Role of mycorrhiza to reduce heavy metal stress

Plants have a system of antioxidant enzymes, which helps to alleviate the effects of various types of stresses. Heavy metals like Cadmium and lead are tolerable for plants to certain extent. The antioxidant enzymes do not function properly at higher concentrations of Cadmium, lead and some other heavy metals. The activities of antioxidant enzymes are reduced due to reactive oxygen species produced as a result of heavy metal stress. The catalase activity was directly inhibited by O2- (Kono and Fridovich, 1982). These ROS are O2-, H2O2, and -OH which can react with many other biomolecules. Several metallic ions are produced by radical displacement reactions. These metallic ions inhibit the activity of antioxidant enzymes. Hence, enzymic antioxidant defense system of plants is affected and adversely inhibits plant growth and productivity. Mycorrhizal fungi are important in phytostabilization of toxic heavy metals. Plants having mycorrhizal association accumulate metallic pollutants by storing these heavy metals in Vesicles as well as in fungal hyphae in their roots, hence these metallic pollutants are immobilized and do not inhibit the growth and uptake of phosphorus and some other micronutrients. Mycorrhizal fungi also release various organic acids which increase the solubilisation of insoluble phosphate compounds present in soil. The unavailable forms of phosphorus are converted into available forms as a result of organic acids produced by fungi. AM fungi release glomalins that are certain metal sorble glycoproteins which increase the immobilization of toxic metals. Another protein is metallothionine released by certain AM fungi, which also reduces the heavy metal toxicity in soil. Mycorrhizal fungi also induce resistance in plants against pathogens, drought and salinity stress. Investigation on heavy metal stress resistant genes in mycorrhizal plants can be very helpful for phytoremediation. This review focuses on the use of AM fungi for phytoremediation.

Detection of Pathogenic Bacteria Staphylococcus aureusand Salmonellasp. from Raw Milk Samples of Different Cities of Pakistan

Food-borne diseases are the main public health problem throughout the world. Milk is important component of human diet including fats, proteins, vitamins and minerals. It is a best source of calcium and phosphorus. Different types of pathogenic bacteria like S. aureus and Salmonella enter in milk and then multiply, after multiplication they become active in causing diseases. These bacteria create serious problems for human health. This study aimed to isolate and identify pathogenic bacteria Staphylococcus aureus and Salmonella from raw milk samples of different cities of Pakistan. Primary screening of raw milk samples was done on the basis of morphological, cultural and biochemical techniques. The final identification was made using 16SrRNA sequence analysis. A total of 200 raw milk samples were collected from different cities of Pakistan. Selective medium xylose lysine deoxycholate agar (XLD) and Mannitol salt agar were used for the identification of Salmonella sp. and S. aureus. Staphylococcus aureus produced yellow colonies with yellow zones on Mannitol salt agar. Staphylococcus aureus exhibited gram-positive character with purple coloration and it was detected as cocci-shaped. Biochemically 91 (45%) samples enhibited Catalase, Coagulase, DNase, Urease, Citrate, fermentation tests positive and indole, oxidase and H2S tests negative with nonmotile character, indicating the presence of Staphylococcus aureus. Salmonella sp. was detected as gram negative rods with pink coloration on gram staining. Biochemically 87 (43%) samples revealed catalase, citrate, H2S and fermentation tests positive while oxidase, DNase, Indole and urease tests negative, indicating the presence of Salmonella sp. in these samples. Of the 200 samples tested, 43% were positive for Salmonella, while 45% samples were contaminated with S. aureus. Effects of Arbuscular Mycorrhizal Fungi on Metals Uptake, Physiological and Biochemical Response of Medicago SativaL. with Increasing Zn and Cd Concentrations in Soil

The effect of mycorrhizal symbiosis on metal accumulation and plant tolerance are not commonly studied in medicinal plants under metal stress. The objective of this study was to assess the impact of mycorrhiza on alfalfa plants with the increase of Zn and Cd toxicity. The experiment was conducted under controlled laboratory conditions. Zinc (Zn) and cadmium (Cd) uptake, some biochemical and physiological parameters were studied in eight-week-old alfalfa plants in response to inoculation or not with arbuscular mycorrhizal fungi (AMF) and with the increase of Zn (0, 100, 300, 900 mg·kg-1) and Cd concentrations (0, 100, 300, 600 mg·kg-1) in soil. The results showed that mycorrhizal (M) plants exhibited tolerance to Zn and Cd up to 300 mg·kg-1 in comparison to non-mycorrhizal (NM) plants which exhibited a significant growth reduction at the same soil Zn and Cd level. M inoculation reduced the Zn and Cd accumulation in shoot and showed higher Zn and Cd contents in roots which showed a different Zn and Cd distribution in AMF associated or non-associated plants. Mycorrhizal plants increased phosphorus (P) contents at all Zn and Cd concentrations except the highest (600 and 900 mg·kg-1) leading significant alterations in biochemical contents such as proline, antioxidant enzymes in leaves and also in nutrients (N, P, K, Cu, Ni, Fe, Mn). Zn and cadmium toxicity cause to increase the proline content in shoot of NM plants, however, proline contents are lower in M plants. Results confirmed that AMF protected alfalfa plants against Zn and Cd toxicity. Mycorrhizal colonization was able to form an efficient symbiosis with alfalfa plants in moderately contaminated Zn and Cd soils (300 mg·kg-1) and play an important role in food quality and safety.
