Lack of CD44 variant 6 expression in rectal cancer invasive front associates with early recurrence

AIm: To investigate the prognostic value of CD44 variant 6 (CD44v6), a membranous adhesion molecule, in rectal cancer. mETHODS: Altogether, 210 rectal cancer samples from 214 patients treated with short-course radiotherapy (RT, n = 90), long-course (chemo) RT (n = 53) or surgery alone (n = 71) were studied with immunohistochemistry for CD44v6. The extent and intensity of membranous and cytoplasmic CD44v6 staining, and the intratumoral membranous staining pattern, were ***: membranous CD44v6 expression was seen in 84% and cytoplasmic expression in 81% of the cases. In 59% of the tumors with membranous CD44v6 expression, the staining pattern in the invasive front was determined as "front-positive" and in 41% as "front-negative". The latter pattern was associated with narrower circumferential margin (P = 0.01), infiltrative growth pattern (P mpared to the "front-positive" tumors. CONCLUSION: The lack of membranous CD44v6 in the rectal cancer invasive front could be used as a method to identify patients at increased risk for recurrent disease.

Prevalence of lactase persistent/non-persistent genotypes and milk consumption in a young population in north-west Russia

AIm:To estimate the prevalence of the lactase non-persistent genotype(C/C-13910)in a northern Russian population in accordance with ethnicity,and to evaluate self-reported milk consumption depending on lactase activity. mETHODS:Blood samples for genotyping lactase activity,defining the C/T-13910 variant by polymerase chain reaction,and direct sequencing were taken from 231 medical students of Russian origin aged 17-26 years. We analyzed milk product consumption by questionnaire which was specially designed for the estimation of milk consumption and abdominal complaints. RESULTS:We found that the prevalence of the C/C-13190 genotype in the northern Russian population was 35.6%.The other genotypes nearby C/T-13910 and associated with lactase activity were not present in the study *** consumption of milk among people with the non-persistent genotype tended to be lower than among the lactose tolerant subjects,but was not statistically ***:An investigation of the lactase persistent genotype in a northern Russian population has not been performed *** genotype did not affect the consumption of milk products in this population which could be explained by low consumption of milk products among the entire study population.

BACKGROUND Anal fistula is a pathological connection between the anal canal and perianal skin, which most commonly develops from an infected anal crypt. While the majority of anal fistulas are idiopathic, they are also associated with Crohn’s disease (CD) and other inflammatory conditions. The prevalence of anal fistula is estimated to be 1-2 per 10000 patients, but population-based studies on anal fistula epidemiology are limited and outdated. AIm To assess the prevalence of anal fistula and relevant comorbidities, with and without CD in the United Kingdom and Europe. mETHODS A retrospective population-representative observational cohort study was performed in The Health Improvement Network (THIN), a United Kingdom primary care database. mid-year point prevalence of anal fistula was calculated on the first of July for each year between 2014 and 2017. Estimates were calculated for anal fistula overall and by CD status and standardized to the United Kingdom and European population. Prevalence of relevant comorbidities including lymphogranuloma venereum, hidradenitis suppurativa, anal presentation of sexually transmitted diseases, diabetes mellitus, and radiation in the pelvic area was reported. RESULTS The United Kingdom-standardized overall point prevalence of anal fistula was 1.80 (95%CI: 1.65-1.94) per 10000 patients in 2017, while the Europe standardized estimate was 1.83 (95%CI: 1.68-1.98) per 10000 patients. Both these standardized point prevalence estimates ranged from 1.89 to 2.36 between 2014-2016. The United Kingdom-standardized point prevalence of anal fistula without CD was 1.35 (95%CI: 1.23-1.48) per 10000 patients, while the Europe-standardized estimate was 1.39 (95%CI: 1.26-1.52) per 10000 patients. In contrast, the standardized point prevalence estimate of anal fistula with CD was lower for both United Kingdom and Europe (0.44;95%CI United Kingdom: 0.37-0.52, 95%CI Europe: 0.37-0.51) per 10000 patients in 2017. Lactase non-persistence and milk consumption in Estonia

AIm: To defi ne the frequency of the C/T-13910 variant associated with lactase persistence/non-persistence trait and to analyze the milk consumption of lactase non- persistent subjects in Estonia. mETHODS: We genotyped 355 Estonians by polymerase chain reaction and direct sequencing. milk consumption was analyzed by a questionnaire, specially developed to analyze milk consumption and abdominal complaints. RESULTS: The frequency of the genotype of the C/ C-13910 (lactase non-persistence) was found to be 24.8% in native Estonians. No other single nucleotide polymorphisms covering the region of 400 bp adjacent to the C/T-13910 variant were found. Lactase non- persistence subjects were found to consume less milk than lactase persistence subjects. CONCLUSION: The frequency of lactase non- persistence defi ned by the C/C-13910 genotype confi rms the results of the previous studies based on indirect methods of determining hypolactasia. milk consumption of lactase non-persistence subjects is consistent with previously reported figures of adult-type hypolactasia in Estonia. However, lactase non-persistence does not prevent the intake of milk in many adults.

Elevating public awareness of waste management at the household level is a cost effective and a win-win social, economic and environmental approach on the table of policymaking. modern and effective waste management bring about broader economic efficiency and social equity, mitigate consumerism, thus, promoting the conservation of natural resources. In this article, a survey study was conducted among the citizens of Wrocław city, Poland. The city is dynamic, touristic, and industrial. Through random sampling, 160 respondents filled in the questionnaire. The results revealed that the participants appeared aware of the benefits of waste management (Wm);however the current Wm systems and infrastructures are not very satisfying. The participants identified several shortcomings in the current Wm systems such as the lack of sufficient colored bins for glass recycling, overfilled bins and heavy lids, and irregular waste collection system. The study proposes a public-oriented outreach campaign targeting waste separation and less-waste behaviors as economically and environmentally beneficial, accompanied by continuous development of Wm regulations and infrastructures and linked it to waste-to-energy systems and technologies to assist in achieving the long-term energy and emissions-reduction targets.
