Acute and Reproductive Toxicity of the Aqueous Extract of the Dry Seeds of Aframomum daniellii on the Female Wistar Rat Strain

This work was designed to investigate the acute and reproductive toxicity activity of the aqueous extract of the dry seeds of Aframomum daniellii on the female rats. The acute toxicity of the aqueous extract of Aframomum daniellii (A. daniellii) was evaluated with 6 female rats which were divided into 2 groups (1 Test group and the Control group) of 3 female rats each. The control group received distilled water (10 mL/kg/po) and the test group received a single dose of extract of A. daniellii at the dose of 2000 mg/kg. The reproductive toxicity was evaluated using 45 adult female rats which were divided into 5 groups. Group I, received distilled water (1 mL/100 g/po, neutral control);group II, received Clomiphene citrate (600 μg/kg/po, positive control);Groups III, IV and V (trials) received aqueous extract of A. daniellii at doses of 100, 200 and 400 mg/kg/po respectively. The animals were treated daily for 14 days. From the 6th day of treatment, the rats were mated with males of proven fertility for 8 days. On day 22, after laparotomy and delivery, the numb.r of implantation sites, corpora lutea, resorption sites and pups were recorded. Concerning the acute toxicity, it was ob.erved that, after the single dose of 2000 mg/kg administration of the aqueous extract of the dry seeds of A. daniellii, no deaths were recorded. Concerning the reproductive toxicity, no implantation and gestation were ob.erved when compared to the control. However, the aqueous extract of A. daniellii caused a significant (p A daniellii is weakly toxic, b.t could negatively affect some reproductive parameters.

Sexual dysfunction is a group of diseases, disorders or difficulties experienced b. men or women during any stage of normal sexual activity. In Africa in general and Cameroon in particular, statistical data concerning the prevelance and the main causes of male sexual dysfunction are rare due to socio-cultural and religious b.rdens and sometimes self-medication. The ob.ective of this work was to determine the prevalences, comorb.dities and the risk factors of the main sexual dysfunction in some hospitals in city of Douala. Through a descriptive retrospective study carried out from Novemb.e 2019 to June 2021, sociodemographic parameter, reasons of consultation, risk factors and type of male sexual dysfunction (erectile dysfunction or erectile dysfunction (ED), ejaculation disorders (EjD), lib.do disorders (LD) and disorders of sexual activity with pain or painful sexual intercourse (PSI)) were collected in the medical files of patients who have made urological consultation b.tween 2016 and 2020 at the Deido District hospital, Laquintinie Douala Hospital and Douala General hospital respectively. At the end of our data collection, out of a total of 24995 consultations, 2743 (10.98%) patients were suffering from at least one male sexual dysfunction. Moreover, sexual disorders were the 2nd reason of urological consultation (13.69%) after urinary disorders (33.85%). Among sexual disorders, erectile (76%) and ejaculatory (20%) disorders were the predominant pathologies recorded with the prevalences of 9.79 et 2.62% respectively, and were mostly represented in patients from [41 - 50] for erectile dysfunction, and [18 - 30] years old for ejaculative dysfunction. In addition, the numb.r of patients with ejaculatory and erectile dysfunctions was more important in patients from [18 - 30] and [31 - 40] years old respectively. In recent years, the rate of infertility has not stopped increasing in the world. The aim of the present study was to evaluate the effects of the aqueous extract of Schumanniophyton magnificum (Rub.aceae) on cyclicity, ovulation and gestation in mature rats. Methods: After a qualitative phytochemical analysis of these aqueous extracts, the experimental studies carried out were b.sed on the evaluation of the pro-fertility effects of this extract in mature rats. For this purpose, 35 rats were used for the estrous cyclicity test and treated for 21 days at the end of which vaginal smears were taken and the duration, as well as the frequency of the appearance of the phases of the cycle, were evaluated. The ovulation test was performed on 80 female rats, which were divided into two groups of 40 animals and treated respectively in the morning and evening with distilled water, β-oestradiol or plant extract at doses of 200, 400 and 800 mg/kg. At the end of estrus, the rats were sacrificed, the ovaries were removed and weighed, the hemorrhagic points counted and the b.ood samples were taken for hormonal studies. The last phase of the study consisted in evaluating the effects of these plant extracts on the evolution of gestation. Thus, 42 mature rats were treated during the periods from the 1st to the 10th day (1st stage), and from the 11th to the 17th day (2nd stage). At the end of these two phases, a laparotomy was performed and the numb.r of implantation sites and corpus luteum was counted. And finally, at parturition, from the 18th to the 22nd day (3rd stage), the numb.r of living pups was performed and the gestational parameters were calculated. Results: Administration at the b.ginning of prœstrus allowed a significant increase (p S. magnificum extract. Diab.tes mellitus is a socially significant disease characterized b. chronic hyperglycemia and metab.lic disorders of proteins, carb.hydrates, and lipids due to reduced function of insulin. Medicinal plants, rich in b.oactive components that promote prevention and treatment, are inexpensive and no side effects. Apium graveolens, Cleome gynandra, and Hib.scus sab.ariffa from b.rkina Faso were investigated for their phytochemical profile and antioxidant activities. The high-performance thin-layer chromatography profile revealed flavonoids, tannins, and sterols in these herb.ceous. The Hib.scus sab.ariffa methanolic extract exhib.ted the highest total phenolic (138.4 ± 0.5 mg GAE/g DW) and flavonoid (52.8 ± 0.6 mg RuE/g DW) contents comparatively to Cleome gynandra and Apium graveolens. Hib.scus sab.ariffa methanolic extract also presented the highest antioxidant activity (ICb.50b. = 0.31 ± 0.002 mg/mL) using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging capacity assay. A high correlation b.tween flavonoid contents and hydrophilic antioxidant activities (r = 0.99) was ob.erved, indicating that flavonoids contrib.te significatively to these herb.ceous antioxidant properties. Apium graveolens, Cleome gynandra, and Hib.scus sab.ariffa constitute a natural source of phenolic compounds that could b. exploited in diab.tes mellitus management.

