BACKGROUND A significant subset of individuals with epilepsy fails to respond to currently available antiepileptic drugs,resulting in heightened mortality rates,psychosocial challenges,and a diminished quality of *** factors,particularly within the SCN1A gene,and the pro-inflammatory cytokine response is important in intricating the drug resistance in idiopathic epilepsy *** this extended study,we determined the correlation of rs6732655A/t single nucleotide polymorphism to understand the causative association of SCN1A gene with epilepsy drug resistance and inflammatory *** to find the correlation of SCN1A gene rs6732655A/t polymorphism with the drug-resistant epilepsy and inflammatory *** the study enrolled 100 age and sex-matched patients of both drug-resistant and drug-responsive epilepsy *** analysed the rs6732655A/t polymorphism to study its association and causative role in drug-resistant epilepsy cases using restriction fragment length polymorphism *** diagnostic performance of interleukin(IL)-1β,IL-6,and high mobility group box 1(HMGB1)protein levels was evaluated in conjunction with genotypic outcome receiver operating characteristic *** At and AA genotypes of rs6732655 SCN1A gene polymorphism were associated with higher risk of drug resistance *** biomarkers IL-6,IL1βand HMGB1 demonstrated diagnostic potential,with cutoff values of 4.63 pg/mL,59.52 pg/mL and 7.99 ng/mL,respectively,offering valuable tools for epilepsy ***,specific genotypes(AA and At)were found to be linked to the elevated levels of IL-1βand IL-6 and potentially reflecting increased oxidative stress and neuro-inflammation in drug-resistant cases supporting the previous reported outcome of high inflammatory markers response in drug resistance *** SCN1A genotypes AA and At are linked to higher drug-resistant epilepsy *** findings underscore the potential influence of inflammation

In this paper,we propose and analyze a fractional-order SIS epidemic model with the saturated treatment and disease *** existence and uniqueness,nonnegativity and finiteness of solutions for our suggested model have been *** equilibria of the model are found and their local and global stability analyses are also ***,the conditions for fractional backward and fractional Hopf bifurcation are also analyzed in both the commensurate and incommensurate fractional-order *** study how the control parameter and the order of the fractional derivative play role in local as well as global stability of equilibrium points and Hopf *** have demonstrated the analytical results of our proposed model system through several numerical simulations.

In this paper, we describe an SIS epidemic model where both the disease transmission rate and treatment function are considered in saturated forms. the dynamical behavior of the system is analyzed. the system is customized by considering the disease trans- mission rate and treatment control as fuzzy numbers and then fuzzy expected value of the infected individuals is determined. the fuzzy basic reproduction number is investi- gated and a threshold condition of pathogen is derived at which the system undergoes a backward bifurcation.

RNAi-Mediated Silencing of ItPK Gene Reduces Phytic Acid Content,Alters transcripts of Phytic Acid Biosynthetic Genes,and Modulates Mineral Distribution in Rice Seeds

Phytic acid is the principal storage form of phosphorus in plant seeds and an essential signalling molecule in several regulatory processes of plant ***,it is known as an anti-nutrient compound owing to its potent chelating ***,reducing the phytic acid content in crops is *** involving regulation of MIPS and IPK1 genes to generate low phytate rice have been reported ***,the functional significance of OsItPK and the effect of its down-regulation on phytic acid content and the associated pleiotropic effects on rice have not yet been *** this study,tissue specific RNA interference(RNAi)-mediated down-regulation of a major ItPK homolog(OsItP5/6K-1)resulted in 46.2%decrease in phytic acid content of t2 transgenic seeds with a subsequent 3-fold enhancement in the inorganic phosphorus *** of OsItP5/6K-1 altered the transcript levels of essential phytic acid pathway genes,without significantly affecting the transcript levels of other OsItPK ***,the mapping of elements through X-ray microfluorescence analysis revealed significant changes in the spatial distribution pattern and translocation of elements in low phytate ***,low phytate polished seeds exhibited 1.3-fold and 1.6-fold enhancement in iron and zinc content in the grain endosperm,*** of OsItP5/6K-1 also altered the amino acid and myo-inositol content of the transgenic *** results successfully established that RNAi-mediated silencing of OsItP5/6K-1 gene significantly reduced the phytate levels in seeds without hampering the germination potential of seeds and plant *** present study provided an insight into the mechanism of phytic acid biosynthesis pathway.

Flood susceptibility modelling using advanced ensemble machine learning models

Floods are one of nature's most destructive disasters because of the immense damage to land,buildings,and human *** is difficult to forecast the areas that are vulnerable to flash flooding due to the dynamic and complex nature of the flash ***,earlier identification of flash flood susceptible sites can be performed using advanced machine learning models for managing flood *** this study,we applied and assessed two new hybrid ensemble models,namely Dagging and Random Subspace(RS)coupled with Artificial Neural Network(ANN),Random Forest(RF),and Support Vector Machine(SVM)which are the other three state-of-the-art machine learning models for modelling flood susceptibility maps at the teesta River basin,the northern region of *** application of these models includes twelve flood influencing factors with 413 current and former flooding points,which were transferred in a GIS *** information gain ratio,the multicollinearity diagnostics tests were employed to determine the association between the occurrences and flood influential *** the validation and the comparison of these models,for the ability to predict the statistical appraisal measures such as Freidman,Wilcoxon signed-rank,and t-paired tests and Receiver Operating Characteristic Curve(ROC)were *** value of the Area Under the Curve(AUC)of ROC was above 0.80 for all *** flood susceptibility modelling,the Dagging model performs superior,followed by RF,the ANN,the SVM,and the RS,then the several benchmark *** approach and solution-oriented outcomes outlined in this paper will assist state and local authorities as well as policy makers in reducing flood-related threats and will also assist in the implementation of effective mitigation strategies to mitigate future damage.

