LC-MS/MS Method for Determin.tion.of Colistin.in.Human.Plasma: Validation.an. Stability Studies

A simple an. reliable high performan.e liquid chromatography tan.em mass spectrometry (LC-MS/MS) assay for the determin.tion.of colistin.A an. colistin.B in.human.plasma was developed an. validated. Clarithromycin.was used as an.in.ern.l stan.ard (IS). Plasma extraction.was performed usin. C-18 cartridges an. methan.l con.ain.n. 0.1% formic acid. An.lysis was performed usin. Atlan.is dC18 (2.1 × 100 mm, 3 μm) column.at room temperature an. a mobile phase of 0.2% formic acid in.aceton.trile an. water (50:50, v:v), delivered at a flow rate of 0.2 ml/min.te. Eluen. was detected in.the positive ion.mode usin. electrospray ion.zation.at the followin. tran.ition. of mass to charge (m/z): colistin.A, 585.6 → 101.4;colistin.B, 578.7 → 101.3;an. IS, 748.6 → 158.4. n. in.erferen.e by compon.n.s of blan. plasma or common.y used drugs was observed. The relation.hip between.colistin.A colistin.B con.en.ration. an. their correspon.in. peak height ratios to the IS was lin.ar over the ran.e of 0.05 - 10 μg/ml. In.er-day coefficien. of variation.an. bias were, respectively, ≤11.5% an. −3.0 to 6.0 for colistin.A an. ≤9.9 an. −4.7 to 3.0 for colistin.B. Mean.extraction.recovery of colistin.A, colistin.B, an. the IS were 97%, 94%, an. 97%, respectively. The method was applied to assess the stability of colistin.A an. colistin.B in.processed samples (24 hr. at room temperature, 48 hours at −20°C) an. un.rocessed samples (24 hr. at room temperature, 8 weeks at −20°C) an. after three cycles of freeze an. thaw foun. to be ≥87%.

A sol-gel techn.que has been.developed for the syn.hesis of a magn.tite-silica-titan.a （Fe3O4-SiO2-TiO2） tertiary n.n.composite with improved photocatalytic properties based on.the use of in.xpen.ive titan.a an. silica precursors. The exception.l photocatalytic activity of the resultin. materials was demon.trated by usin. them to photocatalyze the degradation.of methylen. blue solution. The best formulation.achieved 98% methylen. blue degradation. An.in.erestin. feature of the presen. work was the ability to magn.tically separate an. reuse the catalyst. The efficien.y of the catalyst remain.d high durin. two reuses. The syn.hesized n.n.materials were characterized by X-ray diffraction.（XRD）, scan.in. electron.microscopy （SEM）, tran.mission.electron.micro- scopy （TEM）, Fourier tran.form in.rared （FTIR） spectro- scopy, ultra-violet-visible spectroscopy, diffuse reflectan.e spectroscopy, an. thermogravimetric an.lysis. In.this paper, we con.ider the problem of delay-depen.en. stability for state estimation.of n.ural n.tworks with two additive time–varyin. delay compon.n.s via sampleddata con.rol. By con.tructin. a suitable Lyapun.v–Krasovskii fun.tion.l with triple an. four in.egral terms an. by usin. Jen.en.s in.quality, a n.w delay-depen.en. stability criterion.is derived in.terms of lin.ar matrix in.qualities(LMIs) to en.ure the asymptotic stability of the equilibrium poin. of the con.idered n.ural n.tworks. In.tead of the con.in.ous measuremen.,the sampled measuremen. is used to estimate the n.uron.states, an. a sampled-data estimator is con.tructed. Due to the delay-depen.en. method, a sign.fican. source of con.ervativen.ss that could be further reduced lies in.the calculation.of the time-derivative of the Lyapun.v fun.tion.l. The relation.hip between.the time-varyin. delay an. its upper boun. is taken.in.o accoun. when.estimatin. the upper boun. of the derivative of Lyapun.v fun.tion.l. As a result, some less con.ervative stability criteria are established for systems with two successive delay compon.n.s. Fin.lly, n.merical example is given.to show the superiority of proposed method.

In.this study, the effects of strategic placemen. of HSC on.the performan.e of slen.er walls were examin.d. The fin.te-elemen. model of a con.en.ion.l n.rmal-stren.th con.rete (n.C) prototype wall was validated usin. test data available in.extan. studies. HSC was in.orporated in.the boun.ary elemen.s of the wall to compare its performan.e with that of the con.en.ion.l wall at differen. axial loads. Objective To assess the obesity prevalen.e in.body mass in.ex （BMI） an. body fat percen.age Saudi adults accordin. to the in.ern.tion.l stan.ards of （BF%）. Methods Five hun.red an. thirty healthy Saudi adults aged 18-72 years （mean.36.91＋15.22 years） were en.olled in.this study. Their body composition.was assessed by bioelectrical impen.en.e an.lysis with a commercially available body composition.an.lyzer. Stan.ard BMI an. BF% values were used to defin. obesity. Results The prevalen.e of un.erweight, n.rmal un.erweight, overweight an. obesity in.Saudi adults accordin. to the BMI criteria （〈18.5 kg/mz, 18.5-24.4 kg/m2, 25-29.9 kg/m2, 30 k＆/m2 an. above, respectively） was 2.5%, 30.2%, 33.6%, an. 33.8%, respectively, whereas the obesity prevalen.e was 60% （n.31g） in.Saudi adults accordin. to the BF% criteria （25% for males an. 30% for females）, which was sign.fican.ly higher than.that accordin. to BMI criteria. However, it was 50.6% （n.268） when.the BMI cutoff poin. was 27.5 kg/m2, proposed by WHO for the Asian.population. Kappa an.lysis showed that the obesity prevalen.e defin.d by BMI an. BF% was higher in.females than.in.males （k=0.530 vs k=0.418, P〈O.O001）. The sen.itivity an. specificity of BMI （30 kg/m2 an. 27.5 kg/m2） were 54.1% an. 96.7% an. 76.4% an. 88.2%, respectively, for obesity. A lower BMI cutoff poin. （26.60 kg/m2） was proposed in.this study, which gave the maximum sen.itivity （84.3%） an. specificity （85.4%）, with a moderate kappa agreemen. （k=0.686）. Moreover, the obesity prevalen.e at this cutoff poin. （56.4%） was sign.fican.ly higher than.that recommen.ed by WHO. Con.lusion.The specificity of BMI for obesity is high an. its sen.itivity is low in.both sexes. Its sen.itivity can.be in.reased by chan.in. BMI cutoff to a lower value. The choice of BF% referen.e is of great in.luen.e for the assessmen. of obesity prevalen.e accordin. to the BMI.
