Ron receptor-dependent gene regulation of Kupffer cells during endotoxemia

BACKGROUND: Ron receptor tyrosine kinase signaling in macrophages, including Kupffer cells and alveolar macrophages,suppresses endotoxin-induced proinflammatory cytokine/chemokine production. Further, we have also identified genes from Ron replete and Ron deplete livers that were differentially expressed during the progression of liver inflammation associated with acute liver failure in mice by microarray *** important genes and signaling pathways have been identified downstream of Ron signaling during progression of inflammation by this approach, the precise role that Ron receptor plays in regulating the transcriptional landscape in macrophages, and particular in isolated Kupffer cells, has still not been ***: Kupffer cells were isolated from wild-type(TK+/+)and Ron tyrosine kinase deficient(TK-/-) mice. ex vivo, the cells were treated with lipopolysaccharide(LPS) in the presence or absence of the Ron ligand, hepatocyte growth factor-like protein(HGFL). Microarray and qRT-PCR analyses were utilized to identify alterations in gene expression between ***: Microarray analyses identified genes expressed differentially in TK+/+ and TK-/- Kupffer cells basally as well as after HGFL and LPS treatment. Interestingly, our studies identified Mefv, a gene that codes for the anti-inflammatory protein pyrin, as an HGFL-stimulated Ron-dependent ***, lipocalin 2, a proinflammatory gene, which is induced by LPS, was significantly suppressed by HGFL *** results were validated by qRT-PCR studies on Kupffer cells treated with LPS and ***: The studies herein suggest a novel mechanism whereby HGFL-induced Ron receptor activation promotes the expression of anti-inflammatory genes while inhibiting genes involved in inflammation with a net effect of diminished inflammation in macrophages.

BACKGROUND:Prior experimentation has shown that loss of the tyrosine kinase(TK) signaling domain of the Ron receptor leads to marked hepatocyte protection in a model of lipopolysaccharideinduced acute liver failure(ALF) in D-galactosamine(GalN)sensitized *** aim of this study was to identify the role of Ron in the regulation of hepatic gene ***:Microarray analyses were performed on liver RNA isolated sequentially from wild-type(WT) and TK-/mice during the progression of *** array data were validated using Western and immunohistochemistry analyses as well as with ex vivo culture ***:At baseline,101 genes were differentially expressed between WT and TK-/-livers,which regulate processes involved in hypoxia,proliferation,apoptosis and *** hour after ALF induction,WT livers exhibited increased cytokine expression compared to TK-/-livers,and after 4 hours,an induction of suppressor of cytokine signaling(SOCS) genes as well as JAK-STAT pathway activation were prominent in TK-/livers compared to ***:Our studies suggest a novel hepato-protective mechanism in Ron TK-/-mice wherein increased and sustained SOCS production and JAK-STAT activation in the hepatocyte may inhibit the destructive proinflammatory milieu and promote survival factors which blunt hepatic death and the ensuing development of ALF.

ecological and evolutionary studies on spiders have been featured prominently throughout the contemporary behavio- ral syndromes movement. Here we review the behavioral syndromes literature devoted to spiders, and identify some ways in which behavioral syndromes can impact the function of spiders in ecological communities. We further highlight three general themes within the behavioral syndromes literature for which spiders have served as front running model systems：（1） how trait correlations beget performance trade-offs, （2） the influence that behavioral trait variants have on interspecific interactions and （3） mechanisms that aid in maintaining behavioral variation within-and among-populations. Research on behavioral syndromes con-tinues to grow at an impressive rate, and we feel the success of behavioral syndromes studies in spiders bodes well for their con-tinued prominence.

The Japanese ladybirds, Coccinula crotchi and Coccinula sinensis, are infected with very closely related strains of male-killing Flavobacterium

Male-killing is 1 of 4 known strategies that inherited parasitic endosymbionts have evolved to manipulate their host＇s reproduction. In early male-killing, infected male offspring are killed early in embryogenesis. Within the Insecta, male-killing bacteria have been found in a wide range of hosts. The Coccinellidae families of beetles, better known as ladybirds, are particularly prone to male-killer invasion. In samples of the coccinellid, Coc- cinula crotchi, from Japan, a new male-killing bacterium was revealed by phenotypic assay. Molecular genetic analysis revealed the identity to be a tetracycline-sensitive Flavobac- terium that causes female-biased offspring sex ratio. Furthermore, that Flavobacterium strain was found to be closely related to the Flavobacterium causing male-killing in the congeneric Japanese coccinellid, Coccinula sinensis, which was collected from the same region. However, we found slightly different Flavobacterium strains infecting C. sinen- sis from regions with different environmental conditions. This may be an indication of horizontal transmission of male-killing Flavobacterium between these 2 ladybird spices. Finally, environmental conditions may affect the spread of male-killing bacteria among their hosts.

enhancing the observing capacity for the surface ocean by the use of Volunteer Observing Ship

Knowledge of the surface ocean dynamics and the underlying controlling mechanisms is critical to understand the natural variability of the ocean and to predict its future response to climate *** this paper,we highlight the potential use of Volunteer Observing Ship(VOS),as carrier for automatic underway measuring system and as platform for sample collection,to enhance the observing capacity for the surface *** review the concept,history,present status and future development of the VOS-based in situ surface ocean *** successes of various VOS projects demonstrate that,along with the rapid advancing sensor techniques,VOS is able to improve the temporal resolution and spatial coverage of the surface ocean observation in a highly cost-effective manner.A sustained and efficient marine monitoring system in the future should integrate the advantages of various observing platforms including VOS.

epstein-Barr virus-associated post-transplant lymphoproliferative disorders:beyond chemotherapy treatment

Post-transplant lymphoproliferative disorder(PTLD)is a rare but life-threatening complication of both allogeneic solid organ(SOT)and hematopoietic cell transplantation(HCT).The histology of PTLD ranges from benign polyclonal lymphoproliferation to a lesion indistinguishable from classic monoclonal *** commonly,PTLDs are epstein-Barr virus(eBV)positive and result from loss of immune surveillance over *** for PTLD differs from the treatment for typical non-Hodgkin lymphoma because prognostic factors are different,resistance to treatment is unique,and there are specific concerns for organ *** recipients of HCT have a limited time during which they are at risk for this complication,recipients of SOT have a lifelong requirement for immunosuppression,so approaches that limit compromising or help restore immune surveillance are of high ***,while eBV-positive and eBV-negative PTLDs are not intrinsically resistant to chemotherapy,the poor tolerance of chemotherapy in the post-transplant setting makes it essential to minimize potential treatment-related toxicities and explore alternative treatment ***,reduced-toxicity approaches such as single-agent CD20 monoclonal antibodies or bortezomib,reduced dosing of standard chemotherapeutic agents,and non-chemotherapy-based approaches such as cytotoxic T cells have all been ***,we review the chemotherapy and non-chemotherapy treatment landscape for PTLD.