Expression quantitative trait analyses to identify causal genetic variants for type 2 diabetes susceptibility

type 2 diabetes(t2D) is a common metabolic disorder which is caused by multiple genetic perturbations affecting different biological pathways. Identifying genetic factors modulating the susceptibility of this complex heterogeneous metabolic phenotype in different ethnic and racial groups remains challenging. Despite recent success, the functional role of the t2D susceptibility variants implicated by genome-wide association studies(GWAS) remains largely unknown. Genetic dissection of transcript abundance or expression quantitative trait(eQtL) analysis unravels the genomic architecture of regulatory variants. Availability of eQtL information from tissues relevant for glucose homeostasis in humans opens a new avenue to prioritize GWASimplicated variants that may be involved in triggering a causal chain of events leading to t2D. In this article, we review the progress made in the field of eQtL research and knowledge gained from those studies in understanding transcription regulatory mechanisms in human subjects. We highlight several novel approaches that can integrate eQtL analysis with multiple layers of biological information to identify ethnic-specific causal variants and gene-environment interactions relevant to t2D pathogenesis. Finally, we discuss how the eQtL analysis mediated search for "missing heritability" may lead us to novel biological and molecular mechanisms involved in susceptibility to t2D.

Neural Network Approach to Response of Buildings Due to Earthquake Excitation

the present article investigates the physical phenomena associated with the wave passage effect into a building considering the ground floor as the soft floor with the conformity of the up-to-date scenario of the construction of high rise buildings, due to shear excitation of the base. the aim of the study is to analyse the post-earthquake situation of the building in respect to its health. With this vision, the ensuing problem on two-dimensional building models, non-incorporating soil-structure interaction, is being tackled by both analytical and neural network approaches. Computational results from both ends (of the approaches) show that the wave energy does not always propagate from the ground into the building, but for lower frequency range it sails to the building without any disturbances. However, for higher frequency range, the computational results show that the building experiences large “torsional” deformations, as a result the building may collapse. Finally, both the approaches maintain a good agreement among themselves. the present investigation may lead to a long way in contributing to better and more rational, simplified design criteria.

研究了2个转cry1Ab+cry1Ac杂合基因籼稻品系tt9-3和t19-4对主要非靶标害虫飞虱、叶蝉以及捕食性蜘蛛优势种田间种群动态的影响。结果表明,Bt稻与亲本对照(IR72)田间白背飞虱和黑尾叶蝉成虫的虫口密度在整个水稻生育期间无显著差异,而若虫密度有一定的不同。tt9-3稻田中白背飞虱和黑尾叶蝉若虫密度分别于移栽后早中期和中后期显著高于对照;tt9-4与对照间无显著差异。Bt水稻与亲本对照间锥腹肖蛸、四斑锯螯蛛、食虫沟瘤蛛、拟环纹豹蛛及拟水狼蛛种群数量动态趋势相似,数量上大多无显著差异。综合分析认为,Bt水稻对稻田优势蜘蛛基本上无明显的负影响。

Mathematical Modelling of Mass transfer and Free Convection Current Effects on Unsteady Viscous Flow with Ramped Wall temperature

An exact analysis of the flow of an incompressible viscous fluid past an infinite vertical plate is conducted taking into account the presence of foreign mass or constant mass flux and ramped wall temperature. the dimensionless governing coupled linear partial differential equations are solved using the Laplace transform technique. two different solutions for the fluid velocity are obtained–one valid for the fluids of Schmidt numbers different from unity, and the other for which the Schmidt number is unity. the effects of Prandtl number (Pr), Schmidt number (Sc), time (t) and mass to thermal buoyancy ratio parameter (N) for both aiding and opposing buoyancy effects on the velocity and skin-friction are studied. Also, the heat and mass transfer effects on the flow near a ramped temperature plate have been compared with the flow near a plate with constant temperature.

Electrical Conductivity, Magnetoconductivity and Dielectric Behaviour of (Mg,Ni)-Ferrite below Room temperature

We report a comprehensive study of electrical transport properties of stoichiometric (Mg,Ni)-ferrite in the temperature range 77 ≤ t ≤ 300K, applying magnetic field upto 1t in the frequency range 20 Hz-1 MHz. After ball milling of MgO, NiO and ?-Fe2O3 and annealing at 1473K, a (Mg,Ni)-ferrite phase is obtained. the temperature dependency of dc resistivity indicates the prevalence of a simple hopping type charge transport in all the investigated samples. the activation energy decreases by annealing the samples by 1473K. the dc magnetoresistivity of the samples is positive, which has been explained by using wave function shrinkage model. the frequency dependence of conductivity has been described by power law and the frequency exponent ‘s’ is found to be anomalous temperature dependent for ball milling and annealing samples. the real part of the dielectric permittivity at a fixed frequency was found to follow the power law ?/(f,t) ? tn. the magnitude of the temperature exponent ‘n’ strongly depends on milling time and also on annealing temperature. the dielectric permittivity increases with milling and also with annealing. An analysis of the complex impedance by an ideal equivalent circuit indicates that the grain boundary contribution is dominating over the grain contribution in conduction process.
