Establishment and Characterization of a new Cell Line from Koi Brain(Cyprinus carpio L.)

Koi herpesvirus disease(KHVD) is associated with high mortality in both common carp and koi carp(Cyprinus carpio L.) worldwide. The indirect detection of fish viruses based on the identification of antibodies has emerged as a practical and reliable means of diagnosis. Thus, it is important to create monoclonal antibodies(mAb) against carp Igm. By using hybridoma-monoclonal antibody technology, one hybridoma cell line secreting mAbs against the Igm from carp was established. By western blot analysis, the secreted mAb from cell lines A5-E10 recognized the heavy chain of carp Igm, and did not react with immunoglobulins from three different fish species: grass carp(Ctenopharhyngodon idella), tilapia(Oreochromis mossambicus) and mandarin fish(Siniperca chuatsi).The result demonstrated that mAb was highly specific and sensitive to the Ig of carp, and can be used for monitoring the immunity level of carp, for example, the indirect KHV diagnosis by antibody ELISA. An indirect ELISA method was established using the mAb A5-E10 to detect antibodies in koi sera for the detection KHV antibodies in koi sera. A total number of 200 serum samples from koi were collected from three farms and tested by antibody ELISA. The final results showed that 147(73.5%) samples were confirmed as KHV antibody negative and 53(26.5%) were definitely positive, bearing antibodies against KHV.

A novel permanently growing brain cell line from koi(Cyprinus carpio L.)(KB cell line) was established andits suitability for detection of koi herpesvirus(KHV) was demonstrated in this study. The KB cell line was optimally maintained at 27 °C in Leibovitz’s L-15 medium supplemented with 10 % fetal bovine serum(FBS).It was subcultured more than 100 times, and chromosome analysis revealed that 51.54% of KB cells at passage 80 maintained the abnormal diploid chromosome number 2 n=96 while the modal chromosome number was 2 n = 100. The cell line was cryopreserved in liquid nitrogen at-196℃ and was recovered from storage after one year with good cell viability and vitality. The results of virus isolation demonstrated that KB cells were susceptible to KHV, which was shown by the presence of an obvious cytopathic effect and abundant virus particles. The viral titers of KHV in KB reached 10 TCID/0.1 mL within 7 days. Immunofluorescence and western blot assays confirmed that KB replicated KHV. The newly established KB cell line will serve as a useful tool to elucidate KHV disease(KHVD) pathogenesis.

In order to obtain mg alloys with fine microstructures and high mechanical performances,a novel friction-based processing method,name as“constrained friction processing(CFP)”,was investigated.Via CFP,defect-free mg-Zn-Ca rods with greatly refined grains and high mechanical properties were produced.Compared to the previous as-cast microstructure,the grain size was reduced from more than 1 mm to around 4μm within 3 s by a single process cycle.The compressive yield strength was increased by 350%while the ultimate compressive strength by 53%.According to the established material flow behaviors by“tracer material”,the plastic material was transported by shear deformation.From the base material to the rod,the material experienced three stages,i.***.deformation by the tool,upward flow with additional tilt,followed by upward transportation.The microstructural evolution was revealed by“stop-action”technique.The microstructural development at regions adjacent to the rod is mainly controlled by twinning,dynamic recrystallization(DRX)as well as particle stimulated nucleation,while that within the rod is related to DRX combined with grain growth.

An accurate cartesian method is devised to simulate incompressible viscous flows past an arbitrary moving body.The Navier-Stokes equations are spatially discretized onto a fixed Cartesian mesh.The body is taken into account via the ghost-cell method and the so-called penalty method,resulting in second-order accuracy in velocity.The accuracy and the efficiency of the solver are tested through two-dimensional reference simulations.To show the versatility of this scheme we simulate a threedimensional self propelled jellyfish prototype.

阳朔阶最早由殷保安于2008年提出,命名源自桂林市阳朔县,用以指代上泥盆统锡矿山阶与邵东阶之间的一个地方阶,大致相当于国际年代地层单位法门阶上部。阳朔阶底界以牙形类Palmatolepis rugosa trachytera的首次出现为定义,顶界则以有孔虫Eoendothyra regularis的首现为标志。阳朔阶典型剖面为广西桂林市铁山剖面,自下而上包括“五指山组”和融县组,形成于孤立碳酸盐岩台地边缘,以浅水相的内碎屑、鲕粒泥粒灰岩、粒泥灰岩和颗粒灰岩为特征,局部夹有少量黑色钙质泥岩。在桂林铁山剖面和邻近的小龙门剖面中共采集牙形样品149件,其中94件样品产丰富的牙形类分子,该牙形动物群以Palmatolepis、Neopolygnathus、Polygnathus、Pseudopolygnathus、Bispathodus、Icriodus、Rhodalepis、mehlina和Branmehla属分子为特征,自下而上可识别出6个牙形类生物带,包括Pa.marginifera marginifera带、Pa.rugosa trachytera带、Pa.perlobata postera带、Pa.gracilis expansa带、Bi.costatus带和Bi.ultimus ultimus带,为环古特提斯洋地区法门阶地层对比提供了高精度的生物地层格架。基于详细的牙形类生物地层和沉积相分析,在铁山剖面Pa.rugosa trachytera带上部和Pa.gracilis expansa带顶部浅水相碳酸盐岩序列中识别出两次黑色富有机质沉积事件,对应于相对海平面快速上升和海洋贫氧,可能是法门期全球性Annulata和Dasberg事件在华南边缘海区的沉积响应。

