Impact of Lignosulfonate on Solution Chemistry and Phospholipid Fatty Acid Composition in Soils

An incubation experiment(Exp. 1) with three soils, two from Australia and one from Norway, was carried out to investigate the fate of dissolved BorreGro(a lignosulfonate, produced by Borregaard Lignotech Company, Norway) at different concentrations(0, 10 and 100 mg C L-1) in soil solutions. A rhizobox experiment(Exp. 2) was also done in a Norwegian clay soil, mixed with four levels of BorreGro-carbon(BG-C) added(0, 2, 20 and 200 mg BG-C kg-1) to test the impact of BorreGro on root growth, rhizosphere chemistry(pH, metals and dissolved organic carbon(DOC)) and the composition of phospholipid fatty acids(PLFAs). the BorreGro addition increased the concentration of Mn due to the high concentrations in BorreGro. the BorreGro addition to soil had an indirect but significant impact on the rhizosphere chemistry and PLFAs. the lowest amounts of added BorreGro facilitated the DOC excretion at plant roots, and thereby increased the bacterial and fungal biomass, likely as an effect of increased Mn solubility from BorreGro in the root zone.

Investigation of HCAR2 antagonists as a potential strategy to modulate bovine leukocytes

Background Dairy cows experiencing ketosis after calving suffer greater disease incidence and are at greater risk of leaving the herd. In vitro administration of beta-hydroxybutyric acid(BHBA;the primary blood ketone) has inhibitory effects on the function of bovine leukocytes. BHBA is a ligand of HCAR2 and the activation of these receptors promotes an anti-inflammatory response which may be related with immunosuppression observed in transition dairy cattle. the objective of this study was to identify and test antagonists for HCAR2 in bovine immune cells cultured with *** We observed expression of HCAR2 at the protein level within lymphocytes, monocytes, and granulocytes. the proportion of cells expressing HCAR2 tended to be greater in mid-lactation compared to early lactation cows;the increase was a result of increased proportion of t and B cells expressing HCAR2. Stimulation of HCAR2 with niacin or BHBA promoted Ca^(2+) mobilization in neutrophils and mononuclear cells. Mononuclear cells treated with BHBA had diminished intracellular Ca^(2+) responses when HCAR2 was knocked down by si RNA silencing, indicating Ca^(2+) mobilization was mediated by HCAR2 signaling. two candidate antagonists for HCAR2, synthesized from niacin(NA-1 and NA-5), were tested;monocytes and neutrophils pre-treated with NA-1 and NA-5 had reduced Ca^(2+) mobilization after incubation with BHBA. Furthermore, NA-5 but not NA-1 prevented BHBA-associated reductions in cyclic *** We demonstrated that HCAR2 is present on bovine leukocytes and has greater expression later in lactation. We confirmed that BHBA and niacin derived HCAR2 antagonists alter bovine leukocyte activity. Our results demonstrate that both BHBA and niacin affect bovine leukocyte Ca^(2+) mobilization in a HCAR2-dependent manner.

the thymus begins involution in childhood and historically it was thought to b e nonfunctional by adulthood, thus presenting no contraindication to the routine practice of thymectomy during cardiothoracic surgery. More recent data suggest, however, that the thymus remains active into adulthood and is responsible for t he low-level production of normal t cells. We hypothesize, therefore, that inci dental thymectomy during cardiothoracic surgery in infancy causes long-term cha nges in the cellular immune system. to investigate this hypothesis, we quantifie d peripheral t-cell subsets and t-cell recombination excision circles in child ren with congenital heart disease to measure the impact of cardiothoracic surgic al procedures and thymectomy performed during a period of immunologic developmen t. We found that cardiothoracic surgical procedures, especially if they include thymectomy, impair t-cell production and produce long-term decreases in total lymphocyte count and CD4+and CD8+t-cell subsets, suggesting that long-term m aintenance of lymphocyte populations is disturbed.
