Articular cartilage lesion repair remains a challenging problem due to its avascular,aneural and alymphatic properties resulting in poor regenerative *** chondrocyte transplantation(ACt),a cell-based therapy,has provided a new perspective in articular cartilage management however its translation for lab based models to clinical application needs to be evaluated to ensure safety and efficacy issues are addressed in human *** a series of experiments from in vivo animal trials to clinical pilot models,both in rabbit followed by human experiment were carried out and assessed in our centre to ensure these issues were *** the animal model, ACt was carried out in the right knees of the rabbits while defects on the left knees to serve as a *** and immunohistochemistry assessment were carried out at 3 months and 6 months postoperative and further evaluated by histological and histochemical grading *** our success of animal transplantation work,human ACt(HACt) was conducted in patients with chondral defect identified by MRI and *** assessment of ACt was carried out by radiological evaluation at preoperative,6 and 12 months *** assessment of the human ACt showed good to excellent result in 90%of the cases by which were then evaluated with the Oxford Knee *** comparing to the animal model,the defective area treated with ACt showed significant healing compared to the untreated *** therefore believe that our proposed model of research(which serves as a translational research also known as "bench to bedside" research) proves successful and is an appropriate model for future researches wanting to apply clinical cell *** result of this study will also convince surgeons/clinical practitioners of the efficacy of any experimental therapy and most importantly to further pursue future possibilities in cell-based therapy in reconstructive surgery.
