CRISPR‒Cas7-11 is a type Ⅲ-E CRISPR-associated nuclease that functions as a potent RNA editing ***-peptide repeat fused with Cas/HEF1-associated signal transducer(tPR-CHAt)acts as a regulatory protein that interacts with CRISPR RNA(crRNA)-bound Cas7-11 to form a CRISPR-guided caspase complex(Craspase).However,the precise modulation of Cas7-11’s nuclease activity by tPR-CHAt to enhance its utility requires further ***,we report cryo-electron microscopy(cryo-EM)structures of Desulfonema ishimotonii(Di)Cas7-11-crRNA,complexed with or without the full length or the N-terminus of *** structures unveil the molecular features of the Craspase *** analysis,combined with in vitro nuclease assay and electrophoretic mobility shift assay,reveals that DitPR-CHAt negatively regulates the activity of DiCas7-11 by preventing target RNA from binding through the N-terminal 65 amino acids of DitPR-CHAt(DitPR-CHAt_(NtD)).Our work demonstrates that DitPRCHAt_(NtD) can function as a small unit of DiCas7-11 regulator,potentially enabling safe applications to prevent overcutting and offtarget effects of the CRISPR‒Cas7-11 system.

Chlorine-based disinfectants(CBDs)have been widely used to prevent and control the spread of the COVID-19,which may lead to the formation of carcinogenic *** China,strict disinfection strategies by local governments/communities or volunteering by residents have been implemented to meet the Dynamic COVID Zero(DCZ)***,the amount of CBDs used has not been *** author proposed an urban-scale disinfectant consumption estimation(ALICE)model to quantify weekly CBD *** results show that the CBD consumption for the urban region of Beijing during the DCZ strategy was 3704.0 t(0.43 kg/(cap-yr)),equivalent to a monthly increase of 15 g/cap(70.5%)in CBD consumption compared with that in *** to the scenario analysis,a stricter strategy with a shorter response time toward new cases will decrease the total CBD consumption by 1.2% compared with the baseline estimation.A more precise prevention strategy with a smaller delineation of risk area and a less stringent strategy with a longer response time will lower the total CBD consumption by 0.42% and 0.35%,***,the more precise prevention strategy will reduce CBD consumption of close off and lockdown area(COLD area)by 16.9%,and the stricter strategy will reduce this consumption by 37.7%.this study highlights the impact of pandemic prevention and control strategies on chlorine-based disinfectant consumption and some implications for future environmental pollution and risk assessments.

the fast dynamic properties of the surface of metallic glasses(MGs) play a critical role in determining their potential applications. However, due to the significant difference in thermal history between atomic simulation models and laboratory-made samples, the atomic-scale behaviors of the fast surface dynamics of MGs in experiments remain uncertain. Herein, we prepared model MG films with notable variations in thermal stability using a recently developed efficient annealing protocol, and investigated their atomic-scale dynamics systematically. We found that the dynamics of surface atoms remain invariant, whereas the difference in dynamical heterogeneity between surface and interior regions increases with the improvement of thermal stability. this can be associated with the more pronounced correlation between atomic activation energy spectra and depth from the surface in samples with higher thermal stability. In addition, dynamic anisotropy appears for surface atoms, and their transverse dynamics are faster than normal components, which can also be interpreted by activation energy spectra. Our results reveal the presence of strong liquid-like atomic dynamics confined to the surface of laboratory-made MGs, illuminating the underlying mechanisms for surface engineering design, such as cold joining by ultrasonic vibrations and superlattice growth.

Capsular contracture is a prevalent and severe complication that affects the post-operative outcomes of patients who receive silicone breast *** present,prosthesis replacement is the major treatment for capsular contracture after both breast augmentation procedures and breast reconstruction following breast cancer ***,the mecha-nism(s)underlying breast capsular contracture remains *** study aimed to identify the biological features of breast capsular contracture and reveal the potential underlying mechanism using RNA *** tissues from 12 female patients(15 breast capsules)were divided into low capsular contracture(LCC)and high capsular contracture(HCC)groups based on the Baker ***,41 lipid metabolism-related genes were identified through enrichment analysis,and three of these genes were identified as candidate genes by SVM-RFE and LASSO *** then compared the proportions of the 22 types of im-mune cells between the LCC and HCC groups using a CIBERSORt analysis and explored the Cor-relation between the candidate hub features and immune ***,PRKAR2B was positively correlated with the differentially clustered immune cells,which were M1 macro-phages and follicular helper t cells(area under the ROC=0.786).In addition,the expression of PRKAR2B at the mRNA or protein level was lower in the HCC group than in the LCC *** molecular mechanisms were identified based on the expression levels in the high and low PRKAR2B *** findings indicate that PRKAR2B is a novel diagnostic biomarker for breast capsular contracture and might also influence the grade and progression of capsularcontracture.