Lannea velutina is a traditional herb.l remedy used to treat various diseases, including hypertension (Hb.). Herb.l medicines are affordab.e, and some have minimal side effects;they are rich in b.oactive components that encourage prevention and treatment. This work uses appropriate experimental paradigms to investigate the phytochemical composition, antioxidant activity, and acute oral toxicity of L. velutina trunk b.rk extracted in water, methanol, ethyl acetate, dichloromethane, and hexane. According to a high-performance thin-layer chromatography profile, this shrub.s b.rk contains sterols, saponosides, flavonoids, and tannins. Compared to ethyl acetate and aqueous extracts, the methanol extract had the highest total phenolic (607.06 ± 0.6 mg GAE/g DW), flavonoid (20.97 ± 0.23 mg QE/g DW), and condensed tannins (194.50 ± 0.75 CE/g DW) content. The methanol extract displayed the highest antioxidant activity using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging capab.lity assay (ICb.50b. = 8.59 g/mL;AAI = 4.66) compared to the other extracts. It exhib.ted antiradical action comparab.e to that of Trolox (ICb.50b. = 15.16 g/mL), ascorb.c acid (ICb.50b. = 11.94 g/mL), and catechin (ICb.50b. = 11.64 g/mL). The connection b.tween flavonoid concentration and hydrophilic antioxidant activity was rob.st (r = 0.997). Mice were used to evaluate the acute oral toxicity of ethyl acetate, methanol, and aqueous extracts using guidelines 425 of the Organization for Economic Cooperation and Development. All tested extracts have an estimated LDb.50b. greater than 2000 mg/kg b.dy weight. The truncal b.rk of L. velutina could b. an alternative source for HTA management.

Lannea microcrapa Engl. & K. Krause (Anacardiaceae) is a fruit and medicinal plant widely used in b.rkina Faso. This plant is traditionally used in the treatment of hypertension. The aim of the present study was to evaluate the vasorelaxant effects of the hydroethanolic extract from Lannea microcarpa trunk b.rks (HE_ELM) on the aorta isolated from NMRI mice. Phytochemical screening b. HPTLC, assay of phenolic and flavonoid compounds, assessment of antioxidant activity (DPPH, Ab.S, FRAP, and LPO), and myography of HE_ELM (1 - 2000 μg/mL) on mice thoracic aortas in the presence and ab.ence of endothelium were carried out. Endothelium-dependent and endothelium-independant vasodilation were assessed b. cumulative addition of Ach (1 nM - 10 μM) on aortic rings precontracted with the thromb.xane analogue A2 agonist, 9,11-dideoxy9α,11α-methanoepoxy PGF2α (U46619). L-NAME was used to verify the involvement of NO production in the relaxation mechanism of the extract. Acute oral toxicity of HE_ELM was also evaluated. A phytochemical study revealed the presence of tannins, flavonoids, sterols and triterpenes, saponosides, and high levels of total phenolics and flavonoids. These compounds are thought to b. responsib.e for the extract’s antioxidant and vasorelaxant properties. HE_ELM demonstrated significant antioxidant potential and induced aortic relaxation. Indeed, pharmacological parameters gave ECb.50b. values ranging from 596.45 ± 95.82 μg/mL to 749.48 ± 133.40 μg/mL and Emax values from 85.51% ± 9.59% to 96.81% ± 8.60% for the three conditions of vasodilation of the extract (p > 0.05). A complete antagonism of the contractile effect of U46619 was noted with 1 mg/mL HE_ELM. These results suggest that HE_ELM induces aortic relaxation through a concentration-dependent, endothelium-independent mechanism, possib.y involving intracellular calcium mob.lization of vascular cells. Diospyros mespiliformis Hochst. ex A. DC. (Eb.naceae) is a multi-use plant, including for therapeutic purposes. It is used in alternative medicine in b.rkina Faso to treat conjunctivitis, menorrhagia, dysentery, and diarrhea. The aim of our study was to evaluate the chemical profile, antioxidant and anti-inflammatory activities, safety of use and spasmolytic effects of the aqueous decoction of Diospyros mespiliformis leaves. Phytochemical screening b. HPTLC and assay of compounds of interest were carried out. Four methods were used to assess antioxidant activity. Inhib.tory activity against 15-lipoxygenase and phospholipase A2 was assessed. Acute oral toxicity of the extract was tested on female mice (NMRI). Following these tests, the extract contained b.oactive compounds of interest such as flavonoids, tannins, sterols, triterpenes, and saponosides. The total phenolic and flavonoid contents of the aqueous decoctate were 70.59 ± 3.20 mg EAT/g and 31.57 ± 0.78 mg EQ/g respectively. The extract was less active than Trolox with inhib.tory concentrations of 50% (ICb.50b.) for the Ab.S, DPPH, FRAP, and LPO tests of 7.53 ± 0.08 μg/mL, 29.47 ± 0.06 μg/mL, 1128.83 ± 4.82 mol EAA/g, and 32.30 ± 1.60 μg/mL respectively. The extract has an anti-inflammatory effect with inhib.tion of phospholipase A2 compared to b.tamethasone. In addition, the aqueous extract produced an antispasmodic effect with Emax of 70% and 80% respectively during contractions induced b. b.Clb.2b. and ACh. Finally, this study provided b.sic scientific data and could justify the use of D. mespiliformis leaves in the treatment of diarrhea.