BACKGROUND Geographical(geospatial)clusters have been observed in inflammatory bowel disease(IBD)incidence and linked to environmental determinants of disease,but pediatric spatial patterns in North America are unknown.We hypothesized that we would identify geospatial clusters in the pediatric IBD(PIBD)population of British Columbia(BC),Canada and associate incidence with ethnicity and environmental exposures.AIm To identify PIBD clusters and model how spatial patterns are associated with population ethnicity and environmental exposures.mETHODS One thousand one hundred eighty-three patients were included from a BC Children’s Hospital clinical registry who met the criteria of diagnosis with IBD≤age 16.9 from 2001–2016 with a valid postal code on file.A spatial cluster detection routine was used to identify areas with similar incidence.An ecological analysis employed Poisson rate models of IBD,Crohn’s disease(CD),and ulcerative colitis(UC)cases as functions of areal population ethnicity,rurality,average family size and income,average population exposure to green space,air pollution,and vitamin-D weighted ultraviolet light from the Canadian Environmental Health Research Consortium,and pesticide applications.RESULTS Hot spots(high incidence)were identified in metro Vancouver(IBD,CD,UC),southern Okanagan regions(IBD,CD),and Vancouver Island(CD).Cold spots(low incidence)were identified in Southeastern BC(IBD,CD,UC),Northern BC(IBD,CD),and on BC’s coast(UC).No high incidence hot spots were detected in the densest urban areas.modeling results were represented as incidence rate ratios(IRR)with 95%CI.Novel risk factors for PIBD included fine particulate matter(Pm2.5)pollution(IRR=1.294,CI=1.113-1.507,Pm oil to orchards and grapes(IRR=1.135,CI=1.007-1.270,P=0.033).South Asian population(IRR=1.020,CI=1.011-1.028,Pmily size(IRR=0.467,CI=0.268-0.816,P=0.007),and summer ultraviolet(IBD=0.9993,CI=

We report on an experiment performed at the FLASH2 free-electron laser(FEL)aimed at producing warm dense matter via soft x-ray isochoric heating.In the experiment,we focus on study of the ions emitted during the soft x-ray ablation process using time-of-flight electron multipliers and a shifted maxwell–Boltzmann velocity distribution model.We find that most emitted ions are thermal,but that some impurities chemisorbed on the target surface,such as protons,are accelerated by the electrostatic field created in the plasma by escaped electrons.The morphology of the complex crater structure indicates the presence of several ion groups with varying temperatures.We find that the ion sound velocity is controlled by the ion temperature and show how the ion yield depends on the FEL radiation attenuation length in different materials.

Large benthic foraminifera are an integral component of shallow-water tropical habitats and like many marine calcifiers,are susceptible to ocean acidification(OA)and ocean warming(OW).In particular,the prolific Symbiodiniaceae-bearing and high-magnesium calcite marginopora vertebralis has a low threshold compared to several diatom-bearing and low-magnesium calcite species.In this multi-year mesocosm experiment,we tested three RPC 8.5 climate change scenarios(i)present day,(ii)the year 2050,and(iii)2100.To enable a realistic epiphytic association,these experiments were uniquely conducted using natural carbonate substrate,living calcifying alga,and seagrass.In contrast to previous studies,we detected no reduction in surface-area growth under future climate conditions compared with present day conditions.In terms of calcification,m.vertebralis’epiphytic association to primary producers(i.e.,calcifying algae and seagrasses)potentially ameliorates the effects of OA by buffering against declines in boundary layer pH during periods of photosynthesis(i.e.,CO_(2)removal).Importantly for population maintenance,we observed a strong reduction in asexual fecundity under the 2100 scenario.We propose the additional energy needed to maintain growth might be one reason for drastically reduced asexual reproduction.An alternative explanation could be that the 2℃temperature increase interfered with the environmental synchronization that triggered asexual multiple fission.We conclude that the low levels of reproduction will reduce populations in a high CO_(2)environment and reduce a valuable source of CaCO_(3) sediment production.
