果本研究共136例(41.3%)患者发生了颈淋巴结转移。142例t1-2cN0期患者行肩胛舌骨上清扫,其中22例(15.5%)存在隐匿性淋巴结转移,与其余120例无淋巴结转移者相比,5年总生存率差异无统计学意义(90.2%比92.1%,χ2=0.156,P=0.693)。多因素分析显示t分期、临床分期以及非常规淋巴结转移是舌鳞癌患者出现颈部淋巴结转移的影响因素(P值均th of invasion,DOI)≤5 mm组、5 mm10 mm组的差异有统计学意义[3.1%(2/64)比5.7%(6/106)比13.8%(22/159),χ2=7.907,P=0.005]。结论 cN0舌鳞癌患者选择肩胛舌骨上清扫可获得可靠的控制效果。建议对所有需颈清的患者行非常规淋巴清扫,强烈推荐对存在常规淋巴结转移的患者进行非常规淋巴清扫。

Developing a mucosal vaccine against SARS-CoV-2 is critical for combatting the epidemic.Here,we investigated long-term immune responses and protection against SARS-CoV-2 for the intranasal vaccination of a triple receptor-binding domain(RBD)scaffold protein(3R-NC)adjuvanted with a flagellin protein(KFD)(3R-NC+KFDi.n).In mice,the vaccination elicited RBD-specific broad-neutralizing antibody responses in both serum and mucosal sites sustained at high level over a year.this long-lasting humoral immunity was correlated with the presence of long-lived RBD-specific IgG-and IgA-producing plasma cells,alongside the th17 and tfh17-biased t-cell responses driven by the KFD adjuvant.Based upon these preclinical findings,an open labeled clinical trial was conducted in individuals who had been primed with the inactivated SARS-CoV-2(IAV)vaccine.With a favorable safety profile,the 3R-NC+KFDi.n boost elicited enduring broad-neutralizing IgG in plasma and IgA in salivary secretions.to meet the challenge of frequently emerged variants,we further designed an updated triple-RBD scaffold protein with mutated RBD combinations,which can induce adaptable antibody responses to neutralize the newly emerging variants,including JN.1.Our findings highlight the potential of the KFD-adjuvanted triple-RBD scaffold protein is a promising prototype for the development of a mucosal vaccine against SARS-CoV-2 infection.

Clinical Application of Sex Hormone in Different Physiological Periods in the Diagnosis of Infertility Patients

Background: Infertility is characterized by the inability to conceive after a year of regular unprotected intercourse. Aims: this study aimed to investigate the diagnostic value of sex hormone levels during different physiological periods in the diagnosis of infertility patients. Methods: From December 2019 to May 2021, a total of 93 infertility patients were admitted and selected as the observation group. Among them, 31 cases were in the follicular stage, 31 cases in the ovulation stage, and 31 cases in the luteal stage. Ninety-three healthy women for fertility evaluation due to male infertility were selected as the control group. the control group included 31 women in the follicular phase, 31 women in the ovulatory phase, and 31 women in the luteal phase. the levels of sex hormones (prolactin (PRL), luteinizing hormone (LH), follicle-stimulating hormone (FSH), estradiol (E2), testosterone (t), and progesterone (P)) during different physiological phases were compared between the observation and control groups. Results: the follicular phase showed no significant difference in LH levels between the observation group and the control group. the observation group showed higher levels of PRL and P compared to the control group, while the levels of FSH, E2, and t were lower in the observation group compared to the control group. the ovulation phase showed no significant difference in PRL levels between the two groups. the observation group showed lower levels of LH, FSH, E2, t, and P compared to the control group. the luteal phase showed no statistical difference in E2 levels between the two groups. the observation group showed higher levels of PRL, LH, and FSH compared to the control group, while the levels of t and P were lower in the observation group compared to the control group. Conclusion: Infertile women show variations in hormone levels compared to the normal levels during the follicular phase, ovulatory phase, and luteal phase.
