In Streptococcus pyogenes, we have described the two-component signal transduction sensor and regulatory systems, CovR/S affect the antimicrobial susceptibility including penicillin G before. But the mechanism how two-component sensor protein CovS modulates penicillin G susceptibility has not been elucidated till date. This study aimed to determine how the CovS affected penicillin G susceptibility in Streptococcus pyogenes by northern blot analysis. At first, we investigated the covS mRNA expression under penicillin G induction. We found that the decrease of covS mRNA expression under Penicillin G stimulation. Next we investigated the expression of cell wall synthesis gene, pbp2a and glmM with use of covS knockout mutants from emm1 Streptococcus pyogenes strain 1529. We found that the cell-wall synthesis gene expression of the ?covS mutant strain were lower than that of the wild-type strain. Furthermore the expression of glmM mRNA gene was lower than the expression of pbp2a mRNA gene in the ?covS mutant strain. The covS-complemented strain almost restored the mRNA expression compared to covS mutant strain. The two-component sensor protein CovS affects the susceptibility to penicillin G in Streptococcus pyogenes by modulation of cell-wall synthesis.

On the recovery from the Little Ice Age

A number of published papers and openly available data on sea level changes, glacier retreat, freezing/break-up dates of rivers, sea ice retreat, tree-ring observations, ice cores and changes of the cosmic-ray intensity, from the year 1000 to the present, are studied to examine how the Earth has recovered from the Little Ice Age (LIA). We learn that the recovery from the LIA has proceeded continuously, roughly in a linear manner, from 1800-1850 to the present. The rate of the recovery in terms of temperature is about 0.5°C/100 years and thus it has important implications for understanding the present global warming. It is suggested on the basis of a much longer period covering that the Earth is still in the process of recovery from the LIA;there is no sign to indicate the end of the recovery before 1900. Cosmic-ray intensity data show that solar activity was related to both the LIA and its recovery. The multi-decadal oscillation of a period of 50 to 60 years was superposed on the linear change;it peaked in 1940 and 2000, causing the halting of warming temporarily after 2000. A low loss polarization maintaining photonic crystal fiber has been fabricated. The fiber loss was 1.3 dB/km at 1550 nm. The polarization crosstalk for a 100-m fiber was -35 dB at 1550 nm.

The performance of polymer light emitting devices(PLEDs)based on polyvinyl carbazole(PVK)is improved by introducing a nanoscale interfacial thin layer,made of poly(ethylene oxide)(PEO),between the calcium cathode and the PVK emissive *** is believed that the PEO layer plays a key role in enhancing the device *** comparison to the device with Ca/Al as the cathode,the performance of the PLED with PEO/Ca/Al cathode,including the driving voltage,luminance efficiency is significantly *** improvements are attributed to the introduction of a thin layer of PEO that can lower the interfacial barrier and facilitate electron injection.

In graphite furnace atomic absorption spectrometry (GF-AAS), the atomization process of lead occurring in graphite atomizers with/without a platform plate was investigated when palladium was added to an iron-matrix sample solution containing trace amounts of lead. Absorption profiles of a lead line were meas- ured at various compositions of iron and palladium. Variations in the gas temperature were also estimated with the progress of atomization, by using a two-line method under the assumption of a Boltzmann distribu- tion. Each addition of iron and palladium increased the lead absorbance in both the atomizers, indicating that iron or palladium became an effective matrix modifier for the determination of lead. Especially, palladium played a significant role for controlling chemical species of lead at the charring stage in the platform-type atomizer, to change several chemical species to a single species and eventually to yield a dominant peak of the lead absorbance at the atomizing stage. Furthermore, the addition of palladium delayed the peak after the gas atmosphere in the atomizer was heated to a higher temperature. These phenomena would be because the temperature of the platform at the charring stage was elevated more slowly compared to that of the furnace wall, and also because a thermally-stable compound, such as a palladium-lead solid solution, was produced by their metallurgical reaction during heating of the charring stage. A platform-type atomizer with palladium as the matrix modifier is recommended for the determination of lead in GF-AAS. The optimum condition for this was obtained in a coexistence of 1.0 × 10–2 g/dm3 palladium, when the charring at 973 K and then the atomizing at 3073 K were conducted.

