com-pacted MsW increases the placement efficiency and thus reduces the space requirement for *** composition of MsW,degradation,and compactive efforts are the key factors that control the compaction characteristics of *** paper presents the laboratory investigation of compaction characteristics of fresh and aged MsW at a wide range of compactive *** MsW(s_(1)),Windrows sample(s_(2))of an age of 2 months,5-year-old MsW(s_(3)),10-year-old MsW(s_(4)),and 15-year-old MsW(s_(5))were collected from the working phase of the solid waste management site,Vilholi Nasik,(MH)*** was carried in 1000 mL capacity mold under four different energy levels,i.e.,552(E_(1)),1125(E_(2)),2682(E_(3)),and 5364(E4)kJ/m^(3).The results showed that the maximum dry density(γ_(dmax))was observed less in the s_(1)sample at the standard compactive *** the age of the sample increase from fresh to 5 years,γ_(dmax)was more with lesser optimum moisture content(w_(opt))because of the completion of the biodegradation *** further change in compaction characteristics was observed once the biodegradation process was *** study shows that the particle size of MsW gets reduced as the age of the sample *** reduces the particle size of MsW is one of the contributing factors to increasingγ_(dmax).When compactive energy is changed from E_(1)to E4 theγ_(dmax)increased and w_(opt)decreased for all the samples.A normalized w_(opt)and compaction energy correlation is proposed in this *** correlation can get compaction characteristics of fresh and aged MsW at any compactive effort.

Fluorescently encoded microbeads are in demand for multiplexed applications in different *** to organic dye-based commercially available Luminex's x MAP technology, upconversion nanoparticles(UCNPs) are better alternatives due to their large antistokes shift, photostability, nil background, and single wavelength excitation. Here, we developed a new multiplexed detection system using UCNPs for encoding poly(ethylene glycol) diacrylate(PEGDA) microbeads as well as for labeling reporter antibody. However, to prepare UCNPs-encoded microbeads, currently used swellingbased encapsulation leads to non-uniformity, which is undesirable for fluorescence-based multiplexing. Hence,we utilized droplet microfluidics to obtain encoded microbeads of uniform size, shape, and UCNPs distribution inside. Additionally, PEGDA microbeads lack functionality for probe antibodies conjugation on their *** to functionalize the surface of PEGDA microbeads(acrylic acid incorporation, polydopamine coating)reported thus far quench the fluorescence of UCNPs. Here,PEGDA microbeads surface was coated with silica followed by carboxyl modification without compromising the fluorescence intensity of UCNPs. In this study, droplet microfluidics-assisted UCNPs-encoded microbeads of uniform shape, size, and fluorescence were *** color codes were generated by mixing UCNPs emitting red and green colors at different ratios prior to encapsulation. UCNPs emitting blue color were used to label the reporter antibody. Probe antibodies were covalently immobilized on red UCNPs-encoded microbeads for specific capture of human serum albumin(HsA) as a model protein. The system was also demonstrated for multiplexed detection of both human C-reactive protein(hCRP) and HsA protein by immobilizing anti-h CRP antibodies on green UCNPs.

Precision genome editing is highly desired for crop *** recently emerged CRIsPR/Cas technology offers great potential applications in precision plant genome engineering.A prime editing(PE)approach combining a reverse transcriptase(RT)with a Cas9 nickase and a“priming”extended guide RNA(gRNA)has shown a high frequency for precise genome modification in mammalian cells and several plant ***,the applications of the PE approach in dicot plants are still limited and *** designed and tested prime editors for precision editing of a synthetic sequence in a transient assay and for desirable alleles of 10 loci in tomato by stable *** data obtained by targeted deep sequencing also revealed only low PE efficiencies in both the tobacco and tomato *** assessment of the activities of the PE components uncovered that the fusion of RT to Cas9 and the structure of PE gRNAs(pegRNAs)negatively affected the cleaving activity of the Cas9 *** self-complementarity between the primer binding sequences(PBss)and spacer sequence might pose risks to the activity of the Cas9 ***,modifying the pegRNA sequences by shortening or introducing mismatches to the PBss to reduce their melting temperatures did not enhance the PE efficiency at the MADs-box protein(slMBP21),alcobaca(slALC),and acetolactate synthase 1(slALs1)*** data show challenges of the PE approach in tomato,indicating that a further improvement of the PE system for successful applications is demanded,such as the use of improved expression systems for enriching active PE complexes.

Background： Current research to enrich cattle feed has primarily focused on treatment using white rot fungi, while there are scarce reports using the enzyme tannase, which is discussed only in reviews or in the form of a hypothesis. In this context, the aim of the present study was to evaluate the effect of tannase on wheat straw （Ws） and also the effect of lyophilized tannase at concentrations of 0.1%, 0.2%, and 0.3% （w/w） on Ws followed by fermentation with Ganodermo sp. for 10 d and compared in relation to biochemical parameters, crude protein （CP） content, and nutritional value by calculating the C/N ratio in order to improve the nutritional value of cattle feed. Results： Penicillium charlesii, a tannase-producing microorganism, produced 61.4 IU/mL of tannase in 54 h when 2% （w/v） tannic acid （TA） was initially used as a substrate in medium containing （% w/v） sucrose （1.0）, NaNO3 （1.0）, and MgsO4 （0.08 pH, 5.0） in a 300-L fermentor （working volume 220 L）, and concomitantly fed with 1.0% （w/v） TA after 24 h. The yield of partially purified and lyophilized tannase was 5.8 IU/mg. The tannin-free myco-straw at 0.1% （w/w） tannase showed 37.8% （w/w） lignin degradation with only a 20.4% （w/w） decrease in cellulose content and the in vitro feed digestibility was 32.2%. An increase in CP content （up to 1.28-fold） along with a lower C/N ratio of 25.0%, as compared to myco-straw, was obtained. Conclusions： The use of tannin-free myco-straw has potential to improve the nutritional content of cattle feed. This biological treatment process was safe, eco-friendly, easy to perform, and was less expensive as compared to other treatment methods.

A simple, precise and rapid liquid chromatography-tandem mass spectrometry （LC-Ms/Ms） method has been developed and validated for the simultaneous determination of oseltamivir and oseltamivir carboxylate, a neuraminidase inhibitor, using their deuterated analogs as internal standards （Iss）. The method involved solid phase extraction of the analytes and Iss from 200 μL human plasma with no reconstitution and drying steps. The chromatographic separation was achieved on a symmetry C18 （100 mm × 4.6 mm, 5 μm） column using 10 mM ammonium formate and acetonitrile （30：70, v/v） as the mobile phase in a run time of 2.0 min. Quantitation of analytes and Iss were done by multiple reaction monitoring on a triple quadrupole mass spectrometer in the positive ionization mode. The linearity of the method was established in the concentration range of 0.5-200 ng/mL and 2.0-800 ng/mL for oseltamivir and oseltamivir carboxylate respectively. The mean extraction recovery for oseltamivir （94.4%） and oseltamivir carboxylate （92.7%） from spiked plasma samples was consistent and reproducible. The application of this method was demonstratedby a bioequivalence study in 42 healthy Indian subjects with 75 mg oseltamivir phosphate capsules. The assay reproducibility was established by reanalysis of 151 incurred subject samples.
