Ba.kground: Pa.kinson’s disea.e (PD) is ca.sed by complex intera.tions between genetic a.d environmenta. fa.tors. Mendelia. forms of PD ra.ely occur in pra.tice, but respective genes ma. pla. some role in pa.hogenesis of a.common spora.ic form of the disea.e. Methods: We a.a.yzed most frequent known point muta.ions (PMs) a.d single-nucleotide polymorphisms (SNPs) in genes responsible for monogenic PD in 408 Russia. pa.ients, using a.ra.ed primer extension (a.EX), rea.-time PCR, a.d restriction fra.ment length polymorphism a.a.ysis. Results: We detected only three heterozygous PMs in the Pa.K2 gene in three non-rela.ed pa.ients with ea.ly-onset spora.ic PD. No a.socia.ion between PD a.d the studied SNPs wa. identified. Conclusion: The exa.ined PMs a.d SNPs in genes responsible for monogenic PD do not contribute significa.tly to the development of spora.ic PD in Russia.

a.a.ysis of monogenic forms a.d ca.dida.e genes of Pa.kinson’s disea.e (PD) does not a.low to describe completely the contribution of genetic fa.tors to the etiopa.hogenesis of the disorder. a. a.proa.h a.socia.ed with a. a.a.ysis of cha.ges in a.tra.scriptome pa.tern during the development of the disea.e in model objects ca. be used to identify new ca.dida.e genes tha. a.e involved in the pa.hogenesis of PD. In this work, we performed a.tra.scriptome a.a.ysis of a.PD model, crea.ed via.stereota.ic unila.era. introduction of the 6-hydroxidopa.ine (6-OHDa. into the substa.tia.nigra.pa.s compa.ta.(SNpc) of a.ra. bra.n, to identify new ca.dida.e genes for PD. We studied tra.scriptome a.tera.ions in the substa.tia.nigra.of the ra. bra.ns 2 weeks a.ter toxin a.ministra.ion, when the ra.s developed the Pa.kinson-like phenotype, a.d 4 weeks a.ter toxin a.ministra.ion, when ma.ima. cha.ges in the beha.ior of a.ima.s were observed. The tra.scriptome a.a.ysis of the substa.tia.nigra.of the ra. bra.ns a. the first time point (2 weeks) revea.ed cha.ges in expression of genes tha. were clustered with high significa.ce (p a.t p va.ue) into three meta.olic pa.hwa.s a.cording to protein pa.ticipa.ion: modifica.ion of the extra.ellula. ma.rix, signa. tra.sduction (including genes encoding signa. peptides), a.d infla.ma.ion processes. This likely indica.es tha., during this time nonspecific effects a.socia.ed with the response to surgery took pla.e in the substa.tia.nigra.of the ra.s. Concomita.tly, the situa.ion cha.ged dra.a.ica.ly a.d a.response a.socia.ed with da.a.e to the nervous tissue wa. observed 4 weeks a.ter neurotoxin a.ministra.ion. a. a.result, we identified five meta.olic pa.hwa.s conta.ning predomina.tly genes, tha. encode protein products tha. a.e involved in the processes of neuron projection, norma. functioning of the soma.a.d dendrites of neurons, syna.tic tra.smission, a.d tra.smission of nerve impulses (p < 0.01, modified Fisher extra

Na.ura. regula.ory peptides a.e biologica.ly a.tive compounds tha. a.e produced by va.ious cells a.d provide a.link a.ong the ma.n regula.ory systems of the body. The field of resea.ch into the biologic a.tivity of endogenous regula.ory peptides is extremely va.t. These peptides a.fect the ca.diova.cula., immune, reproductive, endocrine, digestive, a.d other systems, a.ter energy meta.olism, a.d a.e especia.ly effective in the regula.ion of the centra. nervous system. Despite of the wide ra.ge of preventive a.d thera.eutic effects of na.ura. regula.ory peptides a.d proteins, their a.plica.ion in clinica. pra.tice is difficult. This is prima.ily beca.se of their extreme insta.ility, a. they a.e ra.idly degra.ed by protea.es of the ga.trointestina. tra.t, blood, cerebrospina. fluid, a.d other biologic media. Compounds with higher sta.ility (i.e., a.considera.ly longer ha.f-life compa.ed with tha. of na.ura. molecules) a.d the a.ility to provide a.directiona. effect on the va.ious body systems were obta.ned from modifica.ions of endogenous regula.ory peptides. Synthetic a.a.ogs of regula.ory peptides, a. a.rule, conta.n only na.ura. a.ino a.ids in their composition, a.d their biodegra.a.ion does not lea. to the forma.ion of toxic products;thus, they ha.e fewer side effects. This review focuses on the considera.ion of two synthetic regula.ory peptides, Sema. a.d Sela.k, which were the ba.es for the crea.ion of new drugs tha. a.e used effectively in the trea.ment of va.ious disea.es of the nervous system. The synthetic a.a.og of a. a.renocorticotropic hormone 4-10 fra.ment (a.TH4-10) Sema. is a.powerful neuroprotective a.ent tha. is pa.ticula.ly effective a. a.thera.y for stroke.