Single port laparoscopic right hemicolectomy for ileocolic intussusception

A 36-year-old male was admitted with right lower abdominal pain and diarrhea for more than 3 mo. Colonoscopy and a barium enema study revealed a submucosal tumor over the cecum, but computed tomography showed an ileal lipoma. There was no definitive diagnosis preoperatively, but ileocolic intussusception was noted during surgery. Single port laparoscopic radical right hemicolectomy was performed because intra-operative reduction failed. The histological diagnosis of the resected tumor was lipoma. Single port laparoscopic surgery has recently been proven to be safe and feasible. There are advantages compared with conventional laparoscopic surgery, such as smaller incision wounds, fewer port site complications, and easier conversion. However, there are some drawbacks which need to be overcome, such as difficulties in triangulation and instrument clashing. If there are no contraindications to laparoscopy, single port laparoscopic surgery can be performed safely and should be considered for diagnosis and treatment of intussusception in adults. Here, we report the first case of ileocolic intussusception successfully treated by single port laparoscopic surgery.

Percentile analysis of plasma total bilirubin—How different will the rate of phototherapy for jaundice of neonates be by different standards?

Phototherapy of a jaundiced neonate is usually started when bilirubin exceeds a threshold in the standard. There are several standards used in the developed countries even though the guideline of American Academy of Pediatrics is considered to be a global standard. Although the purpose of phototherapy is the prevention of kernicterus, nowadays the prevalence of kernicterus in otherwise healthy term neonates in the developed countries is rare. Meanwhile several potential adverse effects of phototherapy have been reported. In the present study we tried to estimate how different the rate of phototherapy for the jaundice of neonates at lower risk of kernicterus would be by different standards. For this purpose, we utilized the records of plasma total bilirubin (TB) values of 1893 healthy neonates of 38 weeks and more which were measured on day 6 for the percentile analysis. However, this database did not include the TB values of the neonates who received phototherapy by day 6. Then the database was corrected with an assumption that TB on day 6 would have been normally distributed if no phototherapy had been performed. The mean and the standard deviation (SD) of corrected database were 11.29 mg/dl (193 μmol/l) and 3.63 mg/dl (62 μmol/l), respectively. Using a standard distribution with these mean and SD, the percents of TB values which exceed 18.0, 19.0, 20.0 and 21.0 mg/dl on day 6 were estimated 3.22%, 1.70%, 0.82% and 0.35%, respectively. Results of the present report would help to estimate the relative rate of phototherapy that is performed for the neonates who are term and otherwise healthy by different standards.

Cloning and Characterization of δ-Guaiene Synthase Genes Encoding a Sesquiterpene Cyclase from Aquilaria microcarpa Cell Cultures

Three cDNA clones encoding δ-guaiene synthase, a sesquiterpene cyclase, were isolated from tissue cultures of Aquilaria microcarpa, and data mining analysis of the orthologous genes suggested that 10 and 9 amino acid residues of N- and C-terminal ends of the translated products of these clones remained undefined. The recombinant enzyme proteins, to which the putative missing Nand C-terminal amino acid sequences (MSSAKLGSAS and ALLRHAIEI, respectively) were ligated, exhibited the catalytic activities of sesquiterpene biosynthesis. Among these three δ-guaiene synthases, two isoforms were capable of liberating α-guaiene, δ-guaiene, β-elemene plus α-humulene as a minor product, while remaining one isoenzyme generated α-, δ-guaiene and β-elemene but not α-humulene. Although the enzyme protein solely lacking in the N-terminal 10 amino acid residues was capable of synthesizing the sesquiterpenoids, the protein without 9 amino acids at Cterminal did not exhibit the catalytic activity. These results suggest that two types of δ-guaiene synthase;α-, δ-guaiene, β-elemene-producing type, and α-, δ-guaiene, β-elemene plus α-humulene-producing type;concomitantly occur in A. microcarpa cell cultures, and several amino acid residues at C-terminal of the synthase protein are essential to exhibit the catalytic activities as the sesquiterpene cyclase.

Negative Pressure Wound Therapy with Multiple Drainage Holes for the Treatment of Pressure Ulcer with Undermining: Case Reports

Noninvasive treatment of pressure ulcers with undermining is often difficult. To decrease the risk of bleeding in such conditions, negative pressure wound therapy (NPWT) has been applied. We treated a pressure ulcer with wide undermining using NPWT after opening drainage holes in the undermined area. This method can reduce the risk of bleeding and promote the rapid closure of the undermined area.