the finite Berry curvature in topological materials can induce many subtle phenomena, such as the anomalous Hall effect(AHE), spin Hall effect(SHE), anomalous Nernst effect(ANE), non-linear Hall effect(NLHE) and bulk photovoltaic effects. to explore these novel physics as well as their connection and coupling, a precise and effective model should be developed. Here, we propose such a versatile model—a 3D triangular lattice with alternating hopping parameters, which can yield various topological phases,including kagome bands, triply degenerate fermions, double Weyl semimetals and so on. We reveal that this special lattice can present unconventional transport due to its unique topological surface states and the aforementioned topological phenomena, such as AHE, ANE, NLHE and the topological photocurrent effect. In addition, we also provide a number of material candidates that have been synthesized experimentally with this lattice, and discuss two materials, including a non-magnetic triangular system for SHE, NLHE and the shift current, and a ferromagnetic triangular lattice for AHE and ANE. Our work provides an excellent platform, including both the model and materials, for the study of Berry-curvature-related physics.

surfactants were used to assist in the crystallization of *** types of surfactants with different structures(sodium dodecyl sulfate,cetyltrimethylammonium bromide,or betaine hydrochloride[BH])were *** study indicated that three surfactants were all capable of shortening the crystallization induction time of lysozyme,with BH inducing the shortest crystallization induction time(t=1 min)and the largest crystal size(37μm).the molecular interactions among each of the three surfactants aforementioned and lysozyme were also investigated by *** was found that hydrogen bonding dominantly acted between the carboxyl structure of the surfactant BH and the lysozyme,which produced the most favorable conformation for the self-assembly of lysozyme(promote lysozyme orderly aggregation and shorten the crystallization induction time).On this basis,the mechanism of surfactant-promoted lysozyme crystallization was proposed.

Background:total human immunodeficiency virus(HIV)DNA and integrated HIV DNA are widely used markers of HIV *** digital polymerase chain reaction(ddPCR)can be used for absolute quantification without needing a standard ***,we developed duplex ddPCR assays to detect and quantify total HIV DNA and integrated HIV ***:the limit of detection,dynamic ranges,sensitivity,and reproducibility were evaluated by plasmid constructs containing both the HIV long terminal repeat(LtR)and human CD3 gene(for total HIV DNA)and ACH-2 cells(for integrated HIV DNA).Forty-two cases on stable suppressive antiretroviral therapy(ARt)were assayed in total HIV DNA and integrated HIV *** coefficient analysis was performed on the data related to DNA copies and cluster of differentiation 4 positive(CD4^(+))t-cell counts,CD8^(+)t-cell counts and CD4/CD8 t-cell ratio,*** assay linear dynamic range and lower limit of detection(LLOD)were also ***:the assay could detect the presence of HIV-1 copies 100%at concentrations of 6.3 copies/reaction,and the estimated LLOD of the ddPCR assay was 4.4 HIV DNA copies/reaction(95%confidence intervals[CI]:3.6-6.5 copies/reaction)with linearity over a 5-log_(10)-unit range in total HIV DNA *** the integrated HIV DNA assay,the LLOD was 8.0 copies/reaction(95%CI:5.8-16.6 copies/reaction)with linearity over a 3-log 10-unit *** HIV DNA in CD4^(+)t cells was positively associated with integrated HIV DNA(r=0.76,Pth total HIV DNA and integrated HIV DNA in CD4^(+)t cells were inversely correlated with the ratio of CD4/CD8 but positively correlated with the CD8^(+)t-cell ***:this ddPCR assay can quantify total HIV DNA and integrated HIV DNA efficiently with robustness and *** can be readily adapted for measuring HIV DNA with non-B clades,and it could be beneficial for testing in clinical trials.
