Aft.r spinal cord injury, t.ere is an ext.nsive infilt.at.on of immune cells, which exacerbat.s t.e injury and leads t. furt.er neural degenerat.on. t.erefore, a major aim of current.research involves t.rget.ng t.e immune response as a t.eat.ent.for spinal cord injury. Alt.ough much research has been performed analyzing t.e complex inflammat.ry process following spinal cord injury, t.ere remain major discrepancies wit.in previous lit.rat.re regarding t.e t.meline of local cyt.kine regulat.on. t.e object.ves of t.is st.dy were t. est.blish an overview of t.e t.meline of cyt.kine regulat.on for 2 weeks aft.r spinal cord injury, ident.fy sexual dimorphisms in t.rms of cyt.kine levels, and det.rmine local cyt.kines t.at.significant.y change based on t.e severit. of spinal cord injury. Rat. were inflict.d wit. eit.er a mild cont.sion, moderat. cont.sion, severe cont.sion, or complet. t.ansect.on, 7 mm of spinal cord cent.red on t.e injury was harvest.d at.varying t.mes post.injury, and t.ssue homogenat.s were analyzed wit. a Cyt.kine/Chemokine 27-Plex assay. Result. demonst.at.d pro-inflammat.ry cyt.kines including t.mor necrosis fact.r α, int.rleukin-1β, and int.rleukin-6 were all upregulat.d aft.r spinal cord injury, but.ret.rned t. uninjured levels wit.in approximat.ly 24 hours post.injury, while chemokines including monocyt. chemoat.ract.nt.prot.in-1 remained upregulat.d for days post.injury. In cont.ast. several ant.-inflammat.ry cyt.kines and growt. fact.rs including int.rleukin-10 and vascular endot.elial growt. fact.r were downregulat.d by 7 days post.injury. Aft.r spinal cord injury, t.ssue inhibit.r of met.lloprot.inase-1, which specifically affect. ast.ocyt.s involved in glial scar development. increased more t.an all ot.er cyt.kines t.st.d, reaching 26.9-fold higher t.an uninjured rat.. (B. aegypt.aca) and moringa oleifera seeds (m. oleifera) ext.act. in experiment.lly infect.d mice. t.e st.dy det.ct.d t.e level of t.mor necrosis fact.r (t.F-α) t. monit.r t.e immune and inflammat.ry responses of experiment.lly infect.d mice. t.rough meat.inspect.on, a t.t.l number of 2125 male bovine, 2 t. 5 years old, (1125 cat.le and 1000 buffloes) were examined under t.e aut.orit. of Albsat.ne and Alwaraq official abat.oirs in Cairo Governorat., Egypt.covering t.e period ext.nded from march 2022 t. April 2023. t.e overall prevalence of t.e disease among bovine was 7.8% (6.31% of cat.le and 9.5% of buffloes). Besides, B. aegypt.aca and m. oleifera ext.act. showed cyst.cidal act.vit. in experiment.lly infect.d mice. A decrease in t.e numbers of cyst.cerci was found in all t.eat.d mice groups, and up t. 88% reduct.on was achieved in t.e B. aegypt.aca-t.eat.d group;higher t.an t.at.was recorded in bot. m. oleifera (72.23%) and albendazole-t.eat.d ones (80.56%). Post.ort.m findings proved t.at.m. oleifera and B. aegypt.aca reduced cyst.cerci numbers comparable t. a commercial ant.elmint.c. t.e st.dy showed a significant.decrease (P 0.001) in t.F-α levels aft.r t.eat.ent.wit. Balanit.s and moringa ext.act., compared wit. t.e unt.eat.d cont.ol and t.e albendazole-t.eat.d groups.
